Pathology and Laboratory Medicine Faculty Publications

The miR-15/107 group of microRNA (miRNA) gene is increasingly appreciated to serve key functions in humans. These miRNAs regulate gene expression involved in cell division, metabolism, stress response, and angiogenesis in vertebrate species. The miR-15/107 group has also been implicated in human cancers, cardiovascular disease and neurodegenerative disease, including Alzheimer's disease. Here we provide an overview of the following: (1) the evolution of miR-15/107 group member genes; (2) the expression levels of miRNAs in mammalian tissues; (3) evidence for overlapping gene-regulatory functions by different miRNAs; (4) the normal biochemical pathways regulated by miR-15/107 group miRNAs; and (5) the roles played …

Advice From Teens To Teens About Dating: Implications For Healthy Relationships, Heidi Adams Rueda, Lela Rankin Williams

Social Work Faculty Publications

Seventy-five Mexican American and White male and female adolescents were asked in focus groups to offer advice to other adolescents pertaining to dating relationships. Across ethnicities and sexes, “Stay on your feet” was the most prominent advice given, followed by advice to “Know when it's right”. “Have good reasoning…especially about that was a prominent theme among females; Mexican American females focused more on pressure associated with sexual activity while White females embedded their advice more often within futuristic and long-term relationship goals. Females offered roughly three times more relationship advice than did males and dialogued collaboratively at greater length, enriching …

Using Diffusion Distances For Flexible Molecular Shape Comparison, Yu-Shen Liu, Qi Li, Karthik Ramani, William Benjamin Sep 2010

Using Diffusion Distances For Flexible Molecular Shape Comparison, Yu-Shen Liu, Qi Li, Karthik Ramani, William Benjamin

School of Mechanical Engineering Faculty Publications

Background

Many molecules are flexible and undergo significant shape deformation as part of their function, and yet most existing molecular shape comparison (MSC) methods treat them as rigid bodies, which may lead to incorrect shape recognition.

Results

In this paper, we present a new shape descriptor, named Diffusion Distance Shape Descriptor (DDSD), for comparing 3D shapes of flexible molecules. The diffusion distance in our work is considered as an average length of paths connecting two landmark points on the molecular shape in a sense of inner distances. The diffusion distance is robust to flexible shape deformation, in particular to topological …

Alteration Of Micrornas Regulated By C-Myc In Burkitt Lymphoma, Anna Onnis1, Giulia De Falco, Giuseppina Antonicelli, Monica Onorati, Cristiana Bellan, Omar Sherman, Shahin Sayed, Lorenzo Leoncini Sep 2010

Alteration Of Micrornas Regulated By C-Myc In Burkitt Lymphoma, Anna Onnis1, Giulia De Falco, Giuseppina Antonicelli, Monica Onorati, Cristiana Bellan, Omar Sherman, Shahin Sayed, Lorenzo Leoncini

Pathology, East Africa

Background: Burkitt lymphoma (BL) is an aggressive B-cell lymphoma, with a characteristic clinical presentation, morphology and immunophenotype. Over the past years, the typical translocation t(8;14) and its variants have been considered the molecular hallmark of this tumor. However, BL cases with no detectable MYC rearrangement have been identified. Intriguingly, these cases express MYC at levels comparable with cases carrying the translocation. In normal cells c-Myc expression is tightly regulated through a complex feedback loop mechanism. In cancer, MYC is often dysregulated, commonly due to genomic abnormalities. It has recently emerged that this phenomenon may rely on an alteration of post-transcriptional …

The Regulation Of The Alternative Splicing Of Caspase 9, Rachel Goehe Sep 2010

The Regulation Of The Alternative Splicing Of Caspase 9, Rachel Goehe

Theses and Dissertations

The pro-apoptotic, caspase 9a, and the anti-apoptotic, caspase 9b, are derived from the caspase 9 gene by alternative splicing. This study demonstrates that the alternative splicing of caspase 9 is dysregulated in a large percentage of non-small cell lung cancer (NSCLC) tumors of the adenocarcinoma type. Furthermore, modulation of the levels of splice variants of caspase 9 had dramatic effects on the anchorage-independent growth and tumorigenic capacity of NSCLC cells. Due to these findings, the molecular mechanisms regulating the post-transcriptional processing of caspase 9 were therefore examined and an exonic splicing silencer (ESS) regulating the pre-mRNA processing of caspase 9 …

Assessing Plasticity : The Populations Responsible For The Epithelial Engraftment On Marrow-Derived Cells, Justin Brent Cohen Sep 2010

Assessing Plasticity : The Populations Responsible For The Epithelial Engraftment On Marrow-Derived Cells, Justin Brent Cohen

Yale Medicine Thesis Digital Library

Bone marrow-derived cells (BMDCs) have significant plasticity allowing them to give rise to various non-hematopoietic cell types including epithelial cells of the lung, liver, gut, and skin. These findings offer tremendous possibilities for the use of cell therapy to treat tissue injury and disease. However, the specific populations of cells responsible for this phenomenon are still unclear with considerable controversy over the mechanism of this transformation. In this study, we sought to compare the epithelial engraftment ability of two populations enriched for hematopoietic stem cells (HSCs), specifically the fractionated, lineage-depleted, and homed subset (FLH) and the lineage-Sca-1+c-Kit+ sorted population (LSK), …

Microrna-21 Dysregulates The Expression Of Mef2c In Neurons In Monkey And Human Siv/Hiv Neurological Disease., Sowmya V. Yelamanchili, A Datta Chaudhuri, L N. Chen, Huangui Xiong, Howard S. Fox

Journal Articles: Pharmacology & Experimental Neuroscience

MicroRNAs (miRNAs) play important roles in regulating a plethora of physiological and pathophysiogical processes including neurodegeneration. In both HIV associated dementia in humans and its monkey model SIV encephalitis we find miR-21, a miRNA largely known for its link to oncogenesis, to be significantly upregulated in the brain. In situ hybridization of the diseased brain sections revealed induction of miR-21 in neurons. MiR-21 can be induced in neurons by prolonged N-methyl-D-aspartic acid receptor stimulation, an excitotoxic process active in HIV and other neurodegenerative diseases. Introduction of miR-21 into human neurons leads to pathological functional defects. Furthermore, we show that miR-21 …
