Young's schema theory offers a theoretical approach that relates negative childhood experiences, temperament, and early maladaptive schema, to the experience of negative affect and/or depression in adulthood. However, despite the widespread use of schema therapy in clinical practice, little research has explored the pathways theorized by Young. This study explored the pathways posited by Young and colleagues looking at the direct and indirect relationships among negative childhood experience, temperament, early maladaptive schema, and the experience of negative affect in adulthood. We present the development of a versatile spectroscopic imaging tool to allow for imaging with single-molecule sensitivity and high spatial resolution. The microscope allows for near-field and subdiffraction-limited far-field imaging by integrating a shear-force microscope on top of a custom inverted microscope design. The instrument has the ability to image in ambient conditions with optical resolutions on the order of tens of nanometers in the near field. A single low-cost computer controls the microscope with a field programmable gate array data acquisition card. Brain development and behavior are sensitive to environmental stimuli. To gain an understanding of how and to what extent environmental variations, particularly with regard to thermal stress and sensory input, affect brain development, function, and genomic activity, in this dissertation, three interrelated studies were conducted in Drosophila melanogaster.

The first study examined the effects of ecologically-relevant hyperthermia stress on development of the Drosophila mushroom body (MB), a conserved sensory integration and associative center in the insect brain. Model WALP peptides and "next generation" WALP-derived hydrophobic model peptides were employed to discover principles that govern protein-lipid interactions in biological membranes.

Ternary cholesterol-containing lipid mixtures were examined in the presence of WALP peptides of different lengths (acetyl-GWW(LA)_nLWWA-ethanolamide, with n between 3 and 8). Deuterium NMR spectra from labeled lipids reveal that WALP peptides may stabilize lipid ordered "raft" domains and therefore promote lipid phase separation, albeit to a minor extent. The results depend upon whether dioleolyl- or diphytanoyl-phosphatidylcholine is present as the fluid lipid component.

The committing step to apoptosis is regulated by a key protein, Bax, and its ability to integrate and form a pore structure at the outer mitochondrial membrane.

Geochemical models have suggested that the late Cambrian was characterized by a greenhouse climate with high pCO2. Furthermore, stableisotope analyses within the Great Basin have documented a large carbonate isotope (δ13Ccarb) excursion, known as the Steptoean Positive Carbon Isotope Excursion (SPICE). This event has been documented globally, and is interpreted as having resulted from enhanced organic carbon burial. Unless the size of carbon reservoirs in the Cambrian ocean was significantly different from those of the Cenozoic, this forcing should have resulted in a comparable carbon-isotope excursion in organic matter (δ13Corg). Diverse college aged females from a public university in the southwest were asked to participate in an online survey to determine their constraints to physical activity, as well as their current activity level. The groups were compared to establish the differences among the minority groups. Raymore, Crawford, and Godbey’s Hierarchical Model of Constraints was used as a framework when creating the survey. Data analysis showed only one significant difference among the racial/ethnic groups, when looking specifically at the intrapersonal, interpersonal, and structural constraints categories. This study focused on three main areas. These areas involved (1) the relationship among perceived stress, global orientation to life and indicators of physical, psychological and dyadic relational distress, (2) the effect of a brief CBT-based relaxation/guided visualization intervention on these outcome indicators of distress, and (3) changes in physiological indicators during the intervention sessions. The study population was drawn from couples who perceived themselves as living highly stressed lifestyles.

Results suggested that global orientation to life as reflected by differentiation of self inventory (DSI) and sense of coherence scale (SOCS) scale scores mediated the relationship between stress and distress. …

The National Weather Service’s (NWS) river forecast centers provide long-term water resource forecasts for the main river basins in the U.S. The NWS creates seasonal streamflow forecasts using an ensemble prediction model called the Extended Streamflow Prediction (ESP) software. ESP creates runoff volume forecasts by taking the current observed soil moisture and snowpack conditions in the basin and applying them to historical temperature and precipitation scenarios. The ESP treats every historic input year as a likely scenario of future basin conditions. Background: Military and industrial activities have lead to reported release of 2,4-dinitrotoluene (2,4DNT) into soil, groundwater or surface water. It has been reported that 2,4DNT can induce toxic effects on humans and other organisms. However the mechanism of 2,4DNT induced toxicity is still unclear. Although a series of methods for gene network construction have been developed, few instances of applying such technology to generate pathway connected networks have been reported.

This dissertation research comprised three studies focused on vision-specific skills, and their association with functional and academic outcomes for school-age students with visual impairment. The studies involved analysis of secondary data for 850 students with visual impairment who participated in the Special Education Elementary Longitudinal Study (SEELS). Data in the SEELS were gathered using direct assessment and parent and teacher responses for a nationally representative sample of elementary and middle school students.

The Basques are an ancient people, considered by many anthropologists to represent the oldest extant European population. Because of this, they have been the subject of numerous sociological and biological investigations. The Basque Diaspora, a relatively recent demographic expansion of the Basque population, has until now been overlooked in molecular genetic studies. Samples were taken from 53 individuals with Basque ancestry in Boise, Idaho, and the mitochondrial DNA (mtDNA) sequence variation of the first and second hypervariable regions were determined.
