Mesoporous Silica Nanoparticles Inhibit Cellular Respiration, Zhimin Tao, Tewodros Asefa, Cole Duncan, Abhishek Anan, Jerry Goodisman

We studied the effect of two types of mesoporous silica nanoparticles, MCM-41 and SBA-15, on mitochondrial O 2 consumption (respiration) in HL-60 (myeloid) cells, Jurkat (lymphoid) cells, and isolated mitochondria. SBA-15 inhibited cellular respiration at 25-500/ μg/mL; the inhibition was concentration-dependent and time-dependent. The cellular ATP profile paralleled that of respiration. MCM-41 had no noticeable effect on respiration rate. In cells depleted of metabolic fuels, 50 μg/mL SBA-15 delayed the onset of glucose-supported respiration by 12 min and 200 μg/mL SBA-15 by 34 min; MCM-41 also delayed the onset of glucose-supported respiration. The Regulation Of Sirt2 Function By Cyclin-Dependent Kinases Affects Cell Motility, R Pandithage, R Lilischkis, K Harting, A Wolf, Et Al

Cyclin-dependent kinases (Cdks) fulfill key functions in many cellular processes, including cell cycle progression and cytoskeletal dynamics. A limited number of Cdk substrates have been identified with few demonstrated to be regulated by Cdk-dependent phosphorylation. We identify on protein expression arrays novel cyclin E–Cdk2 substrates, including SIRT2, a member of the Sirtuin family of NAD+-dependent deacetylases that targets a-tubulin. We define Ser-331 as the site phosphorylated by cyclin E–Cdk2, cyclin A–Cdk2, and p35–Cdk5 both in vitro and in cells. Importantly, phosphorylation at Ser-331 inhibits the catalytic activity of SIRT2. Gain- and loss-of-function studies demonstrate that SIRT2 interfered with cell adhesion …

Nanotechnology promises to be the defining technology of the 21st century. At an annual investment of $1B, it provides significant contributions to manufacturing, medicine, energy conservation, and the environment. Nanoparticles are structures with at least one dimension in the 1 to 100 nanometer (nm) range. DoD and US Air Force interest in aluminum nanoparticles (AL NPs) stems from its ability to enhance combustion jet fuel, thus increasing fuel efficiency. The addition of AL NPs to JP-8 may pose a unique dermal hazard to aircraft maintenance workers. There is no published data on AL NP toxicity effects on human skin. This …

Analytical and numerical approaches have been developed for modeling temperatures in municipal solid waste landfills. Steps for model formulation and details of boundary conditions are described. The formulation was based on a transient conductive heat transfer analysis. Conventional earth temperature theories were modified for landfill systems by incorporating heat generation functions representing biological decomposition of wastes. Finite element analysis was used for general modeling and parametric evaluations. Thermal properties of materials were determined using field observations and data reported in literature. Racism And Ambulatory Blood Pressure In A Community Sample, Elizabeth Brondolo, Daniel Libby, Ellen-Ge Denton

Objective: Racism has been identified as a psychosocial stressor that may contribute to disparities in the prevalence of cardiovascular disease. The goal of the present article was to investigate the relationship of perceived racism to ambulatory blood pressure (ABP) in a sample of American-born Blacks and Latinos.

Methods: Participants included English-speaking Black or Latino(a) adults between the ages of 24 and 65. They completed daily mood diaries and measures of perceived racism, socioeconomic status, and hostility. Participants were outfitted with ABP monitors; 357 provided data on waking hours only, and 245 provided data on both waking and nocturnal ABP.

This article provides an overview on different types of managing interdisciplinary knowledge integration in environmental urban research projects: 1. "synthesis-then", 2. "synthesis-online", 3. "synthesis-first", 4. "synthesis-as contracted". We present two projects in the context of urban ecology, in order to discuss two types of integration management in more detail. In the "synthesis-first" type of project management, interdisciplinary knowledge integration is organized through "syntheses groups" that represent the relevant perspectives on a case. We introduce "synthesis-first" using the Zurich North case study as an example. In the "synthesis-online" type of project management, interdisciplinary knowledge integration is wanted but occurs only occasionally. …

Actinorhodin, an antibiotic produced by Streptomyces coelicolor, is exported from the cell by the ActA efflux pump. actA is divergently transcribed from actR, which encodes a TetR-like transcriptional repressor. We showed previously that ActR represses transcription by binding to an operator from the actA/actR intergenic region. Importantly, actinorhodin itself or various actinorhodin biosynthetic intermediates can cause ActR to dissociate from its operator, leading to derepression. This suggests that ActR may mediate timely self-resistance to an endogenously produced antibiotic by responding to one of its biosynthetic precursors.
