The reaction of 5,10,15-triarylcorrole with 4-amino-4H-1,2,4-triazole provides another example of corroie ring expansion to give the corresponding 6-azahemiporphycene, a novel porphyrin analogue. The facile oxidation of the corroie ring is a required step for the ring expansion and for this reason the reaction fails in the case of corroles bearing meso-phenyl groups carrying electron-withdrawing substituents. Sterlc requirements also limited the scope of the reaction, which is not successful In the case of 2,6-disubstituted meso-aryl corroles. The occurrence of an initial oxidation is further supported by formation of the 6-azahemiporphycene derivative when the reaction is carried out under the same conditions, …

Small-Molecule Screening Using A Human Primary Cell Model Of Hiv Latency Identifies Compounds That Reverse Latency Without Cellular Activation, Hung Chih Yang, Sifei Xing, Liang Shan, Karen O'Connell

University Faculty Publications and Creative Works

The development of highly active antiretroviral therapy (HAART) to treat individuals infected with HIV-1 has dramatically improved patient outcomes, but HAART still fails to cure the infection. The latent viral reservoir in resting CD4+ T cells is a major barrier to virus eradication. Elimination of this reservoir requires reactivation of the latent virus. However, strategies for reactivating HIV-1 through nonspecific T cell activation have clinically unacceptable toxicities. We describe here the development of what we believe to be a novel in vitro model of HIV-1 latency that we used to search for compounds that can reverse latency. Human primary CD4+ …

Decreased Replication Origin Activity In Temporal Transition Regions, Zeqiang Guan, Christina M. Hughes, Settapong Kosiyatrakul, Paolo Norio, Ranjan Sen, Steven Fiering

Dartmouth Scholarship

In the mammalian genome, early- and late-replicating domains are often separated by temporal transition regions (TTRs) with novel properties and unknown functions. We identified a TTR in the mouse immunoglobulin heavy chain (Igh) locus, which contains replication origins that are silent in embryonic stem cells but activated during B cell development. To investigate which factors contribute to origin activation during B cell development, we systematically modified the genetic and epigenetic status of the endogenous Igh TTR and used a single-molecule approach to analyze DNA replication. Introduction of a transcription unit into the Igh TTR, activation of gene transcription, …

Effects Of Geomorphic Setting On Shallow-Groundwater Exchange And Water Temperature Of Salmon-Bearing Headwater Streams Of The Lower Kenai Peninsula, Alaska, Jason C. Bellino Nov 2009

Effects Of Geomorphic Setting On Shallow-Groundwater Exchange And Water Temperature Of Salmon-Bearing Headwater Streams Of The Lower Kenai Peninsula, Alaska, Jason C. Bellino

USF Tampa Graduate Theses and Dissertations

Temperature is an important physical characteristic of headwater streams that controls the presence and health of juvenile salmonids. Surface water temperature is controlled by many factors including exchanges with groundwater. A study of the hydrology of wetlands associated with headwater streams on the Lower Kenai Peninsula was conducted to determine the effect of geomorphic setting on groundwater discharge to streams and ultimately on in-stream water temperature. Attention was focused on drainage-way and discharge slope wetlands as two end-members of the geomorphic settings of the study area.

Data were collected at 18 study reaches spanning four major watersheds in the study …

Microbial Population Analysis In Leachate From Simulated Solid Waste Bioreactors And Evaluation Of Genetic Relationships And Prevalence Of Vancomycin Resistance Among Environmental Enterococci, Bina S. Nayak Nov 2009

Microbial Population Analysis In Leachate From Simulated Solid Waste Bioreactors And Evaluation Of Genetic Relationships And Prevalence Of Vancomycin Resistance Among Environmental Enterococci, Bina S. Nayak

USF Tampa Graduate Theses and Dissertations

Degradation of the several million tons of solid waste produced in the U.S. annually is microbially mediated, yet little is known about the structure of prokaryotic communities actively involved in the waste degradation process. In the first study, leachates generated during degradation of municipal solid waste (MSW) in the presence (co-disposal) or absence of biosolids were analyzed using laboratory-scale bioreactors over an eight-month period. Archaeal and bacterial community structures were investigated by denaturing gradient gel electrophoresis (DGGE) targeting 16S rRNA genes. Regardless of waste composition, microbial communities in bioreactor leachates exhibited high diversity and temporal trends. Methanogen sequences from a …

The Citric Acid Cycle Of Thiomicrospira Crunogena: An Oddity Amongst The Proteobacteria, Ishtiaque Quasem Nov 2009

The Citric Acid Cycle Of Thiomicrospira Crunogena: An Oddity Amongst The Proteobacteria, Ishtiaque Quasem

USF Tampa Graduate Theses and Dissertations

Thiomicrospira crunogena,

a deep-sea hydrothermal vent chemolithoautotroph,

uses the Calvin-Bensen-Bassham cycle to fix carbon. To meet its biosynthetic needs for

oxaloacetate, oxoglutarate, and succinyl-coA, one would expect that this obligately

autotrophic

Gammaproteobacterium would use a ‘wishbone’ version of the citric acid

cycle (CAC) to synthesize the intermediates necessary for biosynthesis, instead of the

fully oxidative version to minimize carbon loss as carbon dioxide. However, upon

examination of its complete genome sequence, it became apparent that this organism did

not fulfill this expectation.

Instead of a wishbone pathway,

T. crunogena appears to run a fully oxidative

CAC. The cycle is ‘locked’ …

No Beauty We Could Desire-A Lutheran Evaluation Of C. S. Lewis' Sehnsucht, Jonathan Graf Nov 2009

No Beauty We Could Desire-A Lutheran Evaluation Of C. S. Lewis' Sehnsucht, Jonathan Graf

Master of Art Theology Thesis

Together, the experiences and glimpses of what Lewis called' Joy' or Sehnsucht served as the central thread of his life" and provided him with the ontological lens through which he viewed reality. 13 It is the plan of this paper to further examine Lewis' treatment of Sehnsucht" by engaging it in dialogue with a Lutheran theological perspective. In doing so, there are two main objectives that I hope to accomplish. The first objective is to demonstrate that the experience of Sehnsucht should not be merely dismissed or fully embraced; rather, it should be placed within a proper theological narrative that …

Increasing Productivity In Irrigated Agriculture: Agronomic Constraints And Hydrological Realities, Chris Perry, Pasquale Steduto, Richard C. Allen, Charles M. Burt Nov 2009

Increasing Productivity In Irrigated Agriculture: Agronomic Constraints And Hydrological Realities, Chris Perry, Pasquale Steduto, Richard C. Allen, Charles M. Burt

BioResource and Agricultural Engineering

Irrigation is widely criticised as a profligate and wasteful user of water, especially in watershort areas. Improvements to irrigation management are proposed as a way of increasing agricultural production and reducing the demand for water. The terminology for this debate is often flawed, failing to clarify the actual disposition of water used in irrigation into evaporation, transpiration, and return flows that may, depending on local conditions, be recoverable. Once the various flows are properly identified, the existing literature suggests that the scope for saving consumptive use of water through advanced irrigation technologies is often limited. Further, the interactions between evaporation …

Effects Of Stereotyping On Mood, Ovidiu Dobria Nov 2009

Effects Of Stereotyping On Mood, Ovidiu Dobria

College of Liberal Arts & Social Sciences Theses and Dissertations

The study investigated the influence of stereotyping on mood. It was hypothesized that people who were able to solve a problem by applying a stereotype will experience more positive affect compared to people who could not, and that the relationship between stereotyping and mood was mediated by effort and by certainty. 70 undergraduates were asked to predict the academic trajectory of a college applicant based on information provided by a typed college application form and by an essay. For half the participants the applicant was identified as Asian; for the others, the applicant was Anglo. The essay provided information that …

Rna-Guided Rna Cleavage By A Crispr Rna-Cas Protein Complex, Brenton R. Graveley, Sara Olson, Michael O. Duff

UCHC Articles - Research

Compelling evidence indicates that the CRISPR-Cas system protects prokaryotes from viruses and other potential genome invaders. This adaptive prokaryotic immune system arises from the clustered regularly interspaced short palindromic repeats (CRISPRs) found in prokaryotic genomes, which harbor short invader-derived sequences, and the CRISPR-associated (Cas) protein-coding genes. Here we have identified a CRISPR-Cas effector complex that is comprised of small invader-targeting RNAs from the CRISPR loci (termed prokaryotic silencing (psi)RNAs) and the RAMP module (or Cmr) Cas proteins. The psiRNA-Cmr protein complexes cleave complementary target RNAs at a fixed distance from the 3' end of the integral psiRNAs. In Pyrococcus furiosus …

Control Of Electrochemical And Ferryloxy Formation Kinetics Of Cyt P450s In Polyion Films By Heme Iron Spin State And Secondary Structure, Sadagopan Krishnan, Amila Abeykoon, John B. Schenkman, James F. Rusling Nov 2009

Control Of Electrochemical And Ferryloxy Formation Kinetics Of Cyt P450s In Polyion Films By Heme Iron Spin State And Secondary Structure, Sadagopan Krishnan, Amila Abeykoon, John B. Schenkman, James F. Rusling

UCHC Articles - Research

Voltammetry of cytochrome P450 (cyt P450) enzymes in ultrathin films with polyions was related for the first time to electronic and secondary structure. Heterogeneous electron transfer (hET) rate constants for reduction of the cyt P450s depended on heme iron spin state, with low spin cyt P450cam giving a value 40-fold larger than high spin human cyt P450 1A2, with mixed spin human P450 cyt 2E1 at an intermediate value. Asymmetric reduction–oxidation peak separations with increasing scan rates were explained by simulations featuring faster oxidation than reduction. Results are consistent with a square scheme in which oxidized and reduced forms of …
