Intergenic Transcription By Rna Polymerase Ii Coordinates Pol Iv And Pol V In Sirna-Directed Transcriptional Gene Silencing In Arabidopsis, Binglian Zheng, Zhengming Wang, Shengben Li, Bin Yu, Jin -Yuan Liu, Xuemei Chen

Center for Plant Science Innovation: Faculty Publications

Intergenic transcription by RNA Polymerase II (Pol II) is widespread in plant and animal genomes, but the functions of intergenic transcription or the resulting noncoding transcripts are poorly understood. Here, we show that Arabidopsis Pol II is indispensable for endogenous siRNA-mediated transcriptional gene silencing (TGS) at intergenic low-copy-number loci, despite the presence of two other polymerases—Pol IV and Pol V—that specialize in TGS through siRNAs. We show that Pol II produces noncoding scaffold transcripts that originate outside of heterochromatic, siRNA-generating loci. Through these transcripts and physical interactions with the siRNA effector protein ARGONAUTE4 (AGO4), Pol II recruits AGO4/siRNAs to homologous …

Indigenous Sovereignty In State-Native Conflicts: A Comparative Study Of Process And Outcomes, Christina Farnsworth

Electronic Theses and Dissertations

Using the United Nations Declaration on the Rights of Indigenous Peoples, this study was designed to understand how indigenous groups assert their sovereign rights in conflict situations, and how they can be most successful in doing so.

Two instances of indigenous-state conflict were analyzed and compared both to each other and to a baseline of what sovereignty in conflict is, based on the United Nations Declaration. Data to be analyzed and compared was gathered through extensive archival research and interviews with tribal members and other interested parties.

The results documented the interaction between indigenous groups and state/provincial and federal governments …

A Framework For Examining Leadership In Extreme Contexts, Sean T. Hannah, Mary Uhl-Bien, Bruce Avolio, Fabrice L. Cavarretta

Department of Management: Faculty Publications

In this review, we develop a framework to guide future research and to examine the execution of leadership in extreme contexts. We start by defining and distinguishing extreme contexts from crisis and other contexts. A five component typology is developed comprised of magnitude of consequences, form of threat, probability of consequences, location in time and physical or psychological–social proximity. We discuss the unique influences these components have on leadership processes in extreme contexts examining the relevance of organization types such as critical action and high reliability organizations. Further, we present a set of factors that may attenuate or intensify the …

Coral reefs playa critically important rale in both the human and natural world. They are among the most biologically rich systems on earth and are of significant economic value to humans. They act as an important base for fisheries and provide food, jobs, and income to billions of people worldwide. These delicate systems are increasingly threatened by several factors including overexploitation pollution, habitat loss, coral bleaching, and global climate change. The rapid decline and loss of the ancient and complex ecosystems had triggered major concern all over the world. Hawaiian coral reefs have experienced an increasing exposure to anthropogenic threats …

Managing Wild Pigs: A Technical Guide, Ben C. West, Andrea L. Cooper, James B. Armstrong Jan 2009

Managing Wild Pigs: A Technical Guide, Ben C. West, Andrea L. Cooper, James B. Armstrong

Human–Wildlife Interactions Monographs

Wild pigs are and will continue to be a challenging problem for wildlife managers, landowners, farmers, conservationists, and others. Despite ongoing control programs, wild pigs have increased both their range and population size. Because of this growth, wildlife managers will increasingly be involved in dealing with problems caused by wild pigs traditionally, as well as emerging problems such as landscaping damage in suburban areas. Successful management strategies will depend upon persistent, adaptive, and integrated management programs that incorporate sound biological and ecological information. An integrated management approach, in addition to addressing the biological and ecological aspects of wild pigs, will …

Polymorphisms And Biologic Effects Of Acidic Mammalian Chitinase In Asthma, Heather Wachtel Jan 2009

Polymorphisms And Biologic Effects Of Acidic Mammalian Chitinase In Asthma, Heather Wachtel

Yale Medicine Thesis Digital Library

In this study, we hypothesize that human acidic mammalian chitinase (AMCase) binds and is regulated by the epidermal growth factor receptor (EGFR), and that AMCase interacts with Galectin-3 (Gal-3) to mediate anti-apoptotic functions. We further hypothesize that asthma-associated polymorphisms of AMCase alter chitinase activity and modulate anti-apoptotic effects. We investigated the interactions between AMCase, Gal-3 and EGFR by establishing binding and co-expression in vitro; apoptotic effects were evaluated via Annexin V/Propidium Iodide staining. Molecular cloning was performed to generate single nucleotide polymorphisms (SNPs) of AMCase associated with asthma. Our data showed that co-expression of AMCase and EGFR induces chitinase activity; …

That's Ok, It's Only A Rental: The Business Of Renting Dogs, Rachit Anand Jan 2009

That's Ok, It's Only A Rental: The Business Of Renting Dogs, Rachit Anand

Hofstra Law Student Works

Dogs are an integral part of our society today. While the benefits arising from dog ownership are widely accepted, proper care of a dog is also time consuming and can be expensive. This paper presents the concept of renting dogs as a substitute for permanent dog ownership and specifically details the business practices of Flexpetz, Inc., a for-profit company that provides such a rental service. Given the possibility of negative physical and emotional effects on the dogs that may flow from being involved in a “renting” business, the paper surveys various anti-cruelty and animal welfare laws (Federal, State, and in …

Dense Connective Tissue Repair : The Intricacies Of Monitoring Actin Isoforms In Native Tissue Models In Vitro, Eran C. Gwillim

Honors Capstones

In preparation to quantify beta-actin and alpha-smooth muscle actin content in loaded and unloaded native rat tail tendon organ culture models, several methodologies were developed in the present paper. Since alpha-smooth muscle actin has not been monitored in tendon in vitro, developing the methods to culture and quantitatively detect alpha-smooth muscle actin, the protein marker for fibroblast to myofibroblast transition were developed. In addition, quantifying the amount of alpha-smooth muscle actin mRNA, using reverse transcription polymerase chain reaction (RT-PCR) assays was investigated and novel oligonucleotide primers were developed. A method to identify four highly conserved actin isoforms; beta cytoplasmic, gamma …

Nietzsche, Virtue, And The Horror Of Existence, Philip J. Kain Jan 2009

Nietzsche, Virtue, And The Horror Of Existence, Philip J. Kain

Philosophy

Robert Solomon argues that Nietzsche is committed to a virtue ethic like Aristotle's. Solomon’s approach seems unaware of Nietzsche’s belief in the horror of existence. A life that contains as much suffering as Nietzsche expects a life to contain, could not be considered a good life by Aristotle. To go further, as Nietzsche does in his doctrines of eternal recurrence and amor fati, to advocate loving such a fate, to refuse to change the slightest detail, Aristotle would find debased. Nietzsche is committed to a virtue ethic, but not an Aristotelian one.

Local Interaction And Long Distance Connections In The Ulua Valley: The View From Cerro Palenque, Julia A. Hendon

Anthropology Faculty Publications

The site of Cerro Palenque, the largest settlement in the lower Ulua Valley (Sula Valley) in Honduras during the ninth and tenth centuries AD, was a locus of craft production of figurines and pottery, feasting, the ballgame, and other events associated with its ballcourt. Based on the analysis of imported obsidian, the evidence for ritual and craft production, and the layout of the settlement, Cerro Palenque maintained long distance trade connections with Mexico, Guatemala, and Belize. It also took part in local rituals and events with its smaller neighbors in the valley.

Defining Family: Naming, Orientation, And Redemption In The Case Of Terri Schiavo, M. Chad Mcbride, Karen L. Daas, Paige W. Toller Jan 2009

Defining Family: Naming, Orientation, And Redemption In The Case Of Terri Schiavo, M. Chad Mcbride, Karen L. Daas, Paige W. Toller

Iowa Journal of Communication

This paper undertakes a detailed analysis of the Terri Schiavo case as it was covered in popular media. Drawing on Burkean theory, we argue a critical issue in the case was a struggle between Terri's parents and husband to be seen as the more legitimate family in order to determine the duration and extent of Terri's medical care. We discuss how the private debate over Terri's health and the decision to remove her feeding tube entered into the public scenes of legal and political action. This shift to the public scene represented problems for the parties directly involved in the …

The Evolution Of Opioid/Orphanin Receptors In Chordates, Jazalle Deshaun Mcclendon Jan 2009

The Evolution Of Opioid/Orphanin Receptors In Chordates, Jazalle Deshaun Mcclendon

Electronic Theses and Dissertations

The interest in the opioid/orphanin gene family stems from functional similarities that these peptides have to the narcotic opium. Opiates have been extensively studied because of their analgesic properties; however, the reason that these plant products can affect the human central nervous system was a mystery until the discovery of opiate-like peptides. The endogenous opioid peptides are well understood today because they have been fully cloned and characterized in several different organisms including lower chordates. On the other hand, the opioid/orphanin receptors have not been fully cloned or characterized in lower chordates; therefore, to better understand the past and present …

Amino Acid Residues Implicated In The Interaction Of Melanocortin Ligands And Their Receptors: A Study Of Mc2r Selectivity, Kristopher D. Veo Jan 2009

Amino Acid Residues Implicated In The Interaction Of Melanocortin Ligands And Their Receptors: A Study Of Mc2r Selectivity, Kristopher D. Veo

Electronic Theses and Dissertations

Melanocortin receptor ligand selectivity has been a question not easily answered. The inability to functionally express melanocortin 2 receptor (MC2R) has inhibited the study of why MC2R is only stimulated by ACTH, a melanocortin hormone. With the recent discovery of the MC2R accessory protein (MRAP), creating a heterologous system is now feasible. Using a general cell line like CHO-K1 cells, which do not express endogenous MCRs, we were able to create a heterologous expression system and test the selectivity of MC2R using analog variants of ACTH(1-24). Our results indicate an amino acid requirement in the C-terminal portion of ACTH(1-24) for …

Soil Amino Acids At Upper Tree Line, Montane And Lower Tree Line, Kelly C. Owens Jan 2009

Soil Amino Acids At Upper Tree Line, Montane And Lower Tree Line, Kelly C. Owens

Electronic Theses and Dissertations

The classic view of the nitrogen cycle in soils is for plants to take up inorganic N in solution for N nutrition. More recent studies reveal plants can take up low molecular weight dissolved organic N such as amino acids directly from the soil. In ecosystems where the rate of microbial mineralization is limited, plants may take up 10 to 200 percent more amino acid N than mineral N. It is not known if plants take up amino acids in all ecosystems, however recent research shows that plants generally take up amino acids when they are present in high quantities …

Livestock And Vegetation Responses To Stocking Rate And Grazing System, Justin D. Derner Jan 2009

Livestock And Vegetation Responses To Stocking Rate And Grazing System, Justin D. Derner

Range Beef Cow Symposium

Vegetation and livestock responses from a 25 year (1982-2006) grazing system and stocking rate study in the northern mixed-grass prairie at the USDA-Agricultural Research Service, High Plains Grasslands Research Station near Cheyenne, Wyoming are presented here. This study was longest known grazing system X stocking rate study in North America and was one of the few studies to use the same stocking rates for grazing system comparisons. Results from this study have been used to further development of, and validate, rangeland models for tactical (within-year) and strategic (across years) decision support tools for producers.

Estimating Survival Of Precocial Chicks During The Prefledging Period Using A Catch-Curve Analysis And Count-Based Age-Class Data, Conor P. Mcgowan, Joshua J. Millspaugh, Mark R. Ryan, Casey D. Kruse, Greg Pavelka Jan 2009

Estimating Survival Of Precocial Chicks During The Prefledging Period Using A Catch-Curve Analysis And Count-Based Age-Class Data, Conor P. Mcgowan, Joshua J. Millspaugh, Mark R. Ryan, Casey D. Kruse, Greg Pavelka

United States Army Corps of Engineers, Omaha District

ABSTRACT: Estimating reproductive success for birds with pre-cocial young can be difficult because chicks leave nests soon after hatching and individuals or broods can be difficult to track. Researchers often turn to estimating survival during the pre-fledging period and, though effective, mark-recapture based approaches are not always feasible due to cost, time, and animal welfare concerns. Using a threatened population of Piping Plovers (Charadrius melodus) that breeds along the Missouri River, we present an approach for estimating chick survival during the pre-fledging period using long-term (1993–2005), count-based, age-class data. We used a modified catch-curve analysis, and data collected …

Altering The Motivational Function Of Nicotine Through Conditioning Processes, Rick A. Bevins Jan 2009

Altering The Motivational Function Of Nicotine Through Conditioning Processes, Rick A. Bevins

Department of Psychology: Faculty Publications

The collection of chapters in this 55th Nebraska Symposium on Motivation Volume clearly highlights that effective strategies for reducing compulsive tobacco use will require a multifaceted approach in which genetic, neurobiological, individual, and cultural factors are considered. It is difficult, if not impossible, to predict where the next important breakthrough will come from (Bevins & Bardo, 2004; Dethier, 1966; Laidler, 1998). Accordingly, further research that extends and challenges current theory and practice at each of these levels of analysis is needed. The continuing focus of our research program, and the topic of the present chapter, is on the role of …

Acute Tryptophan Depletion And Sweet Food Consumption By Overweight Adults, Sherry L. Pagoto, Bonnie Spring, Dennis E. Mcchargue, Brian Hitsman, Malaina Smith, Bradley Appelhans, Donald Hedeker Jan 2009

Department of Psychology: Faculty Publications

Serotonergic involvement has been implicated in preferential consumption of treat foods. We tested the effect of acute tryptophan depletion (ATD) on food consumption by overweight and lean adults with and without a history of recurrent major depressive disorder (MDD). ATD and taste-matched placebo challenges were administered double-blind in counter-balanced order. Participants were classified as lean (n = 36) or overweight (n=19) on the basis of body mass index (BMI). Total calorie, carbohydrate, protein, and sweet food consumption were assessed via a test meal 8-hours following ATD. Four food items of comparable palatability were offered as a part of the test: …
