Evaluation Of Pilo Substrate Specificity Using Normally Non-Glycosylated Proteins In Pseudomonas Aeruginosa, Matthew A. Henkel

P. aeruginosa 1244 (PA1244) possesses an O-linked glycosylation system by which the glycosyltransferase, PilO, transfers a single preassembled O-antigen repeating unit to the C-terminal serine residue of pilin as a posttranslational modification. Previous work has shown that the group II pilin of PA103 can be engineered for glycosylation by extending a C-terminal serine away from the surface with a short stretch of alanine residues (Horzempa et al., 2006a). As an extension of that study, several normally non-glycosylated proteins [PA683 pilin, PA1244 PilA-E. coli (EC) alkaline phosphatase (PhoA) fusion, and EC PhoA] were engineered with C-terminal amino acid extensions and assessed …

Little is known about the eastern box turtle, Terrapene c. carolina, in West Virginia. The purpose of this study was to compare demographic data between two populations of box turtles, to determine which environmental parameter(s) influence activity patterns, and to describe the optimal habitat of the box turtles at two study sites (Lake and State Park) at Beech Fork Wildlife Management Area. I observed male-skewed sex ratios and low numbers of juveniles in both populations. Male turtles were larger than females in carapace length, but females were larger in carapace height. The high rate of attrition for beginning teachers has been blamed on many factors. Nationally these include low pay, urban versus rural school settings, age, with younger teachers leaving sooner, and other variables making a complex problem with no clear component responsible for the loss of teachers. This study examined the self reported variables that contribute to high school teachers in West Virginia leaving their profession. An electronic survey instrument was distributed to current secondary teachers across the state. This Environmental Assessment (EA) has been prepared to disclose and analyze the environmental consequences of gathering excess wild horses in the Red Desert Wild Horse Herd Management Area (HMA) Complex. A new method of asymmetric phase-transfer-catalyzed alkylation is described. The synthetic utility of this method is demonstrated with the total syntheses of kurasoin A, kurasoin B, and S-naproxenTM, with current work toward S-equol. This methodology is currently being applied to an improved asymmetric total synthesis of the natural compound 12-S-HETE.

The purpose of this study was to examine the effects of professional development (PD) on the infusion of engineering design into high school curricula. Four inservice teachers with backgrounds in physics, chemistry, industrial education, math, and electrical engineering participated in the 2006 National Center of Engineering and Technology Education (NCETE)-sponsored PD workshops at California State University, Los Angeles (CSULA) and provided three sources of data that were used to answer the research question, “What are the effects of PD on infusing engineering design into high school science, technology, engineering, and math (STEM) curricula”? Nitric Oxide releasing biopolymers have the potential to prolong vascular graft and stent potency without adverse systemic vasodilation. It was reported in literature that eNOS-overexpressing endothelial cell seeding of synthetic small diameter vascular grafts decreased human platelet aggregation by 46 and bovine aortic smooth muscle cell proliferation by 67.2 in vitro. We hypothesized that incorporating the enzyme nitric oxide synthase (NOS) in biocompatible polymeric matrix will provide a source of NO that utilizes endogenous compounds to maintain an unlimited supply of NO. Activation of heterodimeric (alpha/beta) integrin transmembrane receptors by the cytoskeletal protein talin is essential for many important cell adhesive responses including cell-extracellular matrix contact, cell motility and survival. A key step in this process involves interaction of phosphotyrosine-binding (PTB) domain in the N-terminal head of talin (talin-H) with integrin beta3membrane-proximal cytoplasmic tails (beta-MP-CTs). Compared to talin-H, intact talin exhibits low potency in inducing integrin activation. Using TROSY NMR spectroscopy, we found that the large C-terminal rod domain of talin (talin-R) interacts with talin-H and allosterically restrains talin in a closed conformation. Cytokines are intracellular messengers that activate multiple signaling pathways and regulate cell survival, differentiation, migration, apoptosis and immune responses. We are investigating the homeostatic control of cytokine-mediated cell signaling using interleukin-4 (IL-4) as a model cytokine. Protein tyrosine phosphorylation is essential for cytokine-dependent signal transduction through the Janus Kinase-Signal Transducer and Activator of Transcription (Jak-STAT) pathway. Therefore, protein tyrosine dephosphorylation is a key mechanism providing the homeostatic control of the pathway. Intravascular ultrasound (IVUS) is increasingly employed for detection and evaluation of coronary artery diseases. Tissue Harmonic Imaging provides different tissue information that could additionally be used to improve diagnostic accuracy. However, current IVUS systems, with their unfocussed transducers, may not be capable of operating in harmonic imaging mode. Thus, there is a need to develop suitable transducers and appropriate techniques to allow imaging in multi modes for complementary diagnostic information. We have previously reported that the level of MyoD expression correlates with the level of apoptosis that occurs in a subpopulation of skeletal myoblasts induced to differentiate by serum withdrawal. Herein we document that MyoD expression dictates the apoptotic threshold in myoblasts and fibroblasts in response to a variety of apoptotic stimuli. Specifically, re-expression of MyoD in skeletal myoblasts rendered defective for both differentiation and apoptosis by the expression of oncogenic Ras restores their ability to undergo both differentiation and apoptosis in response to serum withdrawal. This study investigated the statistical significance of student trust next to the well-tested constructs of personality and motivation to determine whether trust is a significant predictor of course achievement in college math courses. Participants were 175 students who were taking undergraduate math courses in an urban public university. Novel tyramine-based hyaluronan (HA) and collagen hydrogels have been developed in which cross-linking is accomplished via peroxidase-mediated dityramine linkages allowing direct cross-linking in vivo. These TB hydrogels possess advantageous physical properties, which include excellent biocompatibility and the ability to mimic the biological, structural and mechanical properties of normal, healthy tissues, including cartilage, and thus provide for synthetic, implantable biomaterials suitable for a wide range of tissue types. The efficacy of these TB-hydrogels has been previously tested in a number of clinically relevant animal models, which have evaluated their applicability for the repair/replacement of various tissues, including cartilage. Chemicals and drugs are known to be metabolized mostly by Cytochrome P450 xenobiotic metabolizing enzymes. However, the detailed mechanism of nitro-compounds metabolism is still unclear. The activation of nitro-xenobiotics by heme-P450 enzymes is a potential explanation for the origin of nitro-compound carcinogenesis. Investigating the interaction of simple nitro-compounds with redox activity of heme enzymes is therefore critical to explore the mechanism and products of activation. Natural Orifice Translumenal Endoscopic Surgery (NOTES) avoids skin incisions by accessing the abdominal cavity through natural orifices. Benefits include less pain, fewer complications and no scars. The aims of this study were to evaluate safety and efficacy of access techniques for natural orifice surgery, to investigate safety and efficacy of closure methodologies and to compare the inflammatory response induced by NOTES with that following conventional surgery. Access techniques were evaluated for safety and efficacy by measuring resultant injury and time required to access the peritoneal cavity in an acute porcine model.
