Nicotine Exposure In The Developing Bullfrog: Influences On Neuroventilatory Responses To Co₂, Cord M. Brundage

Developmental exposure to the neuroteratogen nicotine may affect ventilatory responses to hypercapnia. Developmental changes in normocapnic and hypercapnic neuroventilation of the isolated bullfrog brainstem preparation have been previously characterized. I investigated the effect of 3- and 10-wk chronic nicotine (30 [mu]g/L) exposure on lung burst frequency exhibited by early and late metamorphic bullfrog tadpoles during normocapnia (1.5 % CO₂) and hypercapnia (5.0 % CO₂). Chronic nicotine exposure impairs the hypercapnic neuroventilatory response of early metamorphic tadpoles following both 3- and 10-wk exposure. Late metamorphic tadpoles demonstrated an impaired hypercapnic neuroventilatory response only after 10-wk exposure. Chronic nicotine exposure had no …

Oligobacterial physiology is mostly unstudied due to cultivation difficulty. New isolation techniques such as extinction culture have produced cultivable representatives of the aquatic environment namely Sphingopyxis alaskensis. Attempts were made to grow the bacterium in batch cultures using glucose and tyrosine as ideal substrates as determined from growth studies. Differential protein expression from cytoplasmic and membrane fractions of the putative culture were compared so as to identify key proteins involved in substrate uptake and metabolism followed by incorporation of protein quantities into mathematical models of oligotroph growth. Errors in growth and maturity estimates can drastically affect the spawner-per-recruit threshold used to recommend commercial fish catch quotas. Growth and maturity parameters for Alaskan sablefish, Anoplopoma fimbria, have not been updated for stock assessment purposes for 20 years, even though sablefish aging has continued. In this study, the old length-stratified data set (1981-1993) was updated and corrected for bias. In addition, newer, randomly collected samples (1996-2004) were analyzed, and new length-at-age, weight-at-age, and maturity-at-age and length parameters were estimated. This study examined the spatial components of inconnu Stenodus leucichthys spawning habitat use in the Selawik River, Alaska. Little is known about inconnu critical habitat needs; however, current studies of inconnu spawning behavior suggest a high level of habitat selectivity. This level of selectivity implies that there are specific habitat characteristics that these fish require for spawning. The purpose of this study was to build a heuristic habitat model that can be used to better understand inconnu spawning site selection in remote Alaskan watersheds. I developed a new technique, DEco-TILLING (an adaptation of Eco-TILLING), to discover useful single nucleotide polymorphisms (SNPs) rapidly and inexpensively. Some chum salmon (Oncorhynchus keta) populations have declined in Western Alaska and in other areas of their geographic range. Possible reasons include climate shifts, by- catch in fisheries, and other perturbations. Genetics offers powerful tools that can be used to monitor this species on the high seas in stock mixtures aiding management of by- catch identification and other contributors to declines. "Steller sea lions (Eumetopias jubatus) have experienced a drastic population decline in the past several decades. Among hypotheses for the decline and failure of the population to recover is decreased reproductive success. Harsh environmental conditions within the species range and the large body size of the animals can limit sampling efforts to investigate these hypotheses. Three captive Steller sea lions were used as models to validate the use of fecal steroid analysis for this species. Most of the current models to explain the diversification of Neotropical birds focus on physical barriers to gene flow. However, for any species the geographic structuring of populations is caused by an interaction between physical barriers to gene flow and a species' propensity to overcome those barriers. The three chapters presented in this dissertation provide three perspectives on this interaction and how it has shaped the diversification of some Neotropical birds. First, the widespread Neotropical lowland forest flycatcher Mionectes oleagineus had three phylogeographic splits across the Andes, resulting in four geographically structured lineages west of the Andes. The three chapters presented in this thesis use molecular markers to examine the evolutionary history of three groups of widespread Neotropical birds. In chapter one, I found that Amazilia tzacatl forms a monophyletic clade and exhibits four genetic clades: Atlantic and Pacific slopes of Middle America, South America, and Isla Coiba. The Escudo Hummingbird (A. t. handleyi) is probably not a full biological species. Specimens from the eastern Darien province of Panama suggest that individuals from Middle and South America colonized this area within the past 25 years. Antarctic icefish (suborder Notothenioidei, family Channichthyidae) do not express hemoglobin (Hb), and 6 of the 16 members of this family do not express myoglobin (Mb) in their cardiac muscle. The loss of Hb and Mb correlates with an increase in mitochondrial volume density, a decrease in the mitochondrial surface-to-volume ratio and a decrease in the surface density of inner mitochondria) membranes in the hearts of icefishes compared to red-blooded notothenioids. Lung Breathing In The Bullfrog: Generating Respiratory Rhythm And Pattern, Brittany L. Davies

This research investigated location of the lung respiratory rhythm generator (RRG) in the bullfrog brainstem using neurokinin-1 (NK1R) and [mu]opioid ([mu]OR) receptor colocalization and characterized the role of these receptors in breathing pattern formation. colocalization was distinct near the facial nucleus in juvenile bullfrogs but not in tadpoles. NK1R intensity exhibited no developmental change, while [mu]OR intensity increased from late-stage tadpoles to juvenile frogs. Substance P (NK1R agonist; bath applied) increased lung burst frequency, lung burst cycle frequency (BCF), episode frequency, lung burst amplitude and area, but decreased number of lung bursts per episode and lung burst duration. When attacked by predators, diverse animals actively or passively release molecules that evoke alarm and related anti-predatory behavior by nearby conspecifics. The actively released molecules are alarm pheromones, whereas the passively released molecules are alarm cues. For example, many insects have alarm-signaling systems that involve active release of alarm pheromones from specialized glands and detection of these signals using specific sensors. Many crustaceans passively release alarm cues, but the nature of the cues,sensors and responses is poorly characterized. Recent findings have implicated gp130 receptor ligands, particularly ciliary neurotrophic factor (CNTF), as potential antiobesity therapeutics. Neuropoietin (NP) is a recently discovered cytokine in the gp130 family that shares functional and structural features with CNTF and signals via the CNTF receptor tripartite complex comprised of CNTFRα, LIF receptor, and gp130. NP plays a role in the development of the nervous system, but the effects of NP on adipocytes have not been previously examined. Because CNTF exerts anti-obesogenic effects in adipocytes and NP shares the same receptor complex, we investigated the effects of NP on adipocyte development and insulin action. Using …

The aim of the present study was to determine the economic and social burden of compulsive hoarding in a large sample of individuals with self-identified hoarding, as well as a separate sample of family members of individuals who hoard. Self-identified hoarding participants (N = 864, 94% female, 65% met research criteria for clinically relevant compulsive hoarding) and family informants (N = 655, 58% described a relative who appeared to meet research criteria for compulsive hoarding), completed an internet survey. Questions were derived in part from those used in the National Comorbidity Survey (NCS), and when possible, hoarding participants were compared …

Incidence rates for adenocarcinomas of the esophagus and gastric cardia have been increasing rapidly, while rates for non‐cardia gastric adenocarcinoma and esophageal squamous cell carcinoma have declined. We examined food group intake as a risk factor for subtypes of esophageal and gastric cancers in a multicenter, population‐based case–control study in Connecticut, New Jersey and western Washington state. Associations between food groups and risk were estimated using adjusted odds ratios (OR), based on increasing intake of one serving per day. Total vegetable intake was associated with decreased risk of esophageal adenocarcinoma (OR = 0.85, 95% CI = 0.75, 0.96). Conversely, total …

This thesis incorporates three studies which explore adult and adolescent male sexual offenders. The first two articles examine the occurrence and effects of female perpetrated sexual abuse within a population of adolescent sexual offenders. The final article explores the occurrence of escalation in the offending histories of adult male sexual offenders. The first article examined the occurrence and characteristics of female perpetrated sexual abuse within a population of adolescent sexual offenders, as well as the arousal and victim chooses of the youth. This is an exploratory study of American veterans' decisions to join the military. Nine veterans from various branches of the military were interviewed regarding their decision process and what factors influenced their decision to join, including how they decided which branch to join, peoples' reactions to their decision, and how they reflect on their decision now. They were also asked their opinion on whether veterans today are receiving the services or acknowledgement that they think they should be getting. During the nonbreeding season, the Cape Romain Region of South Carolina supports ca. one-sixth of the total population of the eastern race (palliatus) of the American Oystercatcher (Haematopus palliatus), which consists of only ca. 11,000 individuals and appears to be declining. I compared the density, size, and orientation of the primary prey, Eastern oysters (Crassostrea virginica) and the foraging behaviors of adult American Oystercatchers among the three largest bays in the Cape Romain Region that American Oystercatchers used as foraging areas.
