The Cul7 E3 Ubiquitin Ligase Targets Insulin Receptor Substrate 1 For Ubiquitin-Dependent Degradation, Pascal J. Lafontant

Recent genetic studies have documented a pivotal growth regulatory role played by the Cullin 7 (CUL7) E3 ubiquitin ligase complex containing the Fbw8-substrate-targeting subunit, Skp1, and the ROC1 RING finger protein. In this report, we identified insulin receptor substrate 1 (IRS-1), a critical mediator of the insulin/insulin-like growth factor 1 signaling, as a proteolytic target of the CUL7 E3 ligase in a manner that depends on mammalian target of rapamycin and the p70 S6 kinase activities. Interestingly, while embryonic fibroblasts of Cul7-/- mice were found to accumulate IRS-1 and exhibit increased activation of IRS-1’s downstream Akt and MEK/ERK pathways, these …

Modeling The Hydrolyzing Action Of Secretory Phospholipase A2 With Ordinary Differential Equations And Monte Carlo Methods, Zijun Lan Dozier

Although cell membranes normally resist the hydrolysis of secretory phospholipase A2, a series of current investigations demonstrated that the changes in lipid order caused by increased calcium has a relationship with the susceptibility to phospholipase A2. To further explore this relationship, we setup ordinary differential equations models, statistic models and stochastic models to compare the response of human erythrocytes to the hydrolyzing action of secretory phospholipase A2 and the relationship between the susceptibility of hydrolysis and the physical properties of secretory phospholipase A2. Furthermore, we use models to determine the ability of calcium ionophore to increased membrane susceptibility.

Background
Caenorhabditis elegans locomotion is a simple behavior that has been widely used to dissect genetic components of behavior, synaptic transmission, and muscle function. Many of the paradigms that have been created to study C. elegans locomotion rely on qualitative experimenter observation. Here we report the implementation of an automated tracking system developed to quantify the locomotion of multiple individual worms in parallel.
Methodology/Principal Findings
Our tracking system generates a consistent measurement of locomotion that allows direct comparison of results across experiments and experimenters and provides a standard method to share data between laboratories. The tracker utilizes a video camera …

The primary purpose of the study was to investigate student involvement and engagement levels and its impact on various aspects of student life on selected African American students at Rowan University. Data were collected by means of a Likert-style survey. In addition, a focus group was conducted consisting of students from the survey sample. Data analysis from the survey suggests that faculty involvement and active/collaborative learning in the classroom are very important to academic success in college. Holding an internship, making class presentations, and tutoring other students were reported as having a positive impact on GPA. From the focus group, …

Methods, compositions and kits for masking and subsequent removal of oil, grease, rust and other stains from a variety of rough solid surfaces, including, but not limited to stone, concrete, asphalt, stucco brick, and ceramic. The methods include coating the stains with an opaque or translucent thin film of a composition that makes the stain practically indistinguishable against the background, exposing the coated stain to the elements, such as, sunlight, air, moisture, resulting in spontaneous transformation, degradation and subsequent removal of the stains from the solid surfaces. Mixing ratios of chloroiodomethane (CH2ClI) in ambient air were quantified in the coastal North Atlantic region (Thompson Farm, Durham, New Hampshire, and Appledore Island, Maine) and two remote Pacific areas (Christmas Island, Kiribati, and Oahu, Hawaii). Average mixing ratios were 0.15 ± 0.18 and 0.68 ± 0.66 parts per trillion by volume (pptv) at Thompson Farm and Appledore Island, respectively, compared to 0.10 ± 0.05 pptv at Christmas Island and 0.04 ± 0.02 pptv in Hawaii. Photolysis constrained the daytime mixing ratios of CH2ClI at all locations with the minimum occurring at 1600 local time. Daily average fluxes to the …

Atopic diseases and asthma are increasing at a remarkable rate on a global scale. It is now well recognized that asthma is a chronic inflammatory disease of the airways. The inflammatory process in many patients is driven by an immunoglobulin E (IgE)-dependent process. Mast cell activation and release of mediators, in response to allergen and IgE, results in a cascade response, culminating in B lymphocyte, T lymphocyte, eosinophil, fibroblast, smooth muscle cell and endothelial activation. This complex cellular interaction, release of cytokines, chemokines and growth factors and inflammatory remodeling of the airways leads to chronic asthma. A subset of patients …

A new method was developed for the determination of uranium in groundwater above 30µg/L (30ppb). This concentration is the maximum contaminant level (MCL) for uranium in drinking water, which was set in December 2003 by the US EPA under the provisions of the Safe Drinking Water Act. The method is rapid and requires little technical training to conduct, allowing it to be used by consumers, in the laboratory, or in the field. The two-step technique involves preconcentrating uranium using a U/TEVA-2 extraction chromatographic resin followed by complexation with a pyridylazo indicator dye, Br-PADAP. Electrospray ionization is a technique widely used in mass spectrometry. Almost every material, specifically large molecules like proteins or polymers can be ionized directly out of solution. During the ionization process molecules are not fragmented. In this work a prototype apparatus for creating three-dimensional patterns in a ultra high vacuum environment using an electrospray ion source was optimized for higher ion currents hence deposition rate by improving the core component of the apparatus, an electrodynamic ion funnel. Alterations in epithelial mucin expression are associated with carcinogenesis, but there are few data in biliary tract cancer (BTC). In pancreatic malignancy, MUC4 is a diagnostic and prognostic tumour marker, whereas MUC5AC has been proposed as a sensitive serological marker for BTC. We assessed MUC4 and MUC5AC expression in (i) prospectively collected bile and serum specimens from 72 patients with biliary obstruction (39 BTC) by real-time reverse transcriptase-PCR (qPCR) and western blot analysis, and (ii) 79 archived biliary tissues (69 BTC) by immunohistochemistry. Schwann cell myelination depends on Krox-20/Egr2 and other promyelin transcription factors that are activated by axonal signals and control the generation of myelin-forming cells. Myelin-forming cells remain remarkably plastic and can revert to the immature phenotype, a process which is seen in injured nerves and demyelinating neuropathies. We report that c-Jun is an important regulator of this plasticity. At physiological levels, c-Jun inhibits myelin gene activation by Krox-20 or cyclic adenosine monophosphate. c-Jun also drives myelinating cells back to the immature state in transected nerves in vivo. Enforced c-Jun expression inhibits myelination in cocultures. Furthermore, c-Jun and Krox-20 show a …

The red-cockaded woodpecker (Picoides borealis) is a listed endangered species, endemic in the southeastern United States. It is a cooperatively breeding species preferring to live in an open, mature and old growth pine ecosystem. The restoration and management of red-cockaded woodpecker habitat is a difficult task within both public and private land. Forest management practices may have adverse effect on nesting and foraging habitat. To delist the red-cockaded woodpecker from the endangered species list, the U.S. Fish and Wildlife Service developed the 2003 Recovery Plan. Lake Erie is the most productive of the North American Great Lakes and experiences annual periods of hypolimnetic hypoxia with unknown consequences for the microbial food web. We established the abundances and mortality rates of microbes in Lake Erie during thermal stratification and determined how they varied with changes in bottom-water dissolved oxygen concentrations. The microbial plankton community (heterotrophic bacteria, Cyanobacteria, eukaryotic phytoplankton, nanozooplankton, microzooplankton) was quantified in surface and bottom waters along with measurements of herbivory and bacterivory rates on eukaryotic and prokaryotic picoplankton and rates of viral lysis of bacteria.
