An integrated optical waveguide type surface plasmon resonance (SPR) sensor having an optical waveguide with a corresponding SPR sensing area, photodetectors, and wave-length tunable laser or any kind of external tunable laser source/coupler formed on a substrate. Naloxone's Pentapeptide Binding Site On Filamin A Blocks Mu Opioid Receptor–Gs Coupling And Creb Activation Of Acute Morphine, Hoau-Yan Wang, Lindsay H. Burns

Chronic morphine causes the mu opioid receptor (MOR) to switch its coupling from Gi/o to Gs, resulting in excitatory signaling via both Gαs and its Gβγ dimer. Ultra-low-dose naloxone (NLX) prevents this switch and attenuates opioid tolerance and dependence. This protective effect is mediated via a high-affinity interaction of NLX to a pentapeptide region in c-terminal filamin A (FLNA), a scaffolding protein interacting with MOR. In organotypic striatal slice cultures, we now show that acute morphine induces a dose-dependent Go-to-Gs coupling switch at 5 and 15 min that resolves by 1 hr. Growth factor binding compounds having a plurality of acyclic isophthalic acid groups attached to a non-peptide organic scaffold and pharmaceutical compositions of the same are disclosed. Methods of administering and using the growth factor binding compounds or the growth factor binding compositions are also taught. These novel growth factor binding compounds are useful for treating angiogenesis, excessive cellular proliferation, tumor growth, and a combination thereof as well as inhibiting growth factor binding to cells and phosphorylation.

N-Thiolated 2-Oxazolidinone-Derived Antibiotics, Edward Turos, Rajesh Kumar Mishra

This invention describes the discovery and synthesis of N-thiolated 2-oxazolidinones as a new class of anti bacterial agents. These compounds can be synthesized from 2-oxazolidinones by Ndeprotection and N-sulfenylation. These new substances were found to exhibit potent anti-bacterial activity, including bacteriostatic properties against Staphylococcus spp., including methicillin resistant Staphylococcus aureus (MRSA), and Bacillus spp., including Bacillus anthracis.

Glutathione Diminishes Tributyltin- And Dibutyltin-Induced Loss Of Lytic Function In Human Natural Killer Cells, Jeralyn J. Powell, Mclisa V. Davis, Margaret M. Whalen

This study investigated whether reduced glutathione (GSH) was able to alter the negative effects of tributyltin (TBT) or dibutyltin (DBT) on the lytic function of human natural killer (NK) cells. NK cells are an initial immune defense against the development of tumors or viral infections. TBT and DBT are widespread environmental contaminants, due to their various industrial applications. Both TBT and DBT have been shown to decrease the ability of NK cells to lyse tumor cells (lytic function). The results indicated that the presence of GSH during the exposure of NK cells to TBT or DBT diminished the negative effect …

This dissertation focuses on the systematic study of techniques for characterization and synthesis of nanoscale materials. We have achieved several goals. Firstly, high number density uniform zinc oxide nanostructure growth has been achieved using thermal evaporation, through control of experimental parameters that include source material temperature, substrate temperature, substrate material, gas flow rate, and choice of catalyst. Aligned zinc oxide nanowires, randomly oriented zinc oxide nanowires, zinc oxide container-shaped structures, and zinc oxide nanobelts have been synthesized with high yield. Secondly, using a one parameter family of lattice fringe geometry curves, we show how to examine the epitaxial relationship between …

Nanotechnology is a term used to describe nanometer scaled systems. This thesis presents various nanomaterials and systems for the investigation of biologically relevant analytes in general, and in particular for their detection, decontamination, or destruction. The validation of short peptide fragments as models for protein aggregation is initially discussed through applying spectroscopic and microscopic techniques to Langmuir monolayer surface chemistry. Following this validation, the use of nanogold as a photoablative material for the destruction of aggregated protein is investigated. Subsequently, the versatility of nanotechnology is shown by investigating a different form of nanogold; namely, gold quantum dots and the interesting …

Modulation of host signaling by the products of microbial activity in the gut may affect weight gain and fat formation.
The relationship between humans and the population of indigenous microorganisms in their intestines (the gut microbiota) is ancient and important. In a recent survey exploring the relationship between mammals and their microbiota it was found that individuals of the same species were more likely to have a similar gut microbiota than mammals of different species [1]. This observation held true regardless of the geographic separation between the two hosts. These results indicate that the composition of the microbiota is dependent …

Expression of human profilin-I does not complement the temperature-sensitive cdc3-124 mutation of the single profilin gene in fission yeast Schizosaccharomyces pombe, resulting in death from cytokinesis defects. Human profilin-I and S. pombe profilin have similar affinities for actin monomers, the FH1 domain of fission yeast formin Cdc12p and poly-L-proline (Lu, J., and Pollard, T. D. (2001) Mol. Biol. Cell 12, 1161-1175), but human profilin-I does not stimulate actin filament elongation by formin Cdc12p like S. pombe profilin. Two crystal structures of S. pombe profilin and homology models of S. pombe profilin bound to actin show how the two profilins bind …

Dietary Resistant Starch Increases Hypothalamic Pomc Expression In Rats, Li Shen, Michael J. Keenan, Roy J. Martin, Richard T. Tulley, Anne M. Raggio, Kathleen L. Mccutcheon, Jun Zhou

Resistant starch (RS) is fermentable dietary fiber. Inclusion of RS in the diet causes decreased body fat accumulation and altered gut hormone profile. This study investigates the effect of feeding RS on the neuropeptide messenger RNA (mRNA) expressions in the arcuate nucleus (ARC) of the hypothalamus and whether vagal afferent nerves are involved. The rats were injected intraperitoneally with capsaicin to destroy unmyelinated small vagal afferent nerve fibers. The cholecystokinin (CCK) food suppression test was performed to validate the effectiveness of the capsaicin treatment.
