Broad Range Amino Acid Specificity Of Rna-Dependent Lipid Remodelling By Multiple Peptide Resistance Factors, Hervé Roy, Michael Ibba Sep 2009

Broad Range Amino Acid Specificity Of Rna-Dependent Lipid Remodelling By Multiple Peptide Resistance Factors, Hervé Roy, Michael Ibba

Biology, Chemistry, and Environmental Sciences Faculty Articles and Research

Aminoacylphosphatidylglycerol synthases (aaPGSs) are multiple peptide resistance factors that transfer amino acids from aminoacyl-tRNAs to phosphatidylglycerol (PG) in the cytoplasmic membrane. Aminoacylation of PG is used by bacteria to decrease the net negative charge of the cell envelope, diminishing affinity for charged molecules and allowing for adaptation to environmental changes. Lys-PGS, which transfers lysine to PG, is essential for the virulence of certain pathogens, providing resistance to both host cationic antimicrobial peptides and therapeutic antibiotics. Ala-PGS was also recently described, but little is known about the possible activities of other members of the highly diverse aaPGS family of proteins. Systematic …

In response to DNA damage, eukaryotic cells activate a series of DNA damage-dependent pathways that serve to arrest cell cycle progression and remove DNA damage. Coordination of cell cycle arrest and damage repair is critical for maintenance of genomic stability. However, this process is still poorly understood. Nucleotide excision repair (NER) and the ATR-dependent cell cycle checkpoint are the major pathways responsible for repair of UV-induced DNA damage. Here we show that ATR physically interacts with the NER factor Xeroderma pigmentosum group A (XPA). Using a mass spectrometry-based protein footprinting method, we found that ATR interacts with a helixturn-helix motif …

Large-Scale Patterns In Trematode Richness And Infection Levels In Marine Crustacean Hosts, David W. Thieltges, Brian L. Fredensborg, Anja Studer, Robert Poulin Sep 2009

Large-Scale Patterns In Trematode Richness And Infection Levels In Marine Crustacean Hosts, David W. Thieltges, Brian L. Fredensborg, Anja Studer, Robert Poulin

School of Integrative Biological & Chemical Sciences (Formerly Dept. of Biology)

Little is known about the patterns of variation in parasitism in marine hosts. Trematodes, the dominant parasites in intertidal systems, are transmitted from their first intermediate hosts (snails) to a range of second intermediate hosts, including crustaceans. Using published studies of trematode infections in crustacean hosts, we investigated general patterns of variation in trematode species richness and infection levels (i.e. percentage of hosts infected and mean number of individual parasites per host). Since the production and release of infective stages in snails is strongly temperature dependent, we also investigated a potential decrease in trematode infection levels with increasing latitude (as …

Defective Rbosomal Products In Blebs (Dribbles) And Methods Of Use To Stimulate An Immune Response, Hong-Ming Hu Sep 2009

Defective Rbosomal Products In Blebs (Dribbles) And Methods Of Use To Stimulate An Immune Response, Hong-Ming Hu

Technology Transfer - Patents

Methods are disclosed for producing defective ribosomal products (DRiPs) in blebs (DRibbles) by contacting cells with a proteasome inhibitor, and in some examples also an autophagy inducer, thereby producing treated cells. DRibbles can be used to load antigen presenting cells (APCs), thereby allowing the APCs to present the DRiPs and antigenic frag ments thereof. Immunogenic compositions that include treated cells, isolated DRibbles, or DRibble-loaded APCs are also disclosed. Methods are also provided for using treated cells, isolated DRibbles, or DRibble-loaded APCs to stimu late an immune response, for example in a subject. For example, DRibbles obtained from a tumor cell …

Xmc Mediates Xctr1-Independent Morphogenesis In Xenopus Laevis†, Tomomi Haremaki, Daniel C. Weinstein Sep 2009

Xmc Mediates Xctr1-Independent Morphogenesis In Xenopus Laevis†, Tomomi Haremaki, Daniel C. Weinstein

Publications and Research

In the frog, Xenopus laevis, fibroblast growth factor (FGF) signaling is required for both mesoderm formation and the morphogenetic movements that drive the elongation of the notochord, a dorsal mesodermal derivative; the coordination of these distinct roles is mediated by the Xenopus Ctr1 (Xctr1) protein: maternal Xctr1 is required for mesodermal differentiation, while the subsequent loss of Xctr1 promotes morphogenesis. The signaling cascade activated by FGF in the presence of Ctr1 has been well characterized; however, the Xctr1-independent, FGF-responsive network remains poorly defined. We have identified Xenopus Marginal Coil (Xmc) as a gene whose expression is highly enriched following Xctr1 …

Accelerometer Based Measurement For The Mapping Of Neck Surface Vibrations During Vocalized Speech, Mark Nolan, Brian Madden, Edward Burke Sep 2009

Accelerometer Based Measurement For The Mapping Of Neck Surface Vibrations During Vocalized Speech, Mark Nolan, Brian Madden, Edward Burke

Conference Papers

This paper presents a detailed study of the vibrations on the surface of the neck during a vocalization of predefined fundamental frequency and intensity. This study was carried out as part of a wider investigation into the use of laryngeal vibrations as a channel of communication. Another potential application of this study is in identifying a suitable location for a hands-free electro-larynx for laryngectomees.

An analog accelerometer, with dimensions 5x5x1.6mm and of mass 80mg, was used to perform the measurements. It was connected to a 12-bit analog-to-digital converter via single strands of insulated wire with a diameter of 100μm. The …

Three-Dimensional Structure Of The Enveloped Bacteriophage Φ12: An Incomplete T = 13 Lattice Is Superposed On An Enclosed T = 1 Shell, Hui Wei, R. Holland Cheng, John Berriman, William J. Rice, David L. Stokes, A. Katz, David Gene Morgan, Paul Gottlieb

Publications and Research

Background Bacteriophage φ12 is a member of the Cystoviridae, a unique group of lipid containing membrane enveloped bacteriophages that infect the bacterial plant pathogen Pseudomonas syringae pv. phaseolicola. The genomes of the virus species contain three double-stranded (dsRNA) segments, and the virus capsid itself is organized in multiple protein shells. The segmented dsRNA genome, the multi-layered arrangement of the capsid and the overall viral replication scheme make the Cystoviridae similar to the Reoviridae. Methodology/Principal Findings We present structural studies of cystovirus φ12 obtained using cryo-electron microscopy and image processing techniques. We have collected images of isolated φ12 virions and generated …

Family Therapists' Responses To Monopolizing, Blaming, Critical And Unempathic Behavior In Parents, Andrew David Sussman Sep 2009

Family Therapists' Responses To Monopolizing, Blaming, Critical And Unempathic Behavior In Parents, Andrew David Sussman

Theses, Dissertations, and Projects

This research was designed to address the question: "How do family therapists respond to monopolizing, blaming, critical and unempathic behavior from parents in family therapy. I was interested to see if they viewed the presentation as resistance, narcissism, or was it attributed to something else? I was also interested in the therapist's background, theoretical framework, training, and how they viewed family interventions. I hypothesized that family therapists would respond to monopolizing, blaming, critical and unempathic behavior in parents in a way that was influenced more by clinical practice experience than theoretical orientation. The instrument was a survey with closed and …

A Modification To The Group Delay And Simulated Annealing Technique For Characterization Of Peripheral Nerve Fiber Size Distributions For Non-Deterministic Sampled Data, Robert B. Szlavik Sep 2009

A Modification To The Group Delay And Simulated Annealing Technique For Characterization Of Peripheral Nerve Fiber Size Distributions For Non-Deterministic Sampled Data, Robert B. Szlavik

Biomedical Engineering

The ability to determine the characteristics of peripheral nerve fiber size distributions would provide additional information to clinicians for the diagnosis of specific pathologies of the peripheral nervous system. Investigation of these conditions, using electro-diagnostic techniques, is advantageous in the sense that such techniques tend to be minimally invasive yet provide valuable diagnostic information. One of the principal electro-diagnostic tools available to the clinician is the nerve conduction velocity test. While the peripheral nerve conduction velocity test can provide useful information to the clinician regarding the viability of the nerve under study, it is a single parameter test that yields …

The emergence of terrestrial life witnessed the need for more sophisticated circulatory systems. This has evolved in birds, mammals and crocodilians into complete septation of the heart into left and right sides, allowing separate pulmonary and systemic circulatory systems, a key requirement for the evolution of endothermy(1-3). However, the evolution of the amniote heart is poorly understood. Reptilian hearts have been the subject of debate in the context of the evolution of cardiac septation: do they possess a single ventricular chamber or two incompletely septated ventricles(4-7)? Here we examine heart development in the red-eared slider turtle, Trachemys scripta elegans (a …
