Genetic And Non-Genetic Factors That Cause Inter-Individual Variations In Cyp2a13 Gene Expression, Hong Wu

The overall goal of this dissertation is to investigate the mechanisms underlying the large interindividual variations in susceptibility to lung carcinogenesis. We focused on CYP2A13, a respiratory tract-specifically expressed enzyme, which is the most efficient P450 towards bioactivation of a tobacco-related carcinogen 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK). The expression level of CYP2A13 varies substantially among human lung samples. Our central hypothesis is that CYP2A13 expression level is affected by both genetic regulators and non-genetic factors. Research on gender and politics often invokes Kanter's (1977) critical mass theory to draw a linkage between women political leaders' descriptive representation and substantive representation. Using the 50 state legislatures as the unit of analysis, I empirically tested the validity of the critical mass theory by investigating the relationship between women's share of legislative seats within lower chambers of state houses and their impact on legislative agenda setting and legislative success in 1995 and 2005. This work is dedicated to the investigation of protein folding with the emphasis on the aggregation and amyloid fibrils formation. The focus of the research is to elucidate the relationship between polypeptide chain sequence and folding/aggregation properties. For this purpose, i) libraries of the de novo macromolecular repetitive β-sheet forming polypeptides were designed, and ii) the strategy for their rapid creation was developed. The strategy enables retention of a basic polypeptide core, yet allowing variations in the repetitive sequence, degree of polymerization and distribution of specific amino acids. Using systematic evolution of ligands by exponential enrichment (SELEX), our lab previously selected a class of competitive RNA aptamers against the alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid (AMPA) receptors. One aptamer in this class was found to have a minimal, functional sequence of 58 nucleotides. In vitro transcription generates two RNA transcripts with the same sequence, which we named M1 and M2, both of which are required to work together to inhibit the AMPA receptors. Although the M1 and M2 species have the same sequence, they cannot be interconverted through unfolding by denaturation/refolding by renaturation. The Influence Of Perceived Origins Of Learning Difficulties And Professional Role On The Acceptability Of The Iq-Achievement Discrepancy And Response-To-Intervention (Rti) Approaches To Learning Disability Classification, Matthew Amore Raso

The present study assessed the acceptability of both the IQ-achievement discrepancy approach and the Response-to-Intervention (RTI) approach to LD classification among teachers, school psychologists, and school administrators. Relationships between each of these variables and perceived origins of learning difficulties among these professionals were also assessed. RTI received more favorable acceptability ratings when compared with the IQ-achievement approach, and more weight was attributed to instructional influences than either biological or environmental influences across all three groups, although significant between-group differences were noted. Biologically and instructionally-based explanations of learning difficulties were significant predictors of IQ-achievement acceptability, but in opposite directions. In contrast, …

This study tests the hypothesis that long–term experience of Apostolic Pentecostal glossolalia or “speaking in tongues” reduces the reactivity of biological stress response to normal or "daily" stressors. Glossolalia is a form of religious dissociation. Dissociation is a universal capacity often conflated with “trance.” It refers to the partitioning of awareness associated with a variety of cross–cultural forms, from daydreaming and denial to possession trance, shamanic spirit journeys, and dissociative identity disorder. Dissociation is believed to reduce or filter stress by mediating evaluation of potential stressors and reactivity of the mechanisms of biological stress response. Plants growing in metalliferous soils may restrict metal uptake and transport depending on metal concentration, sediment characteristics, and plant species. As native plants are replaced by invasives, different patterns of metal cycling can occur, making continued study of this process important. Sediments and tissues of four aquatic plant species/genera: Phragmites australis (common reed); Iris versicolor (blueflag iris); Typha latifolia (broadleaf cattail); and genera Cyperus sp. (sedge) from three urban and two rural sites in Albany County, NY were analyzed for total mercury (Hg­T) by cold vapor atomic absorption spectroscopy. Vegetated, or “green” roofs are used in urban architecture and sustainable building to capture stormwater and enhance urban wildlife. Most research to date has focused on their water retention capabilities, but green roofs also have the potential to treat anthropogenic acid precipitation in urban environments. More specifically, green roofs could reduce sulfate, nitrate and hydrogen ion concentrations in runoff. One previous study has demonstrated that nitrate was reduced in water draining from a green roof (Berndtsson, et al. 2006), but this unpublished thesis appears to be the first comprehensive study of the effects of vegetated roof materials on major ions …

Ammonia is the primary base in the atmosphere. It reacts readily with atmospheric acids such as sulfuric acid, nitric acid, and hydrochloric acid to form ammonium salts that occur predominantly in the fine aerosol particle (PM2.5) fraction. The ability to measure ammonia species (NHx, the sum of gaseous NH3 and particulate NH4+) is essential for the investigation of atmospheric behaviors of NHx. The nucleocapsid (NC) protein NCp7 of the human immunodeficiency virus type 1(HIV 1) is a small basic protein with two zinc finger motifs. NCp7 has key roles in virus replication and structure, which rely on its interactions with nucleic acids. We investigated the interaction of NCp7 with Transactivation Response Element (TAR) DNA which is critical for minus strand transfer during the HIV 1 reverse transcription. The EPR spin probe nitroxide was used as the reporter. Molecular Evolution Of Genes Underlying Phenotypic Differences Between Humans And Chimpanzees, Santhoshi Bandla

Chimpanzees (Pan troglodytes) and bonobos (P. paniscus) are often used as models to study the genetic and morphological changes on the lineage leading to the modern humans (Homo sapiens). Results of this dissertation suggest that, in comparison to other hominoids, chimpanzees and bonobos are more derived in their relative testes sizes and promiscuous mating systems. Phylogenetic analysis of genes that might underlie increased testes size revealed that, in addition to being a sex-determining gene, SRY displays Pan-specific amino acid replacements that make it a compelling candidate as a testes-size determining gene. The placenta forms in eutherian mammals, and is responsible for the nutrition of the developing fetus. However, maternal exposure to environmental pollutants both before and during pregnancy may result in the passage of toxins through the placental barrier and into fetal tissues. The placenta is the only organ derived from both maternal and fetal tissues, and establishes a link between the fetus and the environmental exposures of the mother. The analysis of placentae for the presence of environmental pollutants offers the possibility of exposure measurements in both the mother and the developing fetus. A key assumption of the traditional trait-based approach to the study of crime is that personality traits cause people to act similarly across a wide array of contexts. This approach has been challenged for its failure to acknowledge differences in the social environments to which individuals are exposed. Similarly, community-level explanations of crime have been criticized for failing to acknowledge that there are important individual differences between criminals and non-criminals. This study sought to expand upon existing literature pertaining to feelings toward oneself and aggressive behavior. Specifically, global self-esteem, as well as two specific subsets of self-esteem, defensive self-esteem and narcissistic self-esteem, were examined as predictors of aggressive behavior. Additionally, the relationship between aggression and self-compassion, a recently introduced self-construct moderately correlated with self-esteem, was investigated. College students from a large Northeastern University were invited via email or through a brief classroom presentation to participate in this online study. The advent of Nano-biotechnology has opened a whole new area of biological study that has allowed us to pattern, manipulate, separate, evaluate and detect biological structures and functions that were previously not possible. This dissertation will describe different types of micro-fabricated devices that were designed to quantify and/or evaluate the functional activities of cells of the immune system. The micro-fabricated devices were developed in collaboration with our colleagues at Princeton University as part of our Cellular Micro-dynamics Program of the Nanobiotechnology Center (NBTC) at Cornell University.

Obesity is a major health crisis in the United States, with approximately two-thirds of the population qualifying as overweight and, of those, almost one half qualifying as obese (CDC, 2007). While there are a myriad of causes of obesity, a great deal of evidence shows that many individuals, and particularly women, eat as a coping response to stress (e.g., Greeno & Wing, 1994). Single-particle reconstruction from cryo-electron microscopy (cryo-EM) images is a widely used technique in studying the structure of macromolecular complexes. This technique allows the visualization of molecules in their native hydrated state. The heterogeneity of two-dimensional projection images, resulting from the co-existence of different states of a macromolecular complex, remains a major obstacle, as it impairs the validity of reconstruction and limits the progress toward higher resolution. On the other hand, co-existing structures, if successfully extracted from a single dataset, can provide valuable information about the equilibrium and dynamics of the biological system being studied.

The side chain of a compound plays an important role in its biological function. In our studies, we have found that hydroximinosteroid derivatives with different side chains and position of hydroximino on ring A and B displayed remarkable distinct cytotoxicities against a diversity of cancer cell types. Presence of an oxime group on ring B and a hydroxy on ring A or B resulted in a higher cytotoxicity than other structural motifs. In addition, a cholesterol-type side chain at position 17 was required for the biological activity. Our findings provide new evidence showing the relationship between the chemical structure and …
