The Relative Ages Of Ectomycorrhizal Mushrooms And Their Plant Hosts Estimated Using Bayesian Relaxed Molecular Clock Analyses, David S. Hibbett, P. Brandon Matheny

Biology

Background: Ectomycorrhizae (ECM) are symbioses formed by polyphyletic assemblages of fungi (mostly Agaricomycetes) and plants (mostly Pinaceae and angiosperms in the rosid clade). Efforts to reconstruct the evolution of the ECM habit in Agaricomycetes have yielded vastly different results, ranging from scenarios with many relatively recent origins of the symbiosis and no reversals to the free-living condition; a single ancient origin of ECM and many subsequent transitions to the free-living condition; or multiple gains and losses of the association. To test the plausibility of these scenarios, we performed Bayesian relaxed molecular clock analyses including fungi, plants, and other eukaryotes, based …

Department of Molecular Physiology and Biophysics Faculty Papers

RhoA-activated kinase (ROK) is involved in the disorders of smooth muscle contraction found in hypertension model animals and patients. We examined whether the alpha1-adrenergic receptor agonist-induced ROK signal is perturbed in resistance small mesentery artery (SMA) of Lyon genetically hypertensive (LH) rats, using a ROK antagonist, Y27632. Smooth muscle strips of SMA and aorta were isolated from LH and Lyon normotensive (LN) rats. After Ca(2+)-depletion and pre-treatment with phenylephrine (PE), smooth muscle contraction was induced by serial additions of CaCl(2). In LH SMA Ca(2+) permeated cells to a lesser extent as compared with LN SMA, while CaCl(2)-induced contraction of LH …

Does Goal Generation Vary As A Function Of Depressive Status?, Lindsay Brauer Mar 2009

Does Goal Generation Vary As A Function Of Depressive Status?, Lindsay Brauer

USF Tampa Graduate Theses and Dissertations

Although many theories attempt to explain the manner in which deficits in goal generation may lead to an episode of Major Depressive Disorder (MDD), few studies have actually described how persons with depression vulnerability generate goals for themselves. The current project used a multi-dimensional approach to examine the types, numbers, and domains in which goals are generated, comparing individuals with MDD with individuals with remitted depression, and never-depressed controls. In addition, we examined possible mechanisms that may mediate the relationship between depressive status and goal generation. Surprisingly, results suggested little group difference in the types of goals or subjective ratings …

Thermal Inactivation Of Bacillus Anthracis Using Laser Irradiation Of Micro-Etched Platforms, Jeffrey B. Bacon Mar 2009

Thermal Inactivation Of Bacillus Anthracis Using Laser Irradiation Of Micro-Etched Platforms, Jeffrey B. Bacon

Theses and Dissertations

The purpose of this research was to determine the probability of kill for a thermal inactivation strategy for use against biological agents; specifically the resilient endospore of Bacillus anthracis (Ba). The effort focused on short durations (milliseconds to several seconds) and temperatures (300 to 1300 K) simulating the periphery effects after an explosion generated by conventional munitions. For an improved statistical counting, applied microlithography techniques were used to produce micro-etched glass platforms consisting of 532 circular sample wells, evenly spaced. Small carbon black radiators, which provide fast heating/cooling rate and confined temperature distribution, were produced by populating the etched wells …

In Vitro Toxicity Of Silver Nanoparticles In Human Lung Epithelial Cells, Christina R. Kearns

Theses and Dissertations

Nanotechnology is quickly becoming incorporated into everyday products and uses. Silver nanoparticles, specifically, are being used in commercial products, to include aerosols. The purpose of this research was to determine whether silver nanoparticles are toxic to human lung epithelial cells. Different types (coated vs. uncoated), concentrations (10, 50, 100, and 200 µg/mL) and sizes (coated 5 and 80nm, uncoated 10 and 80nm) of silver nanoparticles were used during this study. Toxicity measurements were completed through in vitro techniques. Another study was also completed on toxicity mechanisms by measuring the reactive oxygen species (ROS) generated. Results showed that silver nanoparticles induce …

The Use Of Spider Webs As Passive Bioaerosol Collectors, Daniel I. Mattei Mar 2009

The Use Of Spider Webs As Passive Bioaerosol Collectors, Daniel I. Mattei

Theses and Dissertations

In this experiment, spider webs demonstrated their suitability as passive bioaerosol collectors. For spider webs to be considered suitable passive collectors webs had to satisfy three basic conditions; (1) collection of microorganisms without discrimination based on species or size, (2) collection under variable environmental conditions, and (3) saturation avoidance in the presence of strong microbial launching sources. Four field sampling locations were used, a waste water treatment facility, a commercial garden center, a secluded state park area, and a parking garage. Microscopy cover glass slides were used as the collection instrument. The methodology assured sterility during collection and promoted in …

Modeling Thermal Inactivation Of Bacillus Spores, Emily A. Knight Mar 2009

Modeling Thermal Inactivation Of Bacillus Spores, Emily A. Knight

Theses and Dissertations

This research models and analyzes methods to damage Bacillus anthracis spores through heat treatment. AFIT researchers have developed methods to characterize the effects of heating spores to high temperatures and for short durations similar to the thermal pulse of conventional weapon detonation. This research models the current experiment and evaluates the rate of thermal diffusion throughout the spores. A micro-model of the effects of dry and wet heating on a spore is presented. Heating a spore energizes adsorbed, absorbed, and chemically bound water molecules. These energized molecules have greater mobility within the spore, as well as between the spore and …

Model-Based Control Using Model And Mechanization Fusion Techniques For Image-Aided Navigation, Constance D. Hendrix Mar 2009

Model-Based Control Using Model And Mechanization Fusion Techniques For Image-Aided Navigation, Constance D. Hendrix

Theses and Dissertations

Unmanned aerial vehicles are no longer used for just reconnaissance. Current requirements call for smaller autonomous vehicles that replace the human in high-risk activities. Many times these activities are performed in GPS-degraded environments. Without GPS providing today's most accurate navigation solution, autonomous navigation in tight areas is more difficult. Today, image-aided navigation is used and other methods are explored to more accurately navigate in such areas (e.g., indoors). This thesis explores the use of inertial measurements and navigation solution updates using cameras with a model-based Linear Quadratic Gaussian controller. To demonstrate the methods behind this research, the controller will provide …

Waste Vegetable Oil As An Alternative Fuel For Diesel Vehicles, Harvey S. Gaber

Theses and Dissertations

Alternative fuels have become a hot topic in the news as the cost of oil remains volatile. Questions of whether acquiring alternative fuels are worth the cost, logistics, and political implications are being asked. A possible solution may be currently thrown away by Wright Patterson Air Force Base’s (WPAFB) dining establishments in the form of Waste Vegetable Oil (WVO). This study researched the benefits and costs of pursuing the installation of a WVO to Straight Vegetable Oil fuel processing center and using the fuel to power some of the base’s diesel vehicles. A pilot program was fielded utilizing the Wright …

A Metagenomic Analysis Of Microbial Contamination In Aviation Fuels, Jerrod P. Mccomb Mar 2009

A Metagenomic Analysis Of Microbial Contamination In Aviation Fuels, Jerrod P. Mccomb

Theses and Dissertations

Microbial contamination in aviation fuel arises due to the impracticality of keeping fuel tanks sterile and the inevitable presence of water from condensation. Microbial contaminants in aviation fuels are a concern because of their potential to degrade the fuel, accelerate corrosion within the fuel tank, and threaten flight safety. This research aids in mitigating those problems by comprehensively characterizing the microbial communities affecting aviation fuels. Advances in molecular biological techniques have allowed for the identification of microorganisms which were not identified by the traditional culture-based methodologies used in previous studies. This study employed a molecular method known as 16S rDNA …

Using Agent-Based Modeling To Evaluate Uas Behaviors In A Target-Rich Environment, Joseph A. Van Kuiken

Theses and Dissertations

The trade-off between accuracy and speed is a re-occurring dilemma in many facets of military performance evaluation. This is an especially important issue in the world of ISR. One of the most progressive areas of ISR capabilities has been the utilization of Unmanned Aircraft Systems (UAS). Many people believe that the future of UAS lies in smaller vehicles flying in swarms. We use the agent-based System Effectiveness and Analysis Simulation (SEAS) to create a simulation environment where different configurations of UAS vehicles can process targets and provide output that allows us to gain insight into the benefits and drawbacks of …
