Cellular And Synaptic Actions Of Acetylcholine In The Lamprey Spinal Cord, Katharina A. Quinlan, James T. Buchanan

This study investigated cellular and synaptic mechanisms of cholinergic neuromodulation in the in vitro lamprey spinal cord. Most spinal neurons tested responded to local application of acetylcholine (ACh) with depolarization and decreased input resistance. The depolarization persisted in the presence of either tetrodotoxin or muscarinic antagonist scopolamine and was abolished with nicotinic antagonist mecamylamine, indicating a direct depolarization through nicotinic ACh receptors. Local application of muscarinic ACh agonists modulated synaptic strength in the spinal cord by decreasing the amplitude of unitary excitatory and inhibitory postsynaptic potentials. The postsynaptic response to direct application of glutamate was unchanged by muscarinic agonists, suggesting …

Human cholangiocarcinomas evade apoptosis by overexpression of Mcl-1. The drug obatoclax (GX15-070) inhibits antiapoptotic members of the Bcl-2 family including Mcl-1. The purpose of this study is to determine if obatoclax sensitizes human cholangiocarcinoma cells to apoptosis. The human cholangiocarcinoma cell lines, KMCH, KMBC, and TFK, were employed for these studies. Protein expression was assessed by immunoblot and protein-protein interactions detected by coprecipitation of the polypeptide of interest with S-tagged Mcl-1. Activation of Bak and Bax was observed by immunocytochemistry with conformation-specific antisera. Obatoclax induced minimal apoptosis alone; however, it increased apoptosis 3- to 13-fold in all three cancer cell …

The notion of African-American mothers and adult daughters sharing breast health and breast cancer knowledge openly and honestly could be the initial step in addressing the high morbidity and mortality that continues to affect this population. Scholars have long contended that the main purpose of education is to guide youth towards more authentic, fruitful ways of being in the world; this ancient wisdom continues to challenge educators to serve as effective role models (e.g., Aristotle, 1963; 1985; Maslow, 1993; Palmer, 1998). Yet, until now, teachers' levels of self-actualization have not been investigated with valid and reliable instruments. Impacts on water quality and quantity are among the most contentious aspects of mining projects. Companies insist that the use of modern technologies will ensure environmentally friendly mining practices. However, evidence of the negative environmental impacts of past mining activity causes local and downstream populations to worry that new mining activities will adversely affect their water supply. We report on one mine site in Peru where water has become a: particularly conflictive issue. We then provide a detailed proposal for a monitoring plan to recover trust among stakeholders. Wetland indices of biological integrity (IBIs) are used to satisfy the water resources monitoring requirements of the Clean Water Act (CWA). However, debate still exists on what classification systems and taxa to base these IBIs upon. Our cumulative research, representing indices of biological integrity designed for regional HGM subclasses, designated HGM management classes and Cowardin et al. (1979) classes for West Virginia. The indices were derived from metrics calculated from anuran, avian, macroinvertebrate, and vegetation communities; each representing increasing levels of resources associated with gathering the necessary data. Background

Perfluorooctanoic acid (PFOA) is a potent hepatocarcinogen and peroxisome proliferator (PP) in rodents. Humans are not susceptible to peroxisome proliferation and are considered refractory to carcinogenesis by PPs. Previous studies with rainbow trout indicate they are also insensitive to peroxisome proliferation by the PP dehydroepiandrosterone (DHEA), but are still susceptible to enhanced hepatocarcinogenesis after chronic exposure.

Objectives

In this study, we used trout as a unique in vivo tumor model to study the potential for PFOA carcinogenesis in the absence of peroxisome proliferation compared with the structurally diverse PPs clofibrate (CLOF) and DHEA. Mechanisms of carcinogenesis were identified from …

Squash bug (Anasa tristis) is a “true bug” with piercingsucking mouthparts (Order Hemiptera) in the leaffooted bug family (Coreidae). It is common throughout the U.S. and found from Canada to Central America. Adults (Fig. 1) emit a foul odor when disturbed and may be called “stink bugs”; however, true stink bugs are in a different true bug family.

Group II metabotropic glutamate (mGlu II) receptors subtype 2 and 3 (mGlu2 and mGlu3) are subtle regulators of neuronal excitability and synaptic plasticity in the hippocampus. In recent years, researchers have investigated the potential neuroprotective and anticonvulsant effects of compounds acting on mGlu II receptors. However, abnormal expression and function of mGlu2 and mGlu3 have been reported in temporal lobe epilepsy, a phenomena that may limit the therapeutic effectiveness of these potentially new antiepileptic drugs. Epileptogenesis in mesial temporal lobe epilepsy is determined by several factors including abnormalities in the expression and function of ion channels. Here, we report a long-lasting deficit in gene expression of Kcnma1 coding for the large-conductance calcium-activated potassium (BK, MaxiK) channel α-subunits after pilocarpine-induced status epilepticus. By using comparative realtime PCR, Taqman gene expression assays, and the delta-delta comparative threshold method we detected a significant reduction in Kcnma1 expression in microdissected dentate gyrus at different intervals after status epilepticus (24 h, 10 days, 1 month, and more than 2 months). This Article explores the widely disputed issue of whether Takings Clause protects against regulatory takings, offering a novel and intermediate solution. Critics of the regulatory takings doctrine have argued that the original meaning of the Fifth Amendment Takings Clause does not cover regulatory takings. They have quickly moved from this claim to the conclusion that the incorporated Takings Clause under the Fourteenth Amendment also does not cover regulatory takings. This study is a mixed-method design focusing on ten White and ten non-White elementary principals working in urban areas. The qualitative methodology uses interviews and observations to examine how principals conceptualize the student achievement gaps and through what practices do they attempt to remedy the condition. The quantitative data resulted from the Quick Discrimination Index and relied on descriptive statistics. The primary audience for this study includes PK-12 public school administrators, particularly those in urban high-diversity, high-achieving schools.

Childhood depression is intimately related to the family context and caregiver-child relations, but only a limited number of treatment outcome studies for depressed youth have formally included members of the family in the intervention. To address this discrepancy the present study evaluated the efficacy of adding caregiver sessions to individual cognitive behavioral therapy (CBT) for the treatment of depressed youth. The treatment included 16 individual sessions and 7 caregiver sessions administered in the child's school to promote accessibility. This dissertation is a case study of an adult drug court in a medium-size Midwestern city. The primary impetus behind the creation of the drug court model was the partial recognition that the "get tough" approach to crime and the "war on drugs" was ineffective in "solving the United States' drug problem. Drug courts represent an integration of a public-health approach and a public-safety strategy of fighting crime and administering "justice." Polycyclic aromatic hydrocarbons (PAHs) are environmental and occupational carcinogens that are produced by the incomplete combustion of organic material, such as from the burning of tobacco, coal, and petroleum products. In addition to causing cancer, exposure to PAHs is hypothesized to contribute to atherosclerosis and to lead to increased incidence rates of cardiovascular disease in populations. Considering the number of deaths attributable to tobacco smoke exposure, ambient air pollution, and occupational hazards, PAHs may be a significant contributor to the prevalence of cardiovascular disease in human populations. Over the past thirty years a consensus has emerged that the word reading difficulties of dyslexic readers stem from deficits in phonological processing. One experimental paradigm that has provided support for this view is the finding that dyslexic readers demonstrate deficits in word retrieval from long term memory on picture naming tasks. Dyslexic readers are able to retrieve fewer words in their receptive vocabularies and are less accurate than normally developing readers. However, the conclusion that dyslexic readers? difficulties in picture naming are the consequence of deficits in phonological processing is inferential. This research investigated a proposed construct of learned technological helplessness (LTH) in female adult learners, which is believed to be impacted by socialization into traditional gender roles, teacher modeling, and age. A two-pronged approach was used to examine women?s attitudes and beliefs about their abilities to use technology as a learning and personal productivity tool. Participants were females, over age 18, on two campuses of a public Midwest research university. Understanding why some organizations are more effective in their service delivery than others is a central discourse in both public administration and political organization. Scholars and practitioners all agree that the defining factor is institutional (state) capacity. Ovarian hyperstimulation used in conjunction with embryo transfer techniques is an assisted reproductive technology that aids cattle producers in reaching their reproductive, genetic, and financial goals; however, viable embryo recovery results are inconsistent, possibly because the reproductive endocrine profile is poorly understood. The objective of these experiments was to evaluate the endocrinological and ovarian dynamics surrounding follicular deviation in hyperstimulated cows.
