Trk: A Neuromodulator Of Age-Specific Behavioral And Neurochemical Responses To Cocaine In Mice., Michelle Niculescu, Shane A Perrine, Jonathan S Miller, Michelle E Ehrlich, Ellen M Unterwald

Farber Institute for Neuroscience Faculty Papers

Responses to psychostimulants vary with age, but the molecular etiologies of these differences are largely unknown. The goal of the present research was to identify age-specific behavioral and molecular adaptations to cocaine and to elucidate the mechanisms involved therein. Postweanling, periadolescent, and adult male CD-1 mice were exposed to cocaine (20 mg/kg) for 7 d. The rewarding effects of cocaine were assessed, as were the response to a Trk antagonist and the regulation of dopamine and cAMP-regulated phosphoprotein, 32 kDa (DARPP-32). Cocaine was rewarding in both periadolescent and adult mice using a conditioned place preference procedure. In contrast, postweanling mice …

Sanders-Brown Center on Aging Faculty Publications

MicroRNAs (miRNAs) are small regulatory RNAs that participate in posttranscriptional gene regulation in a sequence-specific manner. However, little is understood about the role(s) of miRNAs in Alzheimer's disease (AD). We used miRNA expression microarrays on RNA extracted from human brain tissue from the University of Kentucky Alzheimer's Disease Center Brain Bank with near-optimal clinicopathological correlation. Cases were separated into four groups: elderly nondemented with negligible AD-type pathology, nondemented with incipient AD pathology, mild cognitive impairment (MCI) with moderate AD pathology, and AD. Among the AD-related miRNA expression changes, miR-107 was exceptional because miR-107 levels decreased significantly even in patients with …

Microrna, Sex Determination And Floral Meristem Determinacy In Maize, Jo Ann Banks

Department of Botany and Plant Pathology Faculty Publications

Sex determination in the flowers of maize involves the abortion of stamen or pistil development. Recent work investigating genes that control this process reveals that a microRNA is involved in both the sex determination of the male inflorescence and its growth pattern.

Adenomatous Polyposis Coli Is Present Near The Minimal Level Required For Accurate Graded Responses To The Wingless Morphogen, Hassina Benchabane, Edward G. Hughes, Carter M. Takacs, Jason R. Baird, Yashi Ahmed

Dartmouth Scholarship

The mechanisms by which the Wingless (Wg) morphogen modulates the activity of the transcriptional activator Armadillo (Arm) to elicit precise, concentration-dependent cellular responses remain uncertain. Arm is targeted for proteolysis by the Axin/Adenomatous polyposis coli (Apc1 and Apc2)/Zeste-white 3 destruction complex, and Wg-dependent inactivation of destruction complex activity is crucial to trigger Arm signaling. In the prevailing model for Wg transduction, only Axin levels limit destruction complex activity, whereas Apc is present in vast excess. To test this model, we reduced Apc activity to different degrees, and analyzed the effects on three concentration-dependent responses to Arm signaling that specify distinct …

The Rift Between Faiths: Evolution And Intelligent Design, Nicole Sami Jan 2008

The Rift Between Faiths: Evolution And Intelligent Design, Nicole Sami

Reason and Respect

Intelligent design is the concept in which aspects of creationism and evolution come together to form a single theory. Proponents of this theory believe that a higher being created the first forms of life and then over time these creatures adapted to different environments. Teaching intelligent design could potentially cause a problem within society, however: in a democratic society the belief that a person’s opinions should not be impressed upon by another individual is held in high respect. The question therefore becomes, should intelligent design be taught in American public schools?

Monitoring Lys-TrnaLys Phosphatidylglycerol Transferase Activity, Michael Ibba Jan 2008

Monitoring Lys-TrnaLys Phosphatidylglycerol Transferase Activity, Michael Ibba

Biology, Chemistry, and Environmental Sciences Faculty Articles and Research

In some bacteria Lys-tRNALys is used both in translation and for the specific addition of Lys to phosphatidylglycerol in the cytoplasmic membrane. This reaction is catalyzed by the membrane protein MprF, and the lysyl-phosphatidylglycerol formed contributes to the resistance of these bacteria to various cationic antibacterial molecules. Obtaining proteins and reconstituting an in vitro system mimicking membrane conditions is a major challenge to studying the function of membrane proteins, especially when labile substrates such as Lys-tRNALys are required. Here we report methods to obtain a stable enriched membrane fraction containing MprF, and the techniques necessary to quantitatively monitor …

Comparative Secretome Analysis Suggests Low Plant Cell Wall Degrading Capacity In Frankia Symbionts, Juliana E. Mastronunzio, Louis S. Tisa, Philippe Normand, David R. Benson

Molecular, Cellular & Biomedical Sciences

Background: Frankia sp. strains, the nitrogen-fixing facultative endosymbionts of actinorhizal plants, have long been proposed to secrete hydrolytic enzymes such as cellulases, pectinases, and proteases that may contribute to plant root penetration and formation of symbiotic root nodules. These or other secreted proteins might logically be involved in the as yet unknown Molecular interactions between Frankia and their host plants. We compared the genome-based secretomes of three Frankia strains representing diverse host specificities. Signal peptide detection algorithms were used to predict the individual secretomes of each strain, and the set of secreted proteins shared among the strains, termed the core …

Pharmacogenomics: A Primer For Policymakers, Leslie Tucker Jan 2008

Pharmacogenomics: A Primer For Policymakers, Leslie Tucker

National Health Policy Forum

Researchers are exploring how genetic variations among individuals may help explain why a drug can work well in some people and poorly (or not at all) in others, including those who appear to have the same disease. Pharmacogenomics, as this new field is called, aims to help physicians make use of genetic tests to distinguish among patients whose genetic characteristics predispose them to respond in certain ways to certain medicines. If physicians can use this information to quickly and reliably choose the appropriate drug at the most effective dose for each patient, they may produce better patient outcomes and save …

A Generic-Level Phylogenetic Review Of The Macrodactylini (Coleoptera: Scarabaeidae: Melolonthinae), Kerry Katovich

Insecta Mundi

A revision of the generic classification of the tribe Macrodactylini is provided using morphological characters of adults. The revision is based on a taxonomic analysis of 80 genera historically placed in the tribe and a cladistic analysis of 32 genera conforming to the new tribal definition. Synapomorphies for the newly defined Macrodactylini include: the length of the fifth ventrite longer than the fourth ventrite when viewed ventrally, fifth ventrite lacking a complete suture between the tergite and sternite, and the metathoracic tibial spurs (if present) offset, allowing the metatarsus to move past them. Thirty-two genera constitute the newly defined Macrodactylini: …

A Remarkable New Species Of Zethus Fabricius (Hymenoptera, Vespidae, Eumeninae) From Costa Rica, Charles C. Porter

Insecta Mundi

Abstract:
Zethus stangei Porter, new species, is described from Costa Rica. It forms a species group of its own, unique in its grotesquely enlarged male mandibles and reduced clypeus, single mid tibial spur, and in having a medio-apical bulge on the second sternite. It was previously identified as Z. magretti Zavattari but reexamination of the type of this species shows that it actually belongs to the coeruleopennis species group.

Resumen:
Se describe a Zethus stangei Porter, especie nueva, de Costa Rica, cuyas características más notables la colocan en su propio grupo, siendo ellas las grandes y abultadas mandíbulas y el …
