Learning and other forms of plasticity result from changes in transmission at existing synapses or the construction or elimination of synapses. Synapses occur at the juxtaposition of boutons, with their postsynaptic partners, dendrites and cell bodies. It has been assumed that connections in the primary visual cortex (V1) become static after the critical period. Recent studies show, however, that dendritic spines appear and disappear during adulthood in the normal brain. Our first objective was then to determine whether axonal branches and boutons also undergo morphological changes. To do so, we performed longitudinal studies of virally labeled neurons and their processes. …

Structural Studies Of Three Factors That Affect The Prokaryotic Transcription Cycle; Microcin J25, Lambda Q And T4 Gp33, Kelly-Anne Fisi Twist

This thesis examines three factors that affect the prokaryotic transcription cycle; Microcin J25, a peptidyl inhibitor of RNAP; Q antiterminator protein, a bacteriophage protein that modifies RNAP to readthrough termination signals in phage late genes; and Gp33, a T4 phage protein that is a co-activator of T4 phage late gene transcription. We determined that the Microcin J25 peptide (MccJ25) has a lassoed-tail structure using mass spectrometry and NMR. The 21 amino acid MccJ25 peptide is produced from a 58 amino acid pro-peptide, McjA, that is truncated and post-translationally modified by two maturase enzymes. The Epstein-Barr virus (EBV) is a lymphotropic γ-herpes virus infecting over 90% of the human adult population. A striking feature that the virus shares with other γ-herpes viruses is its oncogenic potential. This transforming property can be observed as B cell transformation in vitro and lymphomas as well as epithelial cancers in vivo, but most immunocompetent individuals control EBV infection successfully without the occurrence of disease. Cells of the innate immune system act in synergy to provide a first line of defense against pathogens. The spectacular cellular rearrangements of keratinocytes as epidermal appendages form and the epidermis turns over rely on the rapid and precise modulation of intercellular adhesion. While genetic studies in vertebrates have revealed a pivotal role for classical cadherin-mediated cell adhesion in the regulation of coordinated cell movements in the embryo, little is know about the contribution of classical cadherins to epidermal morphogenesis. Genetic dissection of cadherin function in skin is complicated by overlapping expression of E- and P-cadherin and the fact that their genes are tightly linked, precluding conventional conditional knockout strategies. The Effects Of Dietary Conjugated Linoleic Acid (Cla) On Fatty Acid Composition And Key Enzymes Of Fatty Acid Oxidation In Liver And Muscle Of Geese, Xuhui Zhang, Baowei Wang, Fangyu Long, Lei Wang, Zhigang Yang

The objective of this study was to determine the influence of conjugated linoleic acid (CLA) on fatty acid composition and key enzymes of fatty acid oxidation in liver and muscle of geese. A total of 192 1-day-old geese were randomly assigned to 1 of 4 dietary groups, and were fed for 56 days on diets containing 2.5% soybean oil (group A, control group) or 2.0% soybean oil with 0.5% CLA (group B), 1.0% soybean oil with 1.5% CLA (group C), and 2.5 % CLA (group D). Geese fed CLA had a decreased abdominal fat percentage (AFP) (P < 0.01) compared with the control. It was observed that dietary CLA levels had a significant effect on the body weight, weight gain, feed conversion rate (FCR) (P < 0.05), and feed intake (P < 0.01). Fatty acid composition showed a significant increase of the biologically active cis-9, trans-11 and trans-10, and cis-12 CLA isomers in both liver and muscle of geese fed CLA (P < 0.01). Dietary CLA led to an increase (P < 0.01) in saturated fatty acid (SFA) and a reduction in monounsaturated fatty acid (MUFA) (P < 0.05) concentrations in both liver and muscle. In liver, dietary CLA increased (P < 0.05) acyl CoA oxidase (ACO) activity, but did not affect carnitine palmitoyltransferase-I (CPT-I) activity compared with the control. CPT-I activity was significantly increased by 2.5% dietary CLA in muscle, where ACO activity was decreased at 1.5% CLA level. The results obtained suggested that geese can successfully incorporate CLA in both liver and muscle, which could be beneficial in the human diet, through provision of bioactive fatty acids with no detrimental effects on n-3 PUFA levels.

In many rural communities there exists a stereotype about clergy education perpetuated since slavery. This project offered bi-vocational clergy opportunities to be equipped, empowered and certified for servant leadership. Data captured from 2006 through 2008 was taken from the 25 ordinands. Questions that guided this study were grouped into two sets: examination by quantitative measurements through the passing of a catechism; and by qualitative measurements from personal and telephone interviews. The results of the research revealed a discrepancy in the ordination process where prerequisites and retention studies must play a greater role for future ordinands.

Because current legislation, including OH H.B. 237, is insufficient in that it does not have the potential to significantly increase the number of cord blood donations, it will be necessary to enact legislation that is more demanding. Such legislation should be modeled after current "required request" organ donation laws, which mandate that health professionals actively pursue organ donations by expressly asking the family to consent to donation. Presented herein is an analysis of the equity of epilepsy-related driving restrictions and the role that the state of Ohio may assume in the restructuring of such laws. Part two of this paper discusses the medical aspects of seizures and epilepsy, including basic etiology, treatments, and prognoses. Part three of this paper examines the different types of disabilities and the stigma that impacts individuals with epilepsy. Comparative Research in Law & Political Economy

In many high-income countries, like Canada and the United Kingdom, there has recently been a significant increase in the number of migrant workers entering and participating in their labour markets. This article is concerned with the implications of this phenomenon for protective labour laws and, in particular, for occupational health and safety regulation. We identify a framework for assessing the OHS vulnerabilities of migrant workers, using a layered approach which assists in identifying the risk factors. Using this layer of vulnerability framework, we compare the situation of at-risk migrant workers in Canada and the United Kingdom.

The headgroup of these glycolipids, which consist of a dissacharide i.e. maltose and lactose, provide good hydrophilicity. These glycolipids are comparable to alkyl polyglucosides (APGs), a common non-ionic surfactant used for detergents, particularly in the cosmeceutical industry. APGs are limited to high lipophilicity due to their low degree of polymerisation (~1.5) and their chemical diverse compositions make them unsuitable for bio-related applications. Technical grade glycolipids based on alcohol mixtures from reduced palm oil and palm kernel oil were investigated to explore their application as surfactants. Curriculum integration has a long history. In this paper I catalog several arguments for resistance against integration and present the historical roots of support for those arguments offered by critics of curriculum integration. First, I review some linguistic and usage limitations of the term. Second, I examine several practical and institutional difficulties related to implementing integrated curriculum. Third, I explore some interconnected psychological and sociological dimensions of resistance to curriculum integration. Finally, I consider several epistemological dimensions of resistance to integration, some of which underlie the sociological and psychological aspects. Abstract A key signaling pathway involved in regulating cell growth and proliferation throughout the body is the ERK signaling pathway. ERK is activated via numerous pathways including intracellular calcium release downstream of G-Protein Coupled Receptors (GPCRs). Carbachol, a GPCR-agonist, both increases intracellular calcium and ERK activation in MCF-7 cells. ERK activation and control of cell growth may act through the transcription factor Elk-1. Our goal was to elucidate the specific proteins and kinases upstream of ERK in MCF-7 cells treated with carbachol. Abstract AKT has been implicated in promoting cell survival within certain cells. Current research has shown that hormones that increase the concentration of intracellular calcium can activate AKT that in turn leads to cancer cell survival. Interestingly, LNCaP cells express the M3-subtype of GPCR's that may couple to increases in intracellular calcium and activation of the Ca2+/Calmodulin-dependent Protein Kinases (CaM Ks). Specifically CaM Kinase Kinase (CaM KK) phosphorylates its direct substrates CaM Kinase I, CaM Kinase IV, and AKT. Comparative vertebrate studies into physiological responses to adverse conditions can yield great insight into the design of robust systems and provide fundamental knowledge of stress adaptations. The classically studied cellular stress response is the mitogen-activated protein kinase (MAPK) pathway. Prior investigations in our laboratory indicated a differential response of MAPK family activation to progressive dehydration in cardiac tissue from two species of anuran amphibians: Rana catesbeiana and Bufo marinus. Western blot analysis indicated that pERK increased by three-fold in Bufo and 2-fold in Rana. pJNK levels remained unchanged in Bufo whereas in Rana increased by 50%. pp38 did not exhibit …

The separation between behavioral services and traditional medicine is increasingly being seen as counterproductive on personal and societal levels. Despite this, there has been little research examining how integrated models blending mental and physical health services could be implemented. The literature revealed that behavioral interventions have been incorporated into traditional medical treatments, but this often has been piecemeal in nature and has yielded equivocal results. Intimate partner violence (IPV) is a multifaceted social issue that affects the Christian faith community as it does the secular community. Though the literature reflects some understanding of general correlates and possible antecedents to IPV within the Christian community, the impact of religious and spiritual factors tends to be homogenized and is often misjudged. Allport's theory of intrinsic and extrinsic religious orientation provided a platform for investigating Christian male-perpetrated IPV. Researchers have theorized that student achievement and its contingent effects on self-efficacy are important factors in art education. There is, however, a paucity of research addressing this relationship, which in turn affects students' and educators' levels of success. Accordingly, this study was an investigation of the relationship between art education and self-efficacy in middle school students and tested the constructivist theory, as embodied in Bandera's theories on the foundations of self-efficacy beliefs. Studies reveal a progressive net decrease in the nursing profession across four generations, creating a shortage that poses a critical threat to the health care delivery system and to the health and safety of patients within that system. Research also suggests that generational cohorts reflect social change in attitudes toward work-life balance, organizational commitment, personal-professional relationships, autonomy, focus on career advancement, and actions that represent organizational citizenship behavior (OCB). The current dissertation sought to examine whether there was an association between peer victimization, neuroendocrine functioning, and physical health outcomes. Adolescents and their parent (N = 107) participated in a two-part study. In the first phase, adolescents completed a series of questionnaires either at school or online to assess levels of peer victimization, social support, and health outcomes. They then came into the laboratory with a parent for the second part of the study which consisted of two sessions. The purpose of this research was to examine factors that influence the success of community based marine protected areas in North Sulawesi Indonesia. The research took place in North Sulawesi Province of Indonesia. A multi-methods approach was used including administration of a survey questionnaire to a random sample of household respondents, use of secondary data, key informant interviews and direct observations within the field sites. Results demonstrate that community-based management interventions and community responses are complex and varied. The microbial loop stresses the importance of flagellated protozoa as predators of bacteria and regenerators of nutrients, the outcome of which affects higher trophic levels. The element stoichiometry of flagellates has only been examined while feeding upon bacteria or phytoplankton. In this study, the element stoichiometry of the protozoan flagellate, Ochromonas danica, was determined while growing osmotrophically thus eliminating the potential errors due to element contributions of prey in food vacuoles. In a series of experiments taking place in recirculating artificial stream channels, periphyton communities growing on ceramic tiles were subjected to variations in current velocity and nutrient supply. The first experiment used stream water and manipulated current velocity, subjecting periphyton communities to either high or low current velocities (30 and 10 cm * sec-1 respectively), while in experiments 2-4, modified WC media was used and periphyton communities subjected to different nutrient (low and high) and current regimes (low, high, and variable flow).
