Malaria, caused by the apicomplexan parasite Plasmodium, threatens 40% of the world's population. Transmission between vertebrate and insect hosts depends on the sexual stages of the life-cycle. The male gamete of Plasmodium parasite is the only developmental stage that possesses a flagellum. Very little is known about the identity or function of proteins in the parasite's flagellar biology. Here, we characterise a Plasmodium PF16 homologue using reverse genetics in the mouse malaria parasite Plasmodium berghei. PF16 is a conserved Armadillo-repeat protein that regulates flagellar structure and motility in organisms as diverse as green algae and mice. We show that …

Drug Off-Target Effects Predicted Using Structural Analysis In The Context Of A Metabolic Network Model, Roger L. Chang, Lei Xie, Philip E. Bourne, Bernhard O. Palsson

Recent advances in structural bioinformatics have enabled the prediction of protein-drug off-targets based on their ligand binding sites. Concurrent developments in systems biology allow for prediction of the functional effects of system perturbations using large-scale network models. Integration of these two capabilities provides a framework for evaluating metabolic drug response phenotypes in silico. This combined approach was applied to investigate the hypertensive side effect of the cholesteryl ester transfer protein inhibitor torcetrapib in the context of human renal function. Publications and Research

Genetic analyses in Drosophila epithelia have suggested that the phenomenon of “cell competition” could participate in organ homeostasis. It has been speculated that competition between different cell populations within a growing organ might play a role as either tumor promoter or tumor suppressor, depending on the cellular context. The evolutionarily conserved Hippo (Hpo) signaling pathway regulates organ size and prevents hyperplastic disease from flies to humans by restricting the activity of the transcriptional cofactor Yorkie (yki). Recent data indicate also that mutations in several Hpo pathway members provide cells with a competitive advantage by unknown mechanisms. Here we provide insight …

The Galapagos Islands are renowned for their high degree of endemism. Marine taxa inhabiting the archipelago might be expected to be an exception, because of their utilization of pelagic habitats—the dispersal barrier for terrestrial taxa—as foraging grounds. Magnificent frigatebirds (Fregata magnificens) have a highly vagile lifestyle and wide geographical distribution around the South and Central American coasts. Given the potentially high levels of gene flow among populations, the species provides a good test of the effectiveness of the Galapagos ecosystem in isolating populations of highly dispersive marine species. Hydronephrosis Predicts Symptomatic Ureterolithiasis, But Does Not Predict Need For Intervention In Patients With Suspected Renal Colic, Nicholas Villalon

Computed tomography (CT) is now a first-line test for renal colic, but is costly, potentially harmful, and rarely changes patient care. Hydronephrosis is often present with symptomatic kidney stones, and is reliably determined using point-of-care ultrasound at the bedside, but it is unknown whether the presence of hydronephrosis indicates a large stone that is more likely to require intervention (6mm or greater). We hypothesized that while hydronephrosis would be associated with symptomatic ureterolithiasis, neither the presence nor the degree of hydronephrosis would accurately predict the size of urinary tract stone. Joint Loop End Modeling Improves Covariance Model Based Non-Coding Rna Gene Search, Jennifer Smith

Electrical and Computer Engineering Faculty Publications and Presentations

The effect of more detailed modeling of the interface between stem and loop in non-coding RNA hairpin structures on efficacy of covariance-model-based non-coding RNA gene search is examined. Currently, the prior probabilities of the two stem nucleotides and two loop-end nucleotides at the interface are treated the same as any other stem and loop nucleotides respectively. Laboratory thermodynamic studies show that hairpin stability is dependent on the identities of these four nucleotides, but this is not taken into account in current covariance models. Crystallization of cholesterol in chemical conditions that involve the presence of emulsions of three different types of surface active agents—cationic, anionic, and nonionic—was investigated by means of scanning and transmission electron microscopic analyses. In contrast with the previous attempts to modify the typical plate- and needle-shaped character of biaxially grown cholesterol crystals, the present study indicates that spherical templates of fine emulsion droplets may impose spherical morphologies to cholesterol particles nucleated within.

Abnormal connectivity of the intrinsic anticorrelated networks, the task-negative network (TNN) and task-positive network (TPN), is implicated in schizophrenia. Comparisons between schizophrenic patients and their unaffected siblings offer an opportunity to further understand illness susceptibility and pathophysiology. We hypothesized that schizophrenic patients would demonstrate hyperconnectivity in the intrinsic networks and that similar, but less pronounced, hyperconnectivity would be evident in the networks of the unaffected siblings. Resting-state functional magnetic resonance images were obtained from schizophrenic patients (n=25), their unaffected siblings (n=25), and healthy controls (n=25). Although researchers are aware of the fact that public and group stigma affect mental health help-seeking behavior, there has been no research on level of collective self-esteem and stigma within specific social networks related to members' attitudes toward seeking mental health services. This Article offers a critique of tort remedies grounded in feminist theory of the body. It demonstrates how tort law is invested in a notion of an individuated legal subject, which fails to capture the critical interconnectedness of bodies in a social, political, historical, and colonial context. Taking the "injury" of endocrine disruption in a Canadian Aboriginal community as an example of a contemporary pollution harm, the analysis considers various torts on a conceptual level, and what they might offer the Aamjiwnaang First Nation in the way of remedies. This study explored the experiences of play therapists working cross-racially with children under the age of 7 to determine if and how play therapy is impacted by a cross-racial therapeutic dyad. Specifically, this study explored whether or not clinicians altered their play therapy approach, made adjustments to the available toys, if race was viewed as connected to the presenting problem, and if play or interactional style differed based on a child's race. 12 clinicians currently conducting cross-racial play therapy with children under age 7 were interviewed using a fixed demographic questionnaire and semi-structured interview guide. This study was designed to examine whether clinicians who identified themselves as maintaining a religious or spiritual practice as part of self-care routines found these practices helpful in continuing to practice effectively. To collect information regarding this question, eleven clinicians doing direct service work were interviewed; each clinician was asked the same set of questions. The study was qualitative in nature. Each clinician had a unique way of expressing, carrying out, and maintaining his or her religious and/or spiritual practices. As a whole, however, the eleven stories created a surprisingly cohesive narrative. All eleven clinicians reported maintaining religious and/or spiritual …
