Hnrnp A1 Interacts With The Genomic And Subgenomic Rna Promoters Of Sindbis Virus And Is Required For The Synthesis Of G And Sg Rna, Hongxing Gui, Chi Wei Lu, Sandra Adams, Victor Stollar, Mei Ling Li

Department of Biology Faculty Scholarship and Creative Works

Background. Sindbis virus (SV) is the prototype of alphaviruses which are a group of widely distributed human and animal pathogens. Heterogeneous nuclear ribonucleoprotein (hnRNP) A1 is an RNA-binding protein that shuttles between the nucleus and the cytoplasm. Our recent studies found that hnRNP A1 relocates from nucleus to cytoplasm in Sindbis virus (SV)-infected cells. hnRNP A1 binds to the 5' UTR of SV RNA and facilitates the viral RNA replication and translation. Methods. Making use of standard molecular techniques, virology methods and an in vitro system developed by our lab to assess the role of hnRNP A1 in SV positive …

Effects Of Alternative Prey As A Buffer To Predation Of Channel Catfish (Ictalurus Punctatus) By Largemouth Bass (Micropterus Salmoides), Shelley Nellis Jul 2010

Effects Of Alternative Prey As A Buffer To Predation Of Channel Catfish (Ictalurus Punctatus) By Largemouth Bass (Micropterus Salmoides), Shelley Nellis

Theses and Dissertations

Channel catfish have pectoral spines that lock to defend against gape-limited predators such as largemouth bass. Previous work indicated that spines increase survival of channel catfish exposed to bass but did not determine whether bass avoid catfish if less dangerous species are available. We presented bass with channel catfish and two alternatives, bluegill and goldfish, and compared order of ingestion, ingestion time, percent of successful strikes, and time spent chasing each prey species. We also presented the three species in a jar study that standardized position in the water column as well as in a net-pen study. The order of …

Insights Into The Catalytic Mechanism Of Retro-Aldol Cleavage Of Β-Hydroxy Amino Acids By Escherichia Coli L-Threonine Aldolase, Remsh Soumya Govinda Jul 2010

Insights Into The Catalytic Mechanism Of Retro-Aldol Cleavage Of Β-Hydroxy Amino Acids By Escherichia Coli L-Threonine Aldolase, Remsh Soumya Govinda

Theses and Dissertations

With over 140 vitamin B6 (Pyridoxal 5’-phosphate, PLP) dependent enzymes, serving vital roles in various transamination, decarboxylation, retro-aldol cleavage and synthesis pathways these enzymes constitute the most versatile catalytic systems in nature. Enzymes of this group have an inherent reaction as well as substrate specificity. A single co-factor namely, PLP is used by specific enzymes of this group to serve distinct roles during the catalytic reaction. An ordered evolutionary adaptation in these enzymes has led to specialization achieved by each enzyme for catalyzing specific reactions. L-Threonine aldolase (L-TA) is one such PLP- dependent enzyme that catalyzes the retro-aldol cleavage of …

The Effect Of Cobalt Protoporphyrin And Cobalt Chloride On Heme Oxygenase Expression And Protection From Deoxycholate-Induced Apoptosis, Tina Lawson Jul 2010

The Effect Of Cobalt Protoporphyrin And Cobalt Chloride On Heme Oxygenase Expression And Protection From Deoxycholate-Induced Apoptosis, Tina Lawson

Theses and Dissertations

The inner surface of the stomach is lined by a mucous membrane known as the gastric mucosa. The integrity of the gastric mucosa is critical for protecting the stomach from the low pH and proteolytic environment within the lumen. Both clinically and experimentally, exposure of gastric mucosal cells to bile salts is known to cause injury. Bile salts present in duodenogastric reflux are thought to play a significant role in gastric ulcer formation and alkaline gastritis. In vitro, studies using physiologic concentrations of the secondary bile salt, deoxycholic acid, indicate that bile salts can induce apoptosis in cultured human gastric …

First-Principles Study Of The Variation Of Electron Transport In A Single Molecular Junction With The Length Of The Molecular Wire, Partha Pratim, Ranjit Pati Jul 2010

First-Principles Study Of The Variation Of Electron Transport In A Single Molecular Junction With The Length Of The Molecular Wire, Partha Pratim, Ranjit Pati

Department of Physics Publications

We report a first-principles study of quantum transport in a prototype two-terminal device consisting of a molecular nanowire acting as an inter-connect between two gold electrodes. The wire is composed of a series of bicyclo[1.1.1]pentane (BCP) cage-units. The length of the wire (L) is increased by sequentially increasing the number of BCP cage units in the wire from 1 to 3. A two terminal model device is made out of each of the three wires. A parameter free, nonequilibrium Green’s function approach, in which the bias effect is explicitly included within a many body framework, is used to …

Design And Evaluation Of Chitosan-Calcium Phosphate Scaffolds Constructed From Air Dried And Lyophilized Microspheres, Duong T. Nguyen Jul 2010

Design And Evaluation Of Chitosan-Calcium Phosphate Scaffolds Constructed From Air Dried And Lyophilized Microspheres, Duong T. Nguyen

Electronic Theses and Dissertations Archive

The orthopedic clinical demand for bone grafts is a persistent problem for patients with age-related bone fractures and diseased bone defects. The aim of this study was to use calcium phosphate, a biomimetic ceramic with high compressive strength, and chitosan, a natural biodegradable and biocompatible polymer, to construct microsphere-based composite scaffolds to serve as a bone graft. Two types of scaffolds,(1) air-dried microspheres (AD) and (2) solid air-dried and lyophilized microsphere combination (FDAD), were evaluated in vitro for mineralization and enzymatic degradation. The combination FDAD scaffold showed on average ~80% increase (p<0.01) in cell number per scaffold mass compared to AD because of the larger surface area advantage. Due to the higher cell number, the production of collagen was ~31% greater (p<0.01) on FDAD scaffolds compared to AD scaffolds. However, scanning electron micrographs indicated minimal matrix deposition for both scaffold types. The AD scaffolds had a three-fold compressive strength advantage compared to FDAD scaffolds. These results indicate FDAD scaffolds have more osteogenic potential based on cell growth and collagen elaboration, but AD scaffolds demonstrated higher compressive strength.

International Journal of Exercise Science

The purpose of this study was to pilot a 5-week community-based intervention on improving measures of health-related fitness in overweight children. Data were obtained from 8 overweight and obese 8- to 14-year-old children. Measurements included muscular fitness (curl-ups and modified pull-ups), aerobic capacity (20 meter progressive aerobic cardiovascular endurance run [PACER] test), body composition (tricep and calf skinfolds), body mass index (BMI), and flexibility (back saver sit-and-reach). A significant reduction in BMI was observed at post-test compared to baseline (p = .03). There was a significant decrease in body fat at post-test for boys (p = .013).

Monitoring Training Load In Indian Male Swimmers., Pralay Majumdar Dr., Sri Vidhya Jr.

International Journal of Exercise Science

The present study was taken up to monitor the training load and the impact of training load to the hormone concentrations such as testosterone, cortisol and T/C (Testosterone/Cortisol) ratio during the three phases of training viz preparatory phase, pre-competitive phase and competitive phase in Indian male swimmers. Blood samples were collected at the completion of each phase and the hormone concentrations were quantified. Our results reveal that the testosterone and T/C ratio significantly decreases whereas cortisol increases in the subsequent periodised cycle and concludes that the hormone concentration changes with the intensity and duration of exercise i.e. when the intensity …

Association Of Body Surface Area And Body Composition With Heart Structural Characteristics Of Female Swimmers, Bahreh Sheikh Saraf, Hojat Allah Nikbakht, Mohammad Ali Azarbayjani Jul 2010

Association Of Body Surface Area And Body Composition With Heart Structural Characteristics Of Female Swimmers, Bahreh Sheikh Saraf, Hojat Allah Nikbakht, Mohammad Ali Azarbayjani

International Journal of Exercise Science

In healthy nonathletic populations, some left ventricle (LV) parameters such as LV mass (LVM) and LV end diastolic dimension (LVEDD) can be predicted by some of body size parameters such as body surface area (BSA), fat-free mass (FFM), and height (H). These body size parameters use to remove covariate influence of body size from cardiac dimension variables and allow comparisons to be made between individuals and groups of different body size. Endurance exercise has been associated with changes in LV size and body composition of athletes. The aim of this study was to evaluate in 30 trained female swimmers (age …

Objective And Subjective Assessments Of ‘Normal’ Walking Pace, In Comparison With That Recommended For Moderate Intensity Physical Activity, Kathryn L. Taylor, Claire Fitzsimons, Nanette Mutrie Jul 2010

Objective And Subjective Assessments Of ‘Normal’ Walking Pace, In Comparison With That Recommended For Moderate Intensity Physical Activity, Kathryn L. Taylor, Claire Fitzsimons, Nanette Mutrie

International Journal of Exercise Science

Abstract Despite its common application and widely reported health benefits, walking, in relation to pace and intensity, is under-researched. Few studies have addressed whether people normally walk at a pace which meets the public health recommendations for moderate intensity physical activity (1.34-1.79 ms-1) and there is no known research on individuals’ perceptions of factors which influence walking pace. This study aimed to objectively assess if participants were reaching the pace required for moderate intensity physical activity during ‘normal’ walking. This was examined via a Global Positioning System (GPS) over a 1 km outdoor walk and a timed 150 m trial. …

Grassland Governance And Common-Interest Communities, Anthony Schutz Jul 2010

Grassland Governance And Common-Interest Communities, Anthony Schutz

Nebraska College of Law: Faculty Publications

In the United States, today’s ranches are engaging in small-scale nature-based endeavors to diversify their income base. But the geographic boundary of the land they own creates a relatively small area within which to operate, and fragmented ownership diminishes the ability of any single landowner to produce nature-based income. Collective action among nearby landowners can produce a set of resources from which all members of the group can profit. Such action can enhance the economic, social, and environmental sustainability of grasslands and the populations that use them. This article shows that common-interest communities can be used to provide and allocate …

Delayed Wound Healing: Can Exercise Accelerate It?, K Todd Keylock, Hilary Young Jul 2010

Delayed Wound Healing: Can Exercise Accelerate It?, K Todd Keylock, Hilary Young

International Journal of Exercise Science

Poorly healing wounds affect millions around the world, yet preventive methods and low-cost, effective treatments are few. Wounds heal quickly through well-coordinated phases in those who are healthy and active but can become chronically nonhealing as a result of disease and inactivity. Recently it has been reported that moderate aerobic exercise accelerated healing rates in the aged. High levels of inflammation are known to delay wound healing, and aging and disease are associated with chronically increased inflammation. Therefore, exercise may help speed healing by reducing inflammation to healthier levels not only in the aged, but also in other populations with …

When challenged by low-iron conditions several Gram-negative pathogens secrete a hemophore (HasA) to scavenge hemin from its host and deliver it to a receptor (HasR) on their outer membrane for internalization. Here we report results from studies aimed at probing the structural and dynamic processes at play in the loading of the apo-hemophore secreted by P. aeruginosa (apo-HasAp) with hemin. The structure of apo-HasAp shows a large conformational change in the loop harboring axial ligand His32 relative to the structure of holo-HasAp, whereas the loop bearing the other axial ligand, Tyr75, remains intact. To investigate the role played by the …

Role Of Pyridoxine 5'-Phosphate Oxidase In Metabolism And Transfer Of Pyridoxal 5'-Phosphate, Sayali Karve Jul 2010

Role Of Pyridoxine 5'-Phosphate Oxidase In Metabolism And Transfer Of Pyridoxal 5'-Phosphate, Sayali Karve

Theses and Dissertations

Deficiency of vitamin B6 due to mutations in key B6 metabolizing enzymes is suspected to contribute to several pathologies. Vitamin B6 in its active form, pyridoxal 5’-phosphate (PLP) is a cofactor for over 140 known B6 requiring (or PLP-dependent) enzymes, that serve vital roles in many biochemical reactions. There are three primary vitamin B6 forms, pyridoxine (PN), pyridoxamine (PM) and pyridoxal (PL) which are phosphorylated to pyridoxine 5’-phosphate (PNP), pyridoxamine 5’-phosphate (PMP) and PLP respectively. Pyridoxal kinase (PLK) and pyridoxine 5’-phosphate oxidase (PNPO) are the key enzymes involved in both salvage and de novo pathways of PLP biosynthesis. Mutations in …

Validation Of 3d Radiative Transfer In Coastal-Ocean Water Systems As Modeled By Dirsig, Jacqueline Amy Speir Jul 2010

Validation Of 3d Radiative Transfer In Coastal-Ocean Water Systems As Modeled By Dirsig, Jacqueline Amy Speir

Theses

The radiative transfer equation (RTE) is a mathematical description of radiative gains and losses experienced by a propagating electromagnetic wave in a participating medium. Except for an isotropic lossless vacuum, all other volumes have the potential to scatter, absorb and emit radiant energy. Of these possible events, the global scattering term is the greatest obstacle between a radiative transfer problem and its solution. Historically, the RTE has been solved using a host of analytical approximations and numerical methods. Typical solution models exploit plane-parallel assumptions where it is assumed that optical properties may vary vertically with depth, but have an infinite …

Biological Sciences

Previous work has shown that organosilica-based xerogels have the potential to control biofouling. In this study, modifications of chemistry were investigated with respect to their resistance to marine slimes and to settlement of barnacle cyprids. Adhesion force measurements of bovine serum albumin (BSA)-coated atomic force microscopy (AFM) tips to xerogel surfaces prepared from aminopropylsilyl-, fluorocarbonsilyl-, and hydrocarbonsilyl-containing precursors, indicated that adhesion was significantly less on the xerogel surfaces in comparison to a poly(dimethylsiloxane) elastomer (PDMSE) standard. The strength of adhesion of BSA on the xerogels was highest on surfaces with the highest and the lowest critical surface tensions, gamma(C) and …

A Secondary Data Analysis Of The Practice Intervention Designed To "Wipe Out C. Difficile", Kimberly Aronhalt

Theses and Graduate Projects

Clostridium dfficile infection (CDI) recognized as a global healthcare-associated infection (HAI) causing infectious diarrhea, has increased in incidence, severity, and mortality among patients in the last three decades. Related health care costs to patients and health care facilities are enormous. To prevent HAIs healthcare organizations are charged to have infection prevention control programs in place to address CDI. Surveillance of CDI and prevention strategies are challenged to impact the transmission of CDI due to its anaerobic, spore forming characteristics. At a tertiary medical center, Wipe Out C. dfficile was a targeted approach for five high incident units to reduce CDI. …
