Methods And Devices For Cryopreservation Of Biological Cells And Tissues, Dayong Gao, Gary Van Zant, Xiangdong Cui

Novel methods, compositions, and devices for achieving optimal cooling of living cells during cryopreservation are disclosed. In one aspect, the method comprises gradually cooling the cell to a first predetermined temperature, followed by rapidly cooling the cell to a second predetermined temperature. In another aspect, a device is described for achieving a desired cooling rate for a cell, comprising a first container for holding a cell, a second container for holding the first container, and optionally a frame for holding the first container in a spaced apart relationship with the second container. There is provided a method of modifying vasoactivity by regulating a soluble Aβ pro-inflammatory pathway. Also provided is a method of modifying inflammatory reactions in microglia and neurons by regulating a soluble Aβ pro-inflammatory pathway. A method of treating patients with vascular disease by modifying an intracellular soluble Aβ pro-inflammatory pathway is also provided. A pharmaceutical composition consisting essentially of an effective amount of a soluble Aβ pro-inflammatory pathway regulator in a pharmaceutically effective carrier is also provided.

BACKGROUND: Equine infectious anemia virus (EIAV), a lentivirus that infects horses, has been utilized as an animal model for the study of HIV. Furthermore, the disease associated with the equine lentivirus poses a significant challenge to veterinary medicine around the world. As with all lentiviruses, EIAV has been shown to have a high propensity for genomic sequence and antigenic variation, especially in its envelope (Env) proteins. Recent studies have demonstrated Env variation to be a major determinant of vaccine efficacy, emphasizing the importance of defining natural variation among field isolates of EIAV. Cells of Saccharomyces cerevisiae lacking Apq12, a nuclear envelope (NE)-endoplasmic reticulum (ER) integral membrane protein, are defective in assembly of nuclear pore complexes (NPCs), possibly because of defects in regulating membrane fluidity. We identified BRR6, which encodes an essential integral membrane protein of the NE-ER, as a dosage suppressor of apq12 Delta. Cells carrying the temperature-sensitive brr6-1 allele have been shown to have defects in nucleoporin localization, mRNA metabolism and nuclear transport. Electron microscopy revealed that brr6-1 cells have gross NE abnormalities and proliferation of the ER. brr6-1 cells were hypersensitive to compounds that affect membrane biophysical properties and to …

Rab GTPases and their effectors mediate docking, the initial contact of intracellular membranes preceding bilayer fusion. However, it has been unclear whether Rab proteins and effectors are sufficient for intermembrane interactions. We have recently reported reconstituted membrane fusion that requires yeast vacuolar SNAREs, lipids, and the homotypic fusion and vacuole protein sorting (HOPS)/class C Vps complex, an effector and guanine nucleotide exchange factor for the yeast vacuolar Rab GTPase Ypt7p. We now report reconstitution of lysis-free membrane fusion that requires purified GTP-bound Ypt7p, HOPS complex, vacuolar SNAREs, ATP hydrolysis, and the SNARE disassembly catalysts Sec17p and Sec18p. Purpose: The purpose of our study is to evaluate our institutional experience of treating Tubular Carcinoma of the Breast (TC) and Invasive Ductal Carcinoma (IDC) with Breast Conservation Therapy (BCT), consisting of conservative surgery (CS) and radiation therapy (RT), and to compare clinical-pathologic features and long-term outcomes. Materials and Methods: A review of our institutions tumor registry from 1975-2007 was performed, followed by a central pathology review of available slides, yielding 71 cases of Stage I/II TC and 2238 cases of Stage I/II IDC treated with BCT. Schefflera is the largest genus in the angiosperm family Araliaceae, with about 900 species, of which c. 300 belong to five subgeneric groups in the Neotropical region. Previous phylogenetic studies of Schefflera have been limited to a small number of species from this region, and very little is know about phylogenetic relationships in the Brazilian-centered Didymopanax group of this genus. Therefore, to gain a better understanding of the diversity and evolution of the Didymopanax group of Schefflera, I investigated the systematics of these plants in the broader context of the entire Neotropical clade. In order to achieve a more accurate input value for groundwater age determining models, δ13CVPDB values for soil gas were evaluated at 50 cm depths in locations throughout Utah in order to define correlations between δ13C and environmental parameters. 16 sites were chosen that exploited large changes in elevation and latitude which provided variations in climate, precipitation, plant community, etc. Gas samples were collected over 1-1½ years, and soil samples were collected at depth during installations. Field and laboratory studies were also used to evaluate CO2 and δ13C change with depth. It was discovered that in mountainous recharge areas, the …

Modeling Multilevel Sleep Transitional Data Via Poisson Log-Linear Multilevel Models, Bruce J. Swihart

This paper proposes Poisson log-linear multilevel models to investigate population variability in sleep state transition rates. We specifically propose a Bayesian Poisson regression model that is more flexible, scalable to larger studies, and easily fit than other attempts in the literature. We further use hierarchical random effects to account for pairings of individuals and repeated measures within those individuals, as comparing diseased to non-diseased subjects while minimizing bias is of epidemiologic importance. We estimate essentially non-parametric piecewise constant hazards and smooth them, and allow for time varying covariates and segment of the night comparisons. Swelling-activated Cl− current (ICl,swell) is an outwardly-rectifying current that plays an important role in cardiac electrical activity, cellular volume regulation, apoptosis, and acts as a potential effector of mechanoelectrical feedback. Persistent activation of ICl,swell has been observed in a number of models of cardiovascular disease. Previously we showed that angiotensin II (Ang II), endothelin-1 (ET-1), endothelial growth factor receptor (EGFR), and reactive oxygen species (ROS) produced by NADPH oxidase (NOX) and mitochondria are involved in the activation of ICl,swell by both osmotic swelling and Beta1 integrin stretch.
