Method For Treating A Viral Infection Related Or A Chemical Toxin Related Hepatic Injury With Deltorphin D, Peter R. Oeltgen, Paul D. Bishop, Craig J. Mcclain, Shirish Barve

Pathology and Laboratory Medicine Faculty Patents

A method of modulating cytokine mediated hepatic injury by administering compound-D SEQ ID NO:1 to a mammal. A concentration of the compound in the range of about 0.5 mg/kg to about 20 mg/kg in a physiologically acceptable formulation blocks a cytokine cascade. A therapeutic method of modulating cytokine mediated acute inflammatory, trauma induced and toxin induced hepatic injury, particularly via tumor necrosis factor modulation, is thus disclosed.

Acid-Sensing Ion Channels Interact With And Inhibit Bk K+ Channels, Elena Petroff, Margaret P. Price, Vladislav Snitsarev, Huiyu Gong, Victoria Korovkina, Francois M. Abboud, Michael J. Welsh

Acid-sensing ion channels (ASICs) are neuronal non-voltage-gated cation channels that are activated when extracellular pH falls. They contribute to sensory function and nociception in the peripheral nervous system, and in the brain they contribute to synaptic plasticity and fear responses. Some of the physiologic consequences of disrupting ASIC genes in mice suggested that ASIC channels might modulate neuronal function by mechanisms in addition to their H+-evoked opening. Within ASIC channel's large extracellular domain, we identified sequence resembling that in scorpion toxins that inhibit K+ channels. The causal basis of vertebrate complexity has been sought in genome duplication events (GDEs) that occurred during the emergence of vertebrates, but evidence beyond coincidence is wanting. MicroRNAs (miRNAs) have recently been identified as a viable causal factor in increasing organismal complexity through the action of these ≈22-nt noncoding RNAs in regulating gene expression. Because miRNAs are continuously being added to animalian genomes, and, once integrated into a gene regulatory network, are strongly conserved in primary sequence and rarely secondarily lost, their evolutionary history can be accurately reconstructed. The reference librarian's task is to translate the patron's question into one that can be answered with the library's resources. The first element of that task is to know what the patron wants; the second is to know what resources the library has and how to use them. Reference librarians must learn continuously throughout their careers, both because new resources become available, but also because patrons present questions requiring new resources. This article will focus on how to determine what kind of information the patron needs through the reference interview.

Nuclear Polyglutamine-Containing Protein Aggregates As Active Proteolytic Centers, Min Chen, Lena Singer, Andrea Scharf, Anna Von Mikecz

Biological Sciences Faculty Research & Creative Works

Protein aggregates and nuclear inclusions (NIs) containing components of the ubiquitin-proteasome system (UPS), expanded polyglutamine (polyQ) proteins, and transcriptional coactivators characterize cellular responses to stress and are hallmarks of neurodegenerative diseases. The biological function of polyQ-containing aggregates is unknown. To analyze proteasomal activity within such aggregates, we present a nanoparticle (NP)-based method that enables controlled induction of sodium dodecyl sulfate-resistant inclusions of endogenous nuclear proteins while normal regulatory mechanisms remain in place. Consistent with the idea that the UPS maintains quality control, inhibition of proteasomal proteolysis promotes extra large protein aggregates (1.4-2 μm), whereas formation of NP-induced NIs is found …
