Enzymatic Digestion Of Single Dna Molecules Anchored On Nanogold-Modified Surfaces, Junhong Lü, Ming Ye, Na Duan, Bin Li

To study enzyme-DNA interactions at single molecular level, both the attachment points and the immediate surroundings of surfaces must be carefully considered such that they do not compromise the structural information and biological properties of the sample under investigation. The present work demonstrates the feasibility of enzymatic digestion of single DNA molecules attached to nanoparticle-modified surfaces. With Nanogold linking DNA to the mica surface by electrostatic interactions, advantageous conditions with fewer effects on the length and topography of DNA are obtained, and an appropriate environment for the activities of DNA is created. Despite recent advances in the understanding of the molecular bases of hematological malignancies, the specific mechanisms on how they originate and why some subtypes are more prevalent than others still remain to be elucidated. These two important aspects have been even more difficult to analyze when dealing with individuals under immune suppression because other factors must be considered. Questions still remain as to why individuals with AIDS tend to develop lymphoproliferative disorders differently from those observed in individuals under iatrogenic immunosuppressive therapy. Muc4 Modulation Of Ligand-Independent Erbb2 Signaling, Goldi A. Kozloski

The membrane mucin Muc4 is a heterodimer, bi-functional glycoprotein complex that is normally expressed in epithelial tissue. Functional studies on the extracellular mucin subunit of Muc4 have shown that it acts to promote anti-adhesion properties by sterically interfering with cell-cell and cell-matrix interactions and that the extent of this effect is directly associated with the number of tandem repeats on this subunit. Functional studies on the transmembrane subunit of Muc4 have shown that this subunit participates in intracellular signaling through interaction with the receptor tyrosine kinase ErbB2. Non-group health insurance is coverage that individuals purchase on their own rather than as part of a group. Most states currently permit non-group insurers to underwrite, a process whereby an insurer assesses the health and other characteristics of individuals to determine their likely utilization of health services or risk; insurers then use this assessment to determine whether they will offer coverage and the premium they will charge. Policymakers have identified underwriting and related practices in non-group markets as a target for reform to enable broader access for the currently uninsured. Why Some Species Of Birds Do Not Avoid Inbreeding: Insights From New Zealand Robins And Saddlebacks, Ian G. Jamieson, Sabrina S. Taylor, Lisa N. Tracy, Hanna Kokko, Doug P. Armstrong

When dispersal options are limited and encounters with relatives are likely, individuals need to recognize and avoid mating with kin to avoid the fitness costs of close inbreeding. New Zealand robins and saddlebacks are genetically monogamous and possess life-history traits that predict they should show zero tolerance of close inbreeding. However, of 11 population-years of pedigree data, there was evidence of inbreeding avoidance in only 1 year. We also found no indication that incestuous pairings were avoided or that individuals were choosing genetically dissimilar mates based on microsatellite DNA analysis. Phylogenetic Analyses Reveal Monophyletic Origin Of The Ergot Alkaloid Gene Dmaw In Fungi, Miao Liu, Daniel G. Panaccione, Christopher L. Schardl

Ergot alkaloids are indole-derived mycotoxins that are important in agriculture and medicine. Ergot alkaloids are produced by a few representatives of two distantly related fungal lineages, the Clavicipitaceae and the Trichocomaceae. Comparison of the ergot alkaloid gene clusters from these two lineages revealed differences in the relative positions and orientations of several genes. The question arose: is ergot alkaloid biosynthetic capability from a common origin? We used a molecular phylogenetic approach to gain insights into the evolution of ergot alkaloid biosynthesis. The 4-γ,γ-dimethylallyltryptophan synthase gene, dmaW, encodes the first step in the pathway. Amino acid sequences deduced …

Definitive X-ray crystallographic evidence is obtained for a single hole (or a polaron) to be uniformly distributed on the three equivalent 1,2-dimethoxybenzenoid (or veratrole) rings in the hexamethoxytriptycene cation radical. This conclusion is further supported by electrochemical analysis and by the observation of an intense near-IR transition in its electronic spectrum, as well as by comparison of the spectral and electrochemical characteristics with the model compounds containing one and two dimethoxybenzene rings.

Many cell biologists perform analysis of multinucleated cell data in order to better under- stand the mechanisms that regulate cell division. Sbalzarini, et al., have developed methods for automatically tracking nuclei in cell data in order to aid in this time-consuming analysis. In this paper, we present an implementation of the Sbalzarini tracking algorithm, introduce a new algorithm we developed which is able to identify mitosis events, and present other software tools we have developed to aid in the automated detection of nucleus data.

Classroom Response Systems (CRS) are an in-class technology used to poll students and instantly display an aggregate representation of their responses. CRS have been around since the 1970s and have become increasingly more popular in higher education lecture halls. Even though technology, specifically computers and communications, has improved significantly since the 1970s, CRS have remained surprisingly unchanged. The purpose of this project was to develop an innovative web-based CRS using web services. Structural Basis Of Error-Prone Replication And Stalling At A Thymine Base By Human Dna Polymerase Iota, Kevin N. Kirouac, Hong Ling

Human DNA polymerase iota (pol iota) is a unique member of Y-family polymerases, which preferentially misincorporates nucleotides opposite thymines (T) and halts replication at T bases. The structural basis of the high error rates remains elusive. We present three crystal structures of pol complexed with DNA containing a thymine base, paired with correct or incorrect incoming nucleotides. A narrowed active site supports a pyrimidine to pyrimidine mismatch and excludes Watson-Crick base pairing by pol. The template thymine remains in an anti conformation irrespective of incoming nucleotides. Incoming ddATP adopts a syn conformation with reduced base stacking, whereas incorrect dGTP and …
