Bromeliads As A Breeding Site For The Dengue Vector Aedes Aegypti, Eva B. Shultis Apr 2009

Bromeliads As A Breeding Site For The Dengue Vector Aedes Aegypti, Eva B. Shultis

Independent Study Project (ISP) Collection

Dengue Fever is a major public health concern in tropical and subtropical climates worldwide, including the city of Cairns, Australia, which is currently suffering a severe outbreak. The most important vector of the Dengue virus is the predominantly urban mosquito Aedes aegypti (L.), which lays its eggs in both artificial and natural containers, including the ornamental bromeliad plants found in many household gardens. The ability of larvae to develop to adulthood inside bromeliads has become controversial, however, and bromeliad enthusiasts frequently refuse to have their plants treated with insecticide. The aim of this study was to determine the conditions under …

Molecular Taxonomy, Phylogeny And Biogeography Of Nematodes Belonging To The Trichinella Genus, Edoardo Pozio, Eric Hoberg, Giuseppe La Rosa, Dante S. Zarlenga

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Studying parasites of the genus Trichinella provides scientists of today many advantages. This is a group of zoonotic nematodes that circulates freely among wildlife hosts with one in particular, Trichinella spiralis that is exceptionally well adapted to domestic swine. Recent reports suggest that human infections from hunted animals are on the rise worldwide and numerous countries still experience problems with T. spiralis in their domestic food supplies. Trichinella is a genus whose members are easily propagated in the laboratories, have been used as models to investigate host-parasite relationships and parasitism among clade I organisms, and represent a poorly investigated link …

Laser Levitation Of Solid Particles For Combustion And Gasification Applications, Skigh E. Lewis Mar 2009

Laser Levitation Of Solid Particles For Combustion And Gasification Applications, Skigh E. Lewis

Theses and Dissertations

This dissertation details theoretical and experimental work in the development of a novel combustion diagnostic: laser levitation of solid particles. Theoretical analyses of the forces involved in the suspension of solid particles in a laser beam provide a comprehensive description of the levitation mechanism. Experimental work provides extensive observations and data that describe each of the forces involved, including results from detailed models. Theoretical models establish that a free-convective drag force, light scattering, photon momentum, and other minor forces contribute to the trapping mechanism. The theory quantitatively predicts particle temperature and magnitudes of each of the forces involved. Experimental measurements …

Chemistry Faculty Publications

The hexane- and ethyl acetate-soluble extracts of the leaves of Brassaiopsis glomerulata (Blume) Regel (Araliaceae), collected in Indonesia, were found to inhibit aromatase, the rate-limiting enzyme in the production of estrogens from androgens, in both enzyme- and cell-based aromatase inhibition (AI) assays. Bioassay-guided fractionation led to the isolation of six known compounds of the steroid and triterpenoid classes (1–6) from the hexane extract, of which 6β-hydroxystimasta-4-en-3-one (5), was moderately active in the cell-based AI assay. Fractionation of the ethyl acetate extract afforded seven pure isolates (7–13) of the modified peptide, fatty acid, monoterpenoid, and benzenoid types, including six known compounds …

Osmotic Characteristics And Fertility Of Murine Spermatozoa Collected In Different Solutions, James D. Benson, Si Wei, Hongsheng Men, John K. Critser

Faculty Articles, Papers, and Other Scholarship

Osmotic stress is an important factor that can result in cell damage during cryopreservation. Before ejaculation or collection for cryopreservation, murine spermatozoa are stored in epididymal fluid, a physiologically hyperosmotic environment (w415 mmol/kg). The objectives of this study were to determine the osmotic tolerance limits of sperm motion parameters of ICR and C57BL/6 mouse spermatozoa collected in isosmotic (290 mmol/kg) and hyperosmotic (415 mmol/kg) media, and the effect of the osmolality of sperm collection media on sperm fertility after cryopreservation. Our results indicate that murine spermatozoa collected in media with different osmolalities (290 and 415 mmol/kg Dulbecco’s phosphate buffered saline …

Background

Microarray analysis and 454 cDNA sequencing were used to investigate a centuries-old problem in regenerative biology: the basis of nerve-dependent limb regeneration in salamanders. Innervated (NR) and denervated (DL) forelimbs of Mexican axolotls were amputated and transcripts were sampled after 0, 5, and 14 days of regeneration.

Results

Considerable similarity was observed between NR and DL transcriptional programs at 5 and 14 days post amputation (dpa). Genes with extracellular functions that are critical to wound healing were upregulated while muscle-specific genes were downregulated. Thus, many processes that are regulated during early limb regeneration do not depend upon nerve-derived factors. …

Biology Faculty Publications

BACKGROUND: The basis of genome size variation remains an outstanding question because DNA sequence data are lacking for organisms with large genomes. Sixteen BAC clones from the Mexican axolotl (Ambystoma mexicanum: c-value = 32 x 10(9) bp) were isolated and sequenced to characterize the structure of genic regions.

RESULTS: Annotation of genes within BACs showed that axolotl introns are on average 10x longer than orthologous vertebrate introns and they are predicted to contain more functional elements, including miRNAs and snoRNAs. Loci were discovered within BACs for two novel EST transcripts that are differentially expressed during spinal cord regeneration and skin …

Background

Apoptosis has been speculated to be involved in schizophrenia. In a previously study, we reported the association of the MEGF10 gene with the disease. In this study, we followed the apoptotic engulfment pathway involving the MEGF10, GULP1, ABCA1 and ABCA7 genes and tested their association with the disease.

Methodology/Principal Findings

Ten, eleven and five SNPs were genotyped in the GULP1, ABCA1 and ABCA7 genes respectively for the ISHDSF and ICCSS samples. In all 3 genes, we observed nominally significant associations. Rs2004888 at GULP1 was significant in both ISHDSF and ICCSS samples (p = 0.0083 and …

Abundance And Distribution Of Two Species Of Squilla (Crustacea: Stomatopoda: Squillidae) In The Northern Gulf Of Mexico, Jennifer L. Wortham Jan 2009

Abundance And Distribution Of Two Species Of Squilla (Crustacea: Stomatopoda: Squillidae) In The Northern Gulf Of Mexico, Jennifer L. Wortham

Gulf and Caribbean Research

Stomatopods (mantis shrimps) are predatory benthic crustaceans. Mantis shrimp in the genus Squilla are frequent bycatch animals unintentionally collected in conjunction with the shrimp fishery in the Gulf of Mexico (GOM). Their carcasses are discarded instead of being retained for human consumption, fish meal, or other protein-based food products. The size, depth, salinity, and temperature distributions of these species, as well as their abundance based on gender, were examined to gain biological information that would be necessary if a fishery were to develop in the GOM. I collected samples (n = 2,854) of Squilla empusa and Squilla chydaea in the …

Eugenia Jambolana Lam. Berry Extract Inhibits Growth And Induces Apoptosis Of Human Breast Cancer But Not Non-Tumorigenic Breast Cells, Liya Li, Lynn S. Adams, Shiuan Chen, Caroline Killian, Aftab Ahmed, Navindra P. Seeram

Biomedical and Pharmaceutical Sciences Faculty Publications

The ripe purple berries of the native Indian plant Eugenia jambolana Lam., known as Jamun, are popularly consumed and available in the United States in Florida and Hawaii. Despite the growing body of data on the chemopreventive potential of edible berry extracts, there is paucity of such data for Jamun fruit. Therefore our laboratory initiated the current study with the following objectives: (1) to prepare a standardized Jamun fruit extract (JFE) for biological studies and (2) to investigate the antiproliferative and pro-apoptotic effects of JFE in estrogen dependent/aromatase positive (MCF-7aro), and estrogen independent (MDA-MB-231) breast cancer cells, and in a …

Modeling Thip;Ate Enzyme Chemistry With Cysteine At A High Ph, Emily Michelle Turner Jan 2009

Modeling Thip;Ate Enzyme Chemistry With Cysteine At A High Ph, Emily Michelle Turner

Mahurin Honors College Capstone Experience/Thesis Projects

Thiolate is a highly reactive species critical to the function of many enzymes. We attempted to model the chemistry of two of these enzymes with cysteine in solution at a high pH. To study cysteine proteases, we increased the pH of a cysteine and 4- nitrophenylacetate solution to create a visible color change when thiolyzed that was monitored through UV-Vis spectroscopy to determine rate constants. To study transferases, we attempted to model two reactions. The first was a methyl transfer from guanine to cysteine which mimicked O6-alkylguanine-DNA alkyltransferase. The second was activation of halogenated hydrocarbons by a mechanism similar to …

Morphological And Genetic Diversity Of The New Invasive Species Bactrocera Invadens (Diptera: Tephritidae) In Africa, Mbarak Khamis Fathiya

All PhD Dissertations

Species of the genus Bactrocera in the family Tephritidae, or 'true fruit flies', are among the most important pests of fruits and vegetables. Because of the broad larval host range and cosmopolitan distribution, pest fruit flies are highly invasive, with adults often exhibiting a strong tendency for dispersal and larvae being readily transported to new areas via fruit movement. Recently, a pest species was recorded for the first time in Kenya and has subsequently been found in countries across tropical Africa. The insect was described as Bactrocera invadens, due to its rapid invasion of the African continent. Taxonomic description of …

Insights Into The Population Structure Of Blue Whales In The Eastern North Pacific From Recent Sightings And Photographic Identification, John Calambokidis, Jay Barlow, John K. B. Ford, Todd E. Chandler, Annie B. Douglas

United States Department of Commerce: Staff Publications

Blue whales were widely distributed in the North Pacific prior to the primary period of modern commercial whaling in the early 1900s. Despite concentrations of blue whale catches off British Columbia and in the Gulf of Alaska, there had been few documented sightings in these areas since whaling for blue whales ended in 1965. In contrast, large concentrations of blue whales have been documented off California and Baja California and in the eastern tropical Pacific since the 1970s, but it was not known if these animals were part of the same population that previously ranged into Alaskan waters. We document …

Microbial Diversity And Microbial Abundance In Salt-Saturated Brines: Why Are The Waters Of Hypersaline Lakes Red?, Aharon Oren

Natural Resources and Environmental Issues

Salt-saturated lakes such as the North Arm of Great Salt Lake and saltern crystallizer ponds contain 10-10 and more red microorganisms ml-1. Even the Dead Sea occasionally turns red due to microbial blooms. Three types of organisms may contribute to the coloration: the alga Dunaliella salina rich in carotene, halophilic Archaea (family Halobacteriaceae) containing 50-carbon bacterioruberin carotenoids and sometimes also retinal proteins (bacteriorhodopsin, halorhodopsin), and the recently discovered Salinibacter (Bacteroidetes) which contains pigment salinixanthin (an unusual acylated C40-carotenoid glucoside) as well as different retinal pigments. Bacteriorhodopsin and halorhodopsin enable the cells to directly use light energy for respectively the outward …

Synthesis, Characterization And Biological Activity Of Some Carbazone Schiff Bases And Their Metal Complexes / Mohd. Rizal Bin Razali, Mohd. Rizal Razali

Student Works (2000-2009)

All the metal complexes in this study are synthesized by the general method which the ligand was first prepared by refluxing the precursors of the Schiff base. It was then followed by the addition of the appropriate metal salt to form metal complexes. The characterization of the series of the Schiff base complexes of Ni(II), Cu(II), Zn(II) and Cd(II) with ligand systems containing nitrogen, sulphur and oxygen donor atoms were also carried out. Formation of the Schiff base ligands were confirmed by the presence of v(C=N) band at the range 1550-1650 cm-1 in the IR spectra. In some cases the …

Gonadotropin Releasing Hormone In Agnathans, Scott Ira Kavanaugh Jan 2009

Gonadotropin Releasing Hormone In Agnathans, Scott Ira Kavanaugh

Doctoral Dissertations

Hagfish and lampreys are the only two representatives of Agnathans among extant vertebrates. The regulatory hypothalamic neurohormone, gonadotropin-releasing hormone (GnRH), regulates reproduction in all vertebrates through the hypothalamic-pituitary-gonadal axis. Most vertebrates have at least two forms of GnRH in the brain. In hagfish the primary amino acid structure of GnRH has not been identified as yet, however, indirect methods have shown an immunoreactive GnRH or a GnRH-like peptide in the brain of hagfish. In addition concentrations of brain immunoreactive-GnRH have been correlated with reproductive stages in the Atlantic hagfish ( Myxine glutinosa). Therefore the objective of the first study was …

Historical Changes In The Occurrence And Distribution Of Freshwater Mussels In Kansas, Robert T. Angelo, M. Steve Cringan, Eva Hays, Clint A. Goodrich, Edwin J. Miller, Mark A. Vanscoyoc, Bryan R. Simmons

Great Plains Research: A Journal of Natural and Social Sciences (through 2013)

The surface waters of eastern and central Kansas once supported an impressive variety of native freshwater mussels, but a widespread decline in species richness accompanied the urban, industrial, and agricultural development of this region. Statewide mussel surveys implemented during the past two decades have shed new light on the scope and severity of this decline. Of the 48 mussel species originally known from Kansas, six are now extirpated, one lacks reproductively viable populations (i.e., faces imminent extirpation), and 38 others have suffered evident range reductions or a widespread thinning of former populations. Soil erosion and stream siltation, other forms of …
