Multiple Splice Defects In Abca1 Cause Low Hdl-C In A Family With Hypoalphalipoproteinemia And Premature Coronary Disease, Jeffrey Rhyne, Myrna M. Mantaring, David F. Gardner, Michael Miller Jan 2009

Multiple Splice Defects In Abca1 Cause Low Hdl-C In A Family With Hypoalphalipoproteinemia And Premature Coronary Disease, Jeffrey Rhyne, Myrna M. Mantaring, David F. Gardner, Michael Miller

Internal Medicine Publications

Background Mutations at splice junctions causing exon skipping are uncommon compared to exonic mutations, and two intronic mutations causing an aberrant phenotype have rarely been reported. Despite the high number of functional ABCA1 mutations reported to date, splice variants have been reported infrequently. We screened DNA from a 41 year-old male with low HDL-C (12 mg/dL [0.31 mmol/L]) and a family history of premature coronary heart disease (CHD) using polymerase chain reaction single-strand conformation polymorphism (SSCP) analysis.

Methods Family members with low levels of HDL-C (n = 6) were screened by SSCP for mutations in ABCA1. Samples with altered SSCP …

Estrogen Exposure, Obesity And Thyroid Disease In Women With Severe Pulmonary Hypertension, Lori Sweeney, Norbert F. Voelkel Jan 2009

Estrogen Exposure, Obesity And Thyroid Disease In Women With Severe Pulmonary Hypertension, Lori Sweeney, Norbert F. Voelkel

Internal Medicine Publications

Severe pulmonary hypertension is a lethal group of disorders which preferentially afflicts women. It appears that in recent years the patient profile has shifted towards older, obese, and postmenopausal women, suggesting that endocrine factors may be important. Several studies have revealed an increased prevalence of thyroid disease in these patients, but no studies have evaluated for a coexistence of endocrine factors. In particular, no studies have attempted to evaluate for concurrent thyroid disease, obesity and long-term estrogen exposure in patients.

88 patients attending the Pulmonary Hypertension Association 8th International meeting completed a questionnaire and were interviewed. Information was collected regarding …

The Biological Function Of Interacting Partners Of Zxd Family Proteins, Rupa V. Koneni Jan 2009

The Biological Function Of Interacting Partners Of Zxd Family Proteins, Rupa V. Koneni

ETD Archive

We have identified several protein-protein interactions amongst the members of the zinc finger, X-linked (ZXD) family of proteins (ZXDA, ZXDB, ZXDC and ZXDC2) that have important roles in the regulation of major histocompatibility complex class II and several myeloid-specific genes. The ZXDC and ZXDA proteins share significant nucleotide sequence homology and each contain ten C2H2 zinc fingers and a transcription activation domain. In addition, ZXDC has a C-terminal region that is necessary to interact with a key cofactor, CIITA (Class II transactivator) and activate Major histocompatibility complex class II and class I (MHC II and MHC I respectively) genes. In …

Structural Investigation Of The Complex Of Filamin A Repeat 21 With Integrin Αiib And Β3 Cytoplasmic Tails – A Potential “Transmission” To Regulate Cell Migration, Jianmin Liu Jan 2009

Structural Investigation Of The Complex Of Filamin A Repeat 21 With Integrin Αiib And Β3 Cytoplasmic Tails – A Potential “Transmission” To Regulate Cell Migration, Jianmin Liu

ETD Archive

Cell functions in multi-cellular organisms are strongly depend on the dynamic cooperation between cell adhesion and cytoskeleton reorganization. Integrins, the major cell adhesion receptors, bind to extracellular matrix (ECM) and soluble ligands on the cell surface and link to the actin cytoskeleton inside the cell membrane. In this manner, integrins integrate cell adhesion and cytoskeleton reorganization by acting as a mechanical force transducer and a biochemical signaling hub (Zamir and Geiger 2001). Consequently, integrins are vital for development, immune responses, leukocyte traffic and hemostasis, and a variety of other cellular and physiological processes. Integrins are also are the focal point …

Comparative Study Of Cyanobacteria Of Desert And Semi-Desert Crusts Of Two Different Continents: Africa (Ethiopa) And North America (Usa), Melaku Mesfin

ETD Archive

Cyanobacterial flora of microbiotic crusts of the Ethiopian Rift Valley System and the Great Basin in Idaho and Oregon were studied. Nine species of cyanobacteria were identified and morphologically characterized from the microbiotic crusts of the Rift Valley in Ethiopia. These included one species of Chroococcales, five species of Oscillatoriales, and three species of Nostocales. Similar studies were made for the microbiotic crusts of the Great Basin of Idaho and Oregon. A total of six morphospecies were morphologically characterized and identified, three species of Oscillatoriales, and three in Nostocales, were recorded in the regions. The cyanobacterial flora of the two …

Hemeproteins Bathed In Ionic Liquids;Examining The Role Of Water And Protons In Redox Behavior And Catalytic Function, John Joseph Moran Jan 2009

Hemeproteins Bathed In Ionic Liquids;Examining The Role Of Water And Protons In Redox Behavior And Catalytic Function, John Joseph Moran

ETD Archive

We investigate the changing behaviors of myoglobin and nitric oxide synthase (NOS) in the near-absence of bulk water and/or protons by using ionic liquid butyl methyl imidazolium tetrafluoroborate as a non-aqueous milieu. Through direct charge transfer and metalloprotein-mediated catalytic reduction of oxygen and nitric oxide, we shed light on diverging aspects on how the two hemeproteins face the scarcity of water and/or protons in bulk. Isotopic effect investigations using D2O further elucidates kinetic aspects of proton transfer. Finally, in the case of NOS oxygenase, pterin cofactor binding and NOS-mediated catalytic oxidation of L-arginine in ionic liquids interrogates proton and water …

Fabricating New Miniaturized Biosensors For The Detection Of Dna Damage And Dna Mismatches, N. Indika Perera Jan 2009

Fabricating New Miniaturized Biosensors For The Detection Of Dna Damage And Dna Mismatches, N. Indika Perera

ETD Archive

A large number of genetic diseases and genetic disorders are simply caused by base alterations in the genome. Therefore, developing efficient and cost effective techniques for routine detection of these alterations is of great importance. Different methods involving gel electrophoresis and Polymerase Chain Reaction have been widely employed, but majority of these methods are costly, time consuming, and lack throughput, creating a fundamental gap between the current state-of-the-art and desired characteristics of low-cost, high-speed, simplicity, versatility, and potential for miniaturization. In this study, we attempt to bridge this gap by developing new sensing platforms to detect DNA base mismatches and …

Nitric Oxide Synthase In Confined Environments;Detection And Quantification Of Nitric Oxide Released From Cells And Modified Liposomes Using A Sensitive Metal Catalyst-Pedot Modified Carbon Fiber Electrode, P. A. Reshani H. Perera Jan 2009

Nitric Oxide Synthase In Confined Environments;Detection And Quantification Of Nitric Oxide Released From Cells And Modified Liposomes Using A Sensitive Metal Catalyst-Pedot Modified Carbon Fiber Electrode, P. A. Reshani H. Perera

ETD Archive

Nitric oxide (NO) is a freely diffusible, gaseous free radical, associated with many physiological and pathological processes that include neuronal signaling, immune response, and inflammatory response. NO is produced from L-arginine in an NADPH-dependent reaction catalyzed by a family of nitric oxide synthase (NOS) enzymes. A deficiency in NO plays a role in hypertension, hyperglycaemia, and arteriosclerosis, among other pathological states. Conversely, increased NO levels contribute to arthritis, septic shock, and hypotention. Therefore, measuring and quantifying NO production in biological systems and matrices may be vital in elucidating physiological and pathological processes. The goal of this work is to develop …

Factors Affecting Student Motivation Related To Enrollment And Retention In Music And Performing Ensembles Outside Of The School Environment, Craig T. Klonowski Jan 2009

Factors Affecting Student Motivation Related To Enrollment And Retention In Music And Performing Ensembles Outside Of The School Environment, Craig T. Klonowski

ETD Archive

Despite extensive research regarding student motivation in performing ensembles, little is known about student motivation in ensembles outside of the school environment. The literature regarding motivation in school ensembles shows motivation comes from five main categories: parents and family, peers, directors, environment, and the students themselves. This study asks basic questions regarding motivation of students in a large community performing ensemble, and compares them to responses from the same students regarding their school ensemble experiences. The responses to the questionnaire suggest that motivating factors in community ensembles fall into the same basic categories, but have slightly different nuances based on …

Evolutionary Psychology, Social Emotions And Social Networking Sites: An Integrative Model, Sandra L. Suran Jan 2009

Evolutionary Psychology, Social Emotions And Social Networking Sites: An Integrative Model, Sandra L. Suran

ETD Archive

Humans engage their environment through the combined effort of the mind, body proper and corresponding instinctual emotive devices (Damasio, 1994). These structures are a part of the same phenomena: human biology. Our emotive devices along with the brain and body act as an interwoven organism appraising the environment and making necessary adjustments for it survival and efficient functioning. Moreover, these structures actively engage socially complex signals like those that we experience daily in our social world. Plausibly, when these devices are not utilized the interwoven organism will be in a state of atrophy. Much like an antigen would trigger a …
