A New Triggering Receptor Expressed On Myeloid Cells (Trem) Family Member, Trem-Like 4, Binds To Dead Cells And Is A Dnax Activation Protein 12-Linked Marker For Subsets Of Mouse Macrophages And Dendritic Cells, Hiroaki Hemmi, Juliana Idoyaga, Koji Suda

Publications

Dendritic cells (DCs) are professional APCs that can control immune responses against self and altered self, typically foreign, determinants. DCs can be divided into several subsets, including CD8α+ and CD8α- DCs. These subsets possess specific functions. For example, mouse splenic CD8α+, but not CD8α- DCs selectively take up dying cells and cross-present cell-associated Ags to naive T cells. In this study, we identified genes that were more expressed in CD8α+ than CD8α- DCs by microarray analysis. Only one of these genes, when the extracellular domains were linked to human IgG Fc domain, could …

Antibody To Langerin/Cd207 Localizes Large Numbers Of Cd8Α + Dendritic Cells To The Marginal Zone Of Mouse Spleen, Juliana Idoyaga, Nao Suda, Koji Suda Jan 2009

Antibody To Langerin/Cd207 Localizes Large Numbers Of Cd8Α + Dendritic Cells To The Marginal Zone Of Mouse Spleen, Juliana Idoyaga, Nao Suda, Koji Suda

Publications

Dendritic cells (DCs) are strategically positioned to take up antigens and initiate adaptive immunity. One DC subset expresses CD8αα in mice and is specialized to capture dying cells and process antigens for MHC class I "cross-presentation." Because CD8 + DCs also express DEC205/CD205, which is localized to splenic T cell regions, it is thought that CD8 + DCs also are restricted to T zones. Here, we used a new antibody to Langerin/CD207, which colabels isolated CD8 + CD205 + DCs, to immunolabel spleen sections. The mAb labeled discrete cells with high levels of CD11c and CD8. Surprisingly most CD207 + …

Synthetic Double-Stranded Rnas Are Adjuvants For The Induction Of T Helper 1 And Humoral Immune Responses To Human Papillomavirus In Rhesus Macaques, Christiane P. Stahl-Hennig, Martin Eisenblätter, Edith Yasny Jan 2009

Synthetic Double-Stranded Rnas Are Adjuvants For The Induction Of T Helper 1 And Humoral Immune Responses To Human Papillomavirus In Rhesus Macaques, Christiane P. Stahl-Hennig, Martin Eisenblätter, Edith Yasny

Publications

Toll-like receptor (TLR) ligands are being considered as adjuvants for the induction of antigen-specific immune responses, as in the design of vaccines. Polyriboinosinic-polyribocytoidylic acid (poly I:C), a synthetic double-stranded RNA (dsRNA), is recognized by TLR3 and other intracellular receptors. Poly ICLC is a poly I:C analogue, which has been stabilized against the serum nucleases that are present in the plasma of primates. Poly I:C12U, another analogue, is less toxic but also less stable in vivo than poly I:C, and TLR3 is essential for its recognition. To study the effects of these compounds on the induction of protein-specific immune responses in …

Dendritic Cells Require A Systemic Type I Interferon Response To Mature And Induce Cd4+ Th1 Immunity With Poly Ic As Adjuvant, Maria` P. Longhi, Christine Trumpfheller, Juliana Idoyaga Jan 2009

Dendritic Cells Require A Systemic Type I Interferon Response To Mature And Induce Cd4+ Th1 Immunity With Poly Ic As Adjuvant, Maria` P. Longhi, Christine Trumpfheller, Juliana Idoyaga

Publications

Relative to several other toll-like receptor (TLR) agonists, we found polyinosinic:polycytidylic acid (poly IC) to be the most effective adjuvant for Th1 CD4+ T cell responses to a dendritic cell (DC)-targeted HIV gag protein vaccine in mice. To identify mechanisms for adjuvant action in the intact animal and the polyclonal T cell repertoire, we found poly IC to be the most effective inducer of type I interferon (IFN), which was produced by DEC-205+ DCs, monocytes, and stromal cells. Antibody blocking or deletion of type I IFN receptor showed that IFN was essential for DC maturation and development of …

An Evaluation Of Potential Occupational Exposure To Asbestiform Amphiboles Near A Former Vermiculite Mine, Julie Hart, Terry Spear, Tony Ward, Caitlan Baldwin, Marissa Salo, Mohamed Elasheb

Safety Health & Industrial Hygiene

Amphibole asbestos (AA) has been detected on the surface of tree bark in forests neighboring an abandoned vermiculite mine near Libby, Montana. In the present study, simulations were performed to assess potential AA exposure associated with United States Department of Agriculture Forest Service (FS) occupational activities. Bark samples were collected prior, and personal breathing zone (PBZ) and Tyvek clothing wipe samples were collected during and immediately after trials that simulated FS activities. Transmission electron microscopy (TEM) analyses revealed AA bark concentrations up to 15 million structures per square centimeter (s/cm2). AA was detected in 25% of the PBZ TEM samples. …

Creating Lakes from Open Pit Mines: Processes and Considerations, Emphasis on Northern Environments. This document summarizes the literature of mining pit lakes (through 2007), with a particular focus on issues that are likely to be of special relevance to the creation and management of pit lakes in northern climates. Pit lakes are simply waterbodies formed by filling the open pit left upon the completion of mining operations with water. Like natural lakes, mining pit lakes display a huge diversity in each of these subject areas. However, pit lakes are young and therefore are typically in a non-equilibrium state with respect …

Shedding, Witchcraft, And The Romantic Subject: Feminist Appropriation Of The Witch In Sarah Kirsch’S Zaubersprüche, Qinna Shen Jan 2009

Shedding, Witchcraft, And The Romantic Subject: Feminist Appropriation Of The Witch In Sarah Kirsch’S Zaubersprüche, Qinna Shen

German Faculty Research and Scholarship

Against a background of the feminist appropriation of the witch taking place concurrently in second-wave American, French and West German feminism, the paper examines Sarah Kirsch’s appropriation of the witch as a subversive figure in her poetry cycle Zaubersprüche (Conjurations, 1973). In subverting the traditional image of the witch, Kirsch establishes a new one: that of a feminist witch and a feminist witch-writer. The witch is both the fictive character created by Kirsch, and her own self-designation; in the latter case, writing, especially writing in the experimental fashion, is a form of witchcraft. The paper analyzes the poems …

The Virtues Of Balm In Late Medieval Literature, Elly Truitt Jan 2009

The Virtues Of Balm In Late Medieval Literature, Elly Truitt

History Faculty Research and Scholarship

This article argues that balm, or balsam, was, by the late medieval period, believed to be a panacea, capable of healing wounds and illnesses, and also preventing putrefaction. Natural history and pharmacological texts on balm from the ancient and late antique periods emphasized specific qualities of balm, especially its heat; these were condensed and repeated in medieval encyclopedias. The rarity and cost of balsam, from antiquity through the medieval period, and the high rate of counterfeiting also demonstrate its high demand and significance in medicine and religious ritual. Travel writing and itineraria from the early and central medieval periods added …

Supersession, The Comedia Nueva, And Tirso's La Mejor Espigadera, Matthew D. Stroud Jan 2009

Supersession, The Comedia Nueva, And Tirso's La Mejor Espigadera, Matthew D. Stroud

Modern Languages and Literatures Faculty Research

Given Spain's self-identification with the Roman Catholic Church under the Hapsburgs, what is one to make of the great number of comedias that take as their protagonists figures from the Hebrew Bible, individuals revered by Jews as righteous ancestors, models of behavior, and illustrious examples of the triumphs of the Hebrew people faced with endless persecution and oppression? Most of these plays focus on the actions of men (e.g., King David in Tirso’s La venganza de Tamar, and Joseph and Jacob in Mira’s El más feliz cautiverio), but a number of them focus on righteous Hebrew women such …

Characterization And Differentiation Of Peripheral Blood Derived Multipotent Adult Progenitor Cells, Hari Satya Shankar Addagarla

Theses, Dissertations and Capstones

Stem cells are populations of undifferentiated cells that are found in most tissues and act as precursors for regeneration and maintenance. In the future, they could provide promising therapies for diseases which are to date incurable. Our lab developed a novel cell line from the peripheral blood of adult transgenic green fluorescent protein swine and designated them as Peripheral Blood Derived Multipotent Adult Progenitor Cells (PBD-MAPCs). In this study we characterized the mRNA and protein expression profiles of PBD-MAPCs before and after neural differentiation and investigated the potential of PBD-MAPCs to differentiate into myocardial or neural lineages in vitro. We …

The Ohio River extends 981 miles from Pittsburgh, PA to Cairo, Ill providing drinking water to over three million people, a natural habitat for aquatic life, a public recreation resource, a major transportation route, and a source of water for industry. The Guyandotte River is a highly impacted tributary emptying into the Ohio River in Huntington, WV. The objectives of this study were to determine if sediment load is correlated to the number of antibiotic resistant bacteria (ARB) and determine if a single surface sample is a sufficient representative measurement of ARB populations in a large river. In 2007, subsurface …

The Present Giver And Other Stories On Human Connections, Erin B. Waggoner Jan 2009

The Present Giver And Other Stories On Human Connections, Erin B. Waggoner

Theses, Dissertations and Capstones

The Present Giver and Other Stories on Human Connections is a collection of seven short stories dealing with individuals that struggle to connect to another person. However, the stories also explore that these characters still feel the need to connect, stories very indicative of my own struggles with apathy and relationships. The critical analysis takes on a creative non-fiction approach as a way to show my development as a writer and how these stories relate to what I've learned through the years from my love of reading.

Developing A Monitoring Protocol For The Monkey River Watershed, Belize, Central America, Sean Elliott Collins Jan 2009

Developing A Monitoring Protocol For The Monkey River Watershed, Belize, Central America, Sean Elliott Collins

Theses, Dissertations and Capstones

The study of tropical aquatic systems has been limited. Research in developing countries can be challenging due to inadequate resources and cultural variety. Generally, efforts are concentrated on developing and maintaining economic stability rather than ecological sustainability. The aim of this project was to preliminarily develop and utilize a rapid bioassessment protocol (RBP) for the Monkey River watershed in Belize by determining which metrics best described overall stream health. Like biomonitoring protocols already established for temperate systems, a regional tropical aquatic watershed monitoring program should provide information including stream and watershed health. These protocols score systems on a variety of …

Effect Of Growth Hormone On Hippocampal Synaptic Function During Sleep Deprivation, Eun Young Kim Jan 2009

Effect Of Growth Hormone On Hippocampal Synaptic Function During Sleep Deprivation, Eun Young Kim

Theses, Dissertations and Capstones

Long term potentiation (LTP) in the hippocampus is considered a cellular basis of learning memory. Sleep deprivation, especially rapid eye movement (REM) sleep deprivation, impairs learning and memory as well as LTP. Since most of the previous LTP studies were conducted in the in vitro condition, the full consequences of sleep deprivation (SD) in the living animal are yet to be found. Thus, I tested hippocampal LTP in living animals after 5 days of REM sleep deprivation to determine the effect of SD in vivo. SD also disrupts growth hormone (GH) release. Recent evidence indicates that GH regulates cognitive and …

Loss Of Ski Expression In Testicular Cancer Leads To An Enhanced Invasive Phenotype Through Both Bmp-Dependent And Bmp-Independent Pathways, Amy N. Nash Jan 2009

Loss Of Ski Expression In Testicular Cancer Leads To An Enhanced Invasive Phenotype Through Both Bmp-Dependent And Bmp-Independent Pathways, Amy N. Nash

Theses, Dissertations and Capstones

The proto-oncogene SKI is a transcription factor and a co-repressor of the TGFβ superfamily, including TGF

β and BMP. However, additional data suggests that SKI may function as a tumor suppressor in some cell types. The TGFβ superfamily has been implicated in cancer progression and germ cell migration. Testicular cancer afflicts men during their peak reproductive years and is the most common cancer among men of this age group. Cisplatin-based chemotherapy is the standard treatment for testicular cancer. This treatment can lead to undesirable side effects, including infertility. We have shown that SKI expression is decreased in testicular germ cell …
