Dna Methylation Analysis Of The Mammalian Peg3 Imprinted Domain, Jennifer M. Huang, Joomyeong Kim
Dna Methylation Analysis Of The Mammalian Peg3 Imprinted Domain, Jennifer M. Huang, Joomyeong Kim
Faculty Publications
In this study, we performed the first systematic survey of DNA methylation status of the CpG islands of the PEG3 (Paternally expressed gene 3) imprinted domain in the mouse, cow, and human genomes. Previous studies have shown that the region surrounding the first exon of PEG3 contains a differentially methylated CpG island. In addition, we have discovered two previously unreported differentially methylated regions (DMR): one in the promoter region of mouse Zim3 and another in the promoter region of human USP29. In the cow, the Peg3-CpG island was the only area that showed DMR status. We have also examined the …
Hyperbolic Discounting Is Rational: Valuing The Far Future With Uncertain Discount Rates, J. Doyne Farmer, John Geanakoplos
Hyperbolic Discounting Is Rational: Valuing The Far Future With Uncertain Discount Rates, J. Doyne Farmer, John Geanakoplos
Cowles Foundation Discussion Papers
Conventional economics supposes that agents value the present vs. the future using an exponential discounting function. In contrast, experiments with animals and humans suggest that agents are better described as hyperbolic discounters, whose discount function decays much more slowly at large times, as a power law. This is generally regarded as being time inconsistent or irrational. We show that when agents cannot be sure of their own future one-period discount rates, then hyperbolic discounting can become rational and exponential discounting irrational. This has important implications for environmental economics, as it implies a much larger weight for the far future.
Assessment Of Invasive Plants Along The Upper Passaic River, Susan Marie Jankovic
Assessment Of Invasive Plants Along The Upper Passaic River, Susan Marie Jankovic
Theses, Dissertations and Culminating Projects
The distribution of invasive plants was determined along the floodplain of the Passaic River in three protected park regions. Specifically, Lord Stirling Park, Hatfield Swamp, and Great Piece Meadows were assessed to determine whether or not the top 29 invasive plants as identified by Snyder and Kaufman (2004) were present. Two of the three areas are strongly affected by urbanization which may be a contributing factor to the increase of invasive plants within them. Eleven out of the top twenty nine invasive plants in New Jersey were recorded: Multiflora rose (Rosa multiflora), Lesser celandine (Ranunculus ficaria), Japanese stiltgrass (Microstegium vimineum), …

The success of clinically relevant immunotherapies requires reversing tumor-induced immunosuppression. Here we demonstrated that linear polyethylenimine-based (PEI-based) nanoparticles encapsulating siRNA were preferentially and avidly engulfed by regulatory DCs expressing CD11c and programmed cell death 1–ligand 1 (PD-L1) at ovarian cancer locations in mice. PEI-siRNA uptake transformed these DCs from immunosuppressive cells to efficient antigen-presenting cells that activated tumor-reactive lymphocytes and exerted direct tumoricidal activity, both in vivo and in situ. PEI triggered robust and selective TLR5 activation in vitro and elicited the production of hallmark TLR5-inducible cytokines in WT mice, but not in Tlr5–/– littermates. Thus, PEI is a TLR5 …

Sperm Motility Initiation And Duration In A Euryhaline Fish, Medaka (Oryzias Latipes), H. Yang, T. R. Tiersch
Faculty Publications
The medaka, Oryzias latipes, is a well-recognized fish model for biomedical research. An understanding of gamete characteristics is necessary for experimental manipulations such as artificial fertilization and sperm cryopreservation. The goal of this study was to investigate sperm characteristics of motility initiation, duration, and retention in medaka. First, motility was initiated by osmolality values ranging from 25 to 686 mOsm/kg, which included deionized water and hypotonic, isotonic, and hypertonic Hanks' balanced salt solution. The percentage of motile sperm was >80% when osmolality was /kg and decreased as osmolality increased. This is different from most fish with external fertilization in which …

Signal transducer and activator of transcription 3 (STAT3) is aberrantly activated in human cancer including lung cancer and has been implicated in transformation, tumorigenicity, and metastasis. One putative downstream gene regulated by Stat3 is MUC1 which also has important roles in tumorigenesis. We determined if Stat3 regulates MUC1 in lung cancer cell lines and what function MUC1 plays in lung cancer cell biology. We examined MUC1 expression in non-small cell lung cancer (NSCLC) cell lines and found high levels of MUC1 protein expression associated with higher levels of tyrosine phosphorylated STAT3. STAT3 knockdown downregulated MUC1 expression whereas constitutive STAT3 expression …

Robert Munford & Mercy Otis Warren : How Gender, Geography, And Goals Affected Their Playwrighting, Kylie A. Horney
Master's Theses
This thesis analyzes the Revolutionary-era plays of Robert Munford and Mercy Otis Warren. Munford’s two comedies, The Candidates and The Patriots, are compared to Warren’s three earliest satires, The Adulateur, The Defeat, and The Group, in an effort to explain some of the differences between these two authors. The original printings of these plays from the Early American Imprints series, as well as more recent scholarship on Munford and Warren, are used to investigate the plays and lives of these playwrights. Munford’s and Warren’s backgrounds are explored to account for variations in their works. While the …

Looking Into The Mind Of The Mother : Pup Exposure And Reactivation Of Maternal Circuits, Tricia Lauren Norkunas
Master's Theses
The female rat, among other species, undergoes a fundamental brain re-modeling as a consequence of experiencing the normal and natural events of pregnancy and offspring stimulation. Compelling data show that maternal experiences produce neurobiological modifications in the female leading to specific maternal behaviors, affective states, and the basic underlying female neurobiology necessary to raise viable offspring. This study aims to evaluate the number, quality and selective activation of neurons that develop during the maternal experience. The study showed a trend toward supporting the hypothesis that a “maternal-circuit” is formed through the proliferation of neurons during late-motherhood and lactation, and is …

Cd19 Signaling Is Impaired In Murine Peritoneal And Splenic B-1 B Lymphocytes, Trivikram Dasu, Vishal Sindhava, Stephen H. Clarke, Subbarao Bondada
Microbiology, Immunology, and Molecular Genetics Faculty Publications
B-1 cells reside predominantly within the coelomic cavities, tonsils, Peyer's patches, spleen (a minor fraction – ∼5%) and are absent in the lymph nodes. They are the primary sources of natural IgM in the body. B-1 cells express polyreactive B cell receptors (BCRs) that cross react with self-antigens and are thus implicated in auto-immune disorders. Previously, we reported that peritoneal B-1 cells are deficient in CD19-mediated intracellular signals leading to Ca2+ mobilization. Here, we find that splenic B-1 cells, like peritoneal B-1 cells, are defective in Ca2+ release upon B cell activation by co-cross-linking BCR and CD19. In …

Department of Plant Pathology: Faculty Publications
The complete genome sequence of Triticum mosaic virus (TriMV), a member in the family Potyviridae, has been determined to be 10,266 nucleotides (nt) excluding the 3′ polyadenylated tail. The genome encodes a large polyprotein of 3,112 amino acids with the “hall-mark proteins” of potyviruses, including a small overlapping gene, PIPO, in the P3 cistron. The genome of TriMV has an unusually long 5′ nontranslated region of 739 nt with 12 translation initiation codons and three small open reading frames, which resemble those of the internal ribosome entry site containing 5′ leader sequences of the members of Picornaviridae. Pairwise …

Hierarchical Bayesian Cortical Models: Analysis And Acceleration On Multicore Architectures, Pavan Yalamanchili
All Theses
There is a significant interest in the research community to develop large scale,
high performance implementations of neuromorphic models. These have the potential to
provide significantly stronger information processing capabilities than current computing
algorithms. This thesis examines the parallelization of two recent biologically inspired
hierarchical Bayesian cortical models onto recent multicore architectures. These models
have been developed recently based on new insights from neuroscience and have several
advantages over traditional neural networks. In particular, they need far fewer network
nodes to simulate a large scale cortical model than traditional neural networks, making
them computationally more efficient. This is the first …

Each year, pediatric traumatic brain injury (TBI) accounts for 435,000 emergency department visits, 37,000 hospital admissions, and approximately 2,500 deaths in the United States. TBI results in immediate injury from direct mechanical force and shear. Secondary injury results from the release of biochemical or inflammatory factors that alter the loco-regional milieu in the acute, subacute, and delayed intervals after a mechanical insult. Preliminary preclinical and clinical research is underway to evaluate the benefit from progenitor cell therapeutics, hypertonic saline infusion, and controlled hypothermia. However, all phase III clinical trials investigating pharmacologic monotherapy for TBI have shown no benefit. A recent …
