Chapter 1

DNA barcoding based on the mitochondrial cytochrome c oxidase subunit I (COI) gene sequence is emerging as a useful tool for identifying unknown, whole or partial organisms to species level. However, the application of only a single mitochondrial marker for robust species identification has also come under some criticism due to the possibility of erroneous identifications resulting from species hybridizations and/or the potential presence of nuclear-mitochondrial psuedogenes. The addition of a complementary nuclear DNA barcode has therefore been widely recommended to overcome these potential COI gene limitations, especially in wildlife law enforcement applications where greater confidence in the …

Latitudinal Differences In The Hibernation Characteristics Of Woodchucks (Marmota Monax), Stam M. Zervanos, Christine R. Maher Phd, Jerry A. Waldvogel, Gregory L. Florant Feb 2010

Latitudinal Differences In The Hibernation Characteristics Of Woodchucks (Marmota Monax), Stam M. Zervanos, Christine R. Maher Phd, Jerry A. Waldvogel, Gregory L. Florant

Faculty Publications

There is little information on the phenotypic flexibility of hibernation characteristics within species. To address this issue, we observed differences in hibernation characteristics of three free-ranging populations of woodchucks (Marmota monax) distributed along a latitudinal gradient from Maine to South Carolina. Data from free-ranging animals exhibited a direct relationship between latitude and length of the hibernation season. As expected, woodchucks in the northern latitudes hibernated longer than those in the southern latitudes. Also, the length of interbout arousals decreased with increase in latitude, whereas the length of torpor bouts and the number of arousals increased. Thus, we observed phenotypic plasticity …

Phenotypic And Genotypic Characters Associated With The Polygyne Hybrid Imported Fire Ant, R. B. Garlapati, D. C. Cross, M. A. Caprio, S. Sabanadzovic, J. Chen

Midsouth Entomologist

A study was initiated to identify the origin of polygyny in the hybrid imported fire ant (Solenopsis invicta × richteri) and to confirm the association of polygyne phenotypic and genotypic characters. Polygyny is known to occur in both species in their native ranges in South America. Gp-9, a major gene in imported fire ants, is associated with the expression of social form. Variants of the monogyne allele, Gp-9B, and the polygyne allele, Gp-9b, have been sequenced for both species. The association of phenotypic and genotypic characters in a hybrid imported fire ant colony is strongly indicated by identification of multiple …

Behavioral And Chemical Ecology Of Marine Organisms With Respect To Tetrodotoxin, Becky L. Williams Feb 2010

Behavioral And Chemical Ecology Of Marine Organisms With Respect To Tetrodotoxin, Becky L. Williams

Biology Faculty Publications

The behavioral and chemical ecology of marine organisms that possess tetrodotoxin (TTX) has not been comprehensively reviewed in one work to date. The evidence for TTX as an antipredator defense, as venom, as a sex pheromone, and as an attractant for TTX-sequestering organisms is discussed. Little is known about the adaptive value of TTX in microbial producers; thus, I focus on what is known about metazoans that are purported to accumulate TTX through diet or symbioses. Much of what has been proposed is inferred based on the anatomical distribution of TTX. Direct empirical tests of these hypotheses are absent in …

Hydrological And Oceanic Effects On Polar Motion From Grace And Models, Shaunggen Jin, Don Chambers, Byron D. Tapley Feb 2010

Hydrological And Oceanic Effects On Polar Motion From Grace And Models, Shaunggen Jin, Don Chambers, Byron D. Tapley

Marine Science Faculty Publications

Terrestrial water storage (TWS) and ocean bottom pressure (OBP) are major contributors to the observed polar motion excitations, second only to atmospheric mass movement. However, quantitative assessment of the hydrological and oceanic effects on polar motion remains unclear because of the lack of global observations. In this paper, hydrological and oceanic mass excitations to polar motion are investigated using monthly TWS and OBP derived from the Gravity Recovery and Climate Experiment (GRACE) for January 2003 until December 2008. The results from this analysis are compared with hydrological model excitations from the European Center for Medium-Range Weather Forecasts (ECMWF) and oceanic …

Department of Biochemistry: Faculty Publications

Very-long-chain fatty acids (VLCFAs) are essential for many aspects of plant development and necessary for the synthesis of seed storage triacylglycerols, epicuticular waxes, and sphingolipids. Identification of the acetyl-CoA carboxylase PASTICCINO3 and the 3-hydroxy acyl-CoA dehydratase PASTICCINO2 revealed that VLCFAs are important for cell proliferation and tissue patterning. Here, we show that the immunophilin PASTICCINO1 (PAS1) is also required for VLCFA synthesis. Impairment of PAS1 function results in reduction of VLCFA levels that particularly affects the composition of sphingolipids, known to be important for cell polarity in animals. Moreover, PAS1 associates with several enzymes of …

Carbonates In Skeleton-Poor Seas: New Insights From Cambrian And Ordovician Strata Of Laurentia, Sara B. Pruss, Seth Finnegan, Woodward W. Fischer, Andrew H. Knoll Feb 2010

Carbonates In Skeleton-Poor Seas: New Insights From Cambrian And Ordovician Strata Of Laurentia, Sara B. Pruss, Seth Finnegan, Woodward W. Fischer, Andrew H. Knoll

Geosciences: Faculty Publications

Calcareous skeletons evolved as part of the greater EdiacaranCambrian diversification of marine animals. Skeletons did not become permanent, globally important sources of carbonate sediment, however, until the Ordovician radiation. Representative carbonate facies in a Series 3 (510501 Ma) Cambrian to Tremadocian succession from western Newfoundland, Canada, and Ordovician successions from the Ibex area, Utah, USA, show that, on average, Cambrian and Tremadocian carbonates contain much less skeletal material than do post-Tremadocian sediments. Petrographic point counts of skeletal abundance within facies and proportional facies abundance in measured sections suggest that later Cambrian successions contain on average volume, whereas the skeletal content …

Revised Relative Abundance Estimates And Temporal Activity Of Bats At Three Great Lakes National Parks Based On Acoustic Data, Bruce W. Miller Feb 2010

Revised Relative Abundance Estimates And Temporal Activity Of Bats At Three Great Lakes National Parks Based On Acoustic Data, Bruce W. Miller

United States National Park Service: Publications

Abstract

In this study, Miller re-analyzed acoustic bat data collected from June-August 2003 that was part of a baseline inventory of bat species in three national parks in the Lake Superior region. While the original study presented base-line data on the presence/absence of bat species in these parks, this reanalysis provides estimates of relative abundance and temporal activity of the identified species. Using a suite of recently developed acoustic analysis tools, Miller created species specific filters. This allowed parsing of calls from non-fragmented sequences and differentiate between two species, Myotis septentrionalis and Myotis lucifugus, that were combined into a …

Salivary Nicotine And Cotinine Concentrations In Unstimulated And Stimulated Saliva, Noor Zurani Robson, A.J. Bond, K. Wolff Feb 2010

Salivary Nicotine And Cotinine Concentrations In Unstimulated And Stimulated Saliva, Noor Zurani Robson, A.J. Bond, K. Wolff

Research Publications (2006 to 2010)

Salivary nicotine and salivary cotinine is widely used in clinical and epidemiological studies to validate smoking cessation. However, the reported collection for salivary nicotine and salivary cotinine vary by technique and duration. This study investigated the influence of salivary collection by unstimulation and stimulation technique of the concentration of salivary nicotine and salivary cotinine. It was found that unstimulated technique produced the highest salivary nicotine concentration, whereas stimulated technique produced the highest salivary cotinine concentration. The results of this study suggest that it is important to standardise salivary nicotine and cotinine collection technique.

Bond Strength Of Adhesives To Dentin Contaminated With Smoker's Saliva, Lilliam M Pinzon, Makoto Oguri, Kathy O'Keefe, Vladimir Dusevish, Paulette Spencer, John M Powers, Grayson W Marshall

Faculty, Staff and Student Publications

The purpose of this study was to determine the effects of contamination with smoker's and non-smoker's saliva on the bond strength of resin composite to superficial dentin using different adhesive systems. The interfacial structure between the resin and dentin was evaluated for each treatment using environmental scanning electron microscopy (ESEM). Freshly extracted human molars were ground with 600-grit SiC paper to expose the superficial dentin. Adhesives [One-Up-Bond-F-Plus (OUFP) and Adper-Prompt-L-Pop (APLP)] and resin composite (TPHSpectrum) were bonded to the dentin (n = 8/group, 180 total specimens) under five surface conditions: control (adhesive applied following manufacturers' instructions); saliva, then 5-s air …

The Presence Of Micropterus Salmoides (Largemouth Bass) Influences The Populations Of Rana Draytonii (California Red-Legged Frog) And Pseudacris Regilla (Pacific Treefrog) In Two Ponds In Santa Barbara County, California, Kenneth Lee Gilliland

Master's Theses

Alien fish have been implicated in the decline of Rana draytonii (California red-legged frog) and Pseudacris regilla (Pacific treefrog) populations. Micropterus salmoides (largemouth bass) is a common sport fish that has been introduced into the sympatric range of these two anuran species; however, no studies have investigated the individual influence that this alien fish has on these two species. I conducted M. salmoides control or eradication experiments during a four year study in two ponds within the Transverse Mountain Range of Santa Barbara County, California. Changes in the densities of all life stages of R. draytonii and P. regilla were …

We have used multiplexed high-throughput sequencing to characterize changes in small RNA populations that occur during viral infection in animal cells. Small RNA-based mechanisms such as RNA interference (RNAi) have been shown in plant and invertebrate systems to play a key role in host responses to viral infection. Although homologs of the key RNAi effector pathways are present in mammalian cells, and can launch an RNAi-mediated degradation of experimentally targeted mRNAs, any role for such responses in mammalian host-virus interactions remains to be characterized. Six different viruses were examined in 41 experimentally susceptible and resistant host systems. We identified virus-derived …

Kaposi's Sarcoma-Associated Herpesvirus Glycoproteins B And K8.1 Regulate Virion Egress And Synthesis Of Vascular Endothelial Growth Factor And Viral Interleukin-6 In Bcbl-1 Cells, R. Subramanian, I. Sehgal, O. D'Auvergne, K. G. Kousoulas Feb 2010

Kaposi's Sarcoma-Associated Herpesvirus Glycoproteins B And K8.1 Regulate Virion Egress And Synthesis Of Vascular Endothelial Growth Factor And Viral Interleukin-6 In Bcbl-1 Cells, R. Subramanian, I. Sehgal, O. D'Auvergne, K. G. Kousoulas

Faculty Publications

Kaposi's sarcoma-associated herpesvirus (KSHV) viral glycoproteins play important roles in the infectious life cycle and have been implicated in KSHV-associated endothelial cell transformation, angiogenesis, and KS-induced malignancies. KSHV-associated primary effusion lymphomas (PELs) secrete high levels of vascular endothelial growth factor (VEGF) and viral interleukin-6 (vIL-6) in vitro and VEGF, vIL-6, and basic-fibroblast growth factor (b-FGF) in mouse xenografts. KSHV-encoded glycoproteins B (gB) and K8.1 stimulate VEGF secretion, most likely mediated by direct or indirect binding to cell surface receptors, including the gB-specific αVβ3 and α3β1 integrins. In this study, the short interfering RNA (siRNA)-mediated inhibition of either gB or K8.1 …

The Combined Effects Of Rivers And Refugia Generate Extreme Cryptic Fragmentation Within The Common Ground Skink (Scincella Lateralis), Nathan D. Jackson, Christopher C. Austin Feb 2010

The Combined Effects Of Rivers And Refugia Generate Extreme Cryptic Fragmentation Within The Common Ground Skink (Scincella Lateralis), Nathan D. Jackson, Christopher C. Austin

Faculty Publications

Rivers can act as both islands of mesic refugia for terrestrial organisms during times of aridification and barriers to gene flow, though evidence for long-term isolation by rivers is mixed. Understanding the extent to which riverine barrier effects can be heightened for populations trapped in mesic refugia can help explain maintenance and generation of diversity in the face of Pleistocene climate change. Herein, we implement phylogenetic and population genetic approaches to investigate the phylogeographic structure and history of the ground skink, Scincella lateralis, using mtDNA and eight nuclear loci. We then test several predictions of a river-refugia model of diversification. …
