Numerical Analysis Of A Fractional Step Theta-Method For Fluid Flow Problems, John Chrispell Aug 2008

Numerical Analysis Of A Fractional Step Theta-Method For Fluid Flow Problems, John Chrispell

All Dissertations

The accurate numerical approximation of viscoelastic fluid flow poses two difficulties: the large number of unknowns in the approximating algebraic system (corresponding to velocity, pressure, and stress), and the different mathematical types of the modeling equations. Specifically, the viscoelastic modeling equations have a hyperbolic constitutive equation coupled to a parabolic conservation of momentum equation. An appealing approximation approach is to use a fractional step $\theta$-method. The $\theta$-method is an operator splitting technique that may be used to decouple mathematical equations of different types as well as separate the updates of distinct modeling equation variables when modeling mixed systems of partial …

The Effects Of Indirubin Derivitives On Gene Expression And Cellular Functions In The Murine Macrophage, Abigail Babcock Aug 2008

The Effects Of Indirubin Derivitives On Gene Expression And Cellular Functions In The Murine Macrophage, Abigail Babcock

All Dissertations

Indirubin is the active ingredient in the ancient herbal remedy, Danggui Longhui Wan, which is used as an anti-leukemic, anti-inflammatory, and detoxification treatment. Indirubin-3'-monoxime (IO) is the most widely studied of the indirubin derivatives. Most have been shown to inhibit cyclin-dependent kinases and glycogen synthase kinases, triggering cell cycle arrest and apoptosis with different levels of activity depending on the particular structure. These kinases inhibited by indirubin(s) play key roles in cellular proliferation and immune functions, therefore synthetic indirubins may have significant applications as therapeutic agents for cancer, inflammation, and neurological diseases.
Indirubin is also a potent aryl hydrocarbon receptor …

Estimating Geocenter Variations From A Combination Of Grace And Ocean Model Output, Sean Swenson, D. Chambers, John Wahr

Marine Science Faculty Publications

In this study, we estimate a time series of geocenter anomalies from a combination of data from the Gravity Recovery and Climate Experiment (GRACE) satellite mission and the output from ocean models. A matrix equation is derived relating total geocenter variations to the GRACE coefficients of degrees two and higher and to the oceanic component of the degree one coefficients. We estimate the oceanic component from t wo state-of-the-art ocean models. Results are compared to independent estimates of geocenter derived from other satellite data, such as satellite laser ranging and GPS. Finally, we compute degree one coefficients that are consistent …

The Role Of Metal Coordnation In The Inhibition Of Iron(Ii)- And Copper(I)-Mediated Dna Damage By Organoselenium And Organosulfur Compounds, Erin Battin

All Dissertations

Copper and iron are two widely-studied transition metals associated with hydroxyl radical (OH) generation: Fe2+/Cu+ + H2O2 = Fe3+/Cu2+ + OH + OH-. Metal-mediated oxidative damage is a major cause of cellular DNA damage and death, and has been implicated in cancer, neurodegenerative, and cardiovascular diseases. Because sulfur and selenium antioxidants ameliorate oxidative damage, the ability of twenty biological sulfur- and selenium-containing compounds to inhibit metal-mediated DNA damage caused by hydroxyl radical has been quantified. In the Cu+/H2O2 system, six sulfur compounds and three selenium compounds inhibited DNA damage with IC50 values ranging from 3.3 to 25.5 μM. Several sulfur …

Engineered Micro-Environments And Vibrational Culture Systems For Vocal Fold Tissue Engineering, Jaishankar Kutty Aug 2008

Engineered Micro-Environments And Vibrational Culture Systems For Vocal Fold Tissue Engineering, Jaishankar Kutty

All Dissertations

Voice is produced by the conversion of aerodynamic energy from exhalation to acoustical energy for voice production by the vocal folds (membranous connective tissue) located in the larynx. The quality of voice depends on the biomechanical properties of the multi-layered vocal fold tissue which derive from its extracellular matrix (ECM) organization and composition. The wound healing response to vocal fold injuries is characterized by scarring and subsequent dysphonia due to alterations in the biomechanical properties of the tissue.
The work presented here is motivated by the importance of voice in maintaining quality of life and the inability of current treatment …

Effects Of Polyphenol Compounds On Iron-And Copper-Mediated Dna Damage: Mechanisms And Predictive Models, Nathan Perron Aug 2008

Effects Of Polyphenol Compounds On Iron-And Copper-Mediated Dna Damage: Mechanisms And Predictive Models, Nathan Perron

All Dissertations

Polyphenol compounds are widely recognized for their antioxidant activity; however, under some conditions these compounds can also exhibit prooxidant activity. This work examined the interactions of polyphenols with iron and copper to improve understanding of both their antioxidant and prooxidant effects. Polyphenols with catechol and gallol groups effectively inhibit iron-mediated oxidative DNA damage by chelating iron, with IC50 values ranging from 1-59 µM. Gallate compounds are more potent than their catecholate analogues in every instance, and their antioxidant potencies correlate to pKa values for the most acidic phenolic hydrogen. This correlation represents the first predictive model of …

Mothering The Mother : Can A Postpartum Doula Enhance Maternal Self Confidence And Maternal Empathy In A Primiparous Mother?, Binda Colebrook

Theses, Dissertations, and Projects

This study asks whether postpartum doulas enhance maternal empathy and maternal self-confidence in primiparous (first time) mothers in the postpartum phase and whether this is true across a socioeconomically diverse sample of women. A postpartum doula provides support to new mothers and their families in the first weeks postpartum. While quite a lot of research has been done on labor support doulas, less is known about the impact of the postpartum doula. No studies were found on whether they enhance maternal self-confidence and empathy. Because there are so few studies on postpartum doulas, an exploratory, qualitative method was used. In …

The Leaf Ionome As A Multivariable System To Detect A Plant's Physiological Status, Ivan R. Baxter, Olga Vitek, Brett Lahner, Balasubramaniam Muthukumar, Monica Borghi, Joe Morrissey, Mary Lou Guerinot, David E. Salt Aug 2008

The Leaf Ionome As A Multivariable System To Detect A Plant's Physiological Status, Ivan R. Baxter, Olga Vitek, Brett Lahner, Balasubramaniam Muthukumar, Monica Borghi, Joe Morrissey, Mary Lou Guerinot, David E. Salt

Dartmouth Scholarship

The contention that quantitative profiles of biomolecules contain information about the physiological state of the organism has motivated a variety of high-throughput molecular profiling experiments. However, unbiased discovery and validation of biomolecular signatures from these experiments remains a challenge. Here we show that the Arabidopsis thaliana (Arabidopsis) leaf ionome, or elemental composition, contains such signatures, and we establish statistical models that connect these multivariable signatures to defined physiological responses, such as iron (Fe) and phosphorus (P) homeostasis. Iron is essential for plant growth and development, but potentially toxic at elevated levels. Because of this, shoot Fe concentrations are …

Strategies For Expanding The Halogen Bonding Periodic Table And Desigining Complementary Halogen/Hydrogen Bonding Synthons, Hadi Arman

All Dissertations

Halogen bonding is the donation of electron density from an electron donor, such as N, O, S, Se atoms, to an electron acceptor such as a dihalogen or organohalogen. This interaction plays an important role in biological systems, such as the thyroid gland and is useful in many other fields such as drug design, crystal engineering, synthetic chemistry, material science and bioinorganic chemistry. However, currently there is a lack of information for this interaction in systems involving organoiodines with sulfur and selenium electron donors.
A variety of sulfur and selenium Lewis base donors were investigated with various organoiodines. The main …

Cues For Cellular Assembly Of Vascular Elastin Networks, Chandrasekhar Kothapalli Aug 2008

Cues For Cellular Assembly Of Vascular Elastin Networks, Chandrasekhar Kothapalli

All Dissertations

Elastin, a structural protein distributed in the extracellular matrix of vascular tissues is critical to the maintenance of vascular mechanics, besides regulation of cell-signaling pathways involved in injury response and morphogenesis. Thus, congenital absence or disease-mediated degradation of vascular elastin and its malformation within native vessels due to innately poor elastin synthesis by adult vascular cells compromise vascular homeostasis. Current elastin regenerative strategies using tissue engineering principles are limited by the progressive destabilization of tropoelastin mRNA expression in adult vascular cells and the unavailability of scaffolds that can provide cellular cues necessary to up-regulate elastin synthesis and regenerate faithful mimics …

All Dissertations

During the past two decades, advances in microelectromechanical systems (MEMS) have spurred efforts worldwide to develop sensing platforms based on smart microcantilevers. A microcantilever beam is one of the simplest MEMS structures which forms the basis for portable, fast and highly sensitive schemes that are capable of measuring small deflections in static or dynamic response due to changes in external parameters such as mass, pressure, charge, etc.
In this dissertation, I mainly focus on MEMS sensors with transducers in the form of microcantilevers. Variations in the microcantilever's response such as resonant frequency, amplitude, phase and quality factor when exposed to …

Synthesis And Pharmacology Of N-Alkyl-3-(Halo-Naphthoyl)Indoles, Valerie Smith Aug 2008

Synthesis And Pharmacology Of N-Alkyl-3-(Halo-Naphthoyl)Indoles, Valerie Smith

All Dissertations

The medicinal values of marijuana have often been overshadowed due to the psychotropic, 'recreational' effects it brings about in a user. For hundreds of years the therapeutic uses of cannabis sativa were appreciated, but the inability to control the dosage of the main constituent, and therefore the psychotropic effects, rendered the drug a liability. The elucidation of the structure of this main psychoactive constituent, ∆9-tetrahydrocannabinol (∆9-THC) by Gaoni and Mechoulam in 1964 opened the door to the discovery of a vast array of knowledge about cannabinoids. From the multiple classes of cannabinoids and the ways these compounds interact with the …

ABSTRACT

A series of bis(3,4-ethylenedioxythiophene)-thiophene derivatives (BEDOT-Ts) and bis(3,4-ethylenedioxythiophene)-benzene derivatives (BEDOT-Bs) monomers were electrochemically polymerized onto glassy carbon or ITO-coated glass electrodes. Cyclic voltammetry (CV), infrared (IR), UV-vis-NIR and photoluminescence (PL) spectroscopy were employed to investigate and characterize properties of resulting polymers. dc-conductivities of these polymers were measured by the four-point probe method.
Cyclic voltammetry studies show that the redox half potentials of PBEDOT-3BT, PBEDOT-3OT, PBEDOT-3TBS and PBEDOT-3MPT are -290, -297, -325 and -284 mV versus Ag/Ag+, respectively. The redox half potentials of PBEDOT-TOL, PBEDOT-BOB and PBEDOT-FB are -130, -147 and -327 mV vs. Ag/Ag+, respectively, and a bit higher …

Chitosan Derivatives For Tissue Engineering, Yongzhi Qiu Aug 2008

Chitosan Derivatives For Tissue Engineering, Yongzhi Qiu

All Dissertations

Chitosan, a naturally occurring polysaccharide, and its derivatives have been widely explored for biomedical applications due to their biocompatibility and biodegradability. In our studies, we developed a series of chitosan derivatives through chemical modifications. These chitosan derivatives not only possess better processibility in scaffolds fabrication, but also show excellent potentials in tissue engineering applications, including blood vessel and bone tissue engineering.
The excellent antithrombogenic property is crucial for vascular engineering applications, especially in engineering small-diameter blood vessels. In our studies, chitosan was chemically modified by phthalization and the phthalized chitosan exhibited great antithrombogenic property. Through a wet- phase-inversion process, tubular …

Excessive exercise may induce osteoarthritis (OA), or degeneration, of articular cartilage, which is a leading cause of lameness and decreased use in horses. The purposes of this study were to utilize a sheep model to determine the effects of circular and linear exercise on the macroscopic and microscopic characteristics of articular cartilage in the metacarpal-phalangeal (MCP) joint and to evaluate biochemical changes in serum and synovial fluid compared to non-exercised control lambs.
Twenty lambs were randomly assigned to three groups: circular exercise (C, 8.5m diameter; n = 8), straight-line exercise (S, treadmill; n = 8) and non-exercised control (CON, n …

The Spaceflight Environment And The Skeletal System, Eric Bandstra Aug 2008

The Spaceflight Environment And The Skeletal System, Eric Bandstra

All Dissertations

The spaceflight environment presents many challenges to the human body. Bone loss in astronauts is a well known consequence of reduced loading in the weightless environment of low-earth orbit. Recent studies have also indicated that spaceflight relevant types of radiation (at relatively high doses) have deleterious effects on trabecular bone. Both of these represent potential skeletal challenges during long-duration spaceflight. The objective of these studies is to examine the response of bone to models of the spaceflight environment.
Astronauts will likely absorb doses of protons and heavy ions during lengthy missions outside the Earth's magnetosphere. Following exposure to a range …

Changes In Brain Biochemistry And Feeding Behavior In Hybrid Striped Bass Exposed To Environmental Contaminants, Kristen Gaworecki Aug 2008

Changes In Brain Biochemistry And Feeding Behavior In Hybrid Striped Bass Exposed To Environmental Contaminants, Kristen Gaworecki

All Dissertations

Biochemical endpoints are useful when determining contaminant exposure in aquatic organisms. However, behaviors can also be impaired by environmental contaminants. Links between changes in brain biochemistry and behavior have been made, and understanding these relationships could increase the utility of these endpoints in ecological risk assessments. The overall goal of this research was to determine relationships between an ecologically relevant fish behavior and brain biochemistry. To accomplish this goal, I developed a behavioral bioassay that quantified the time it took exposed hybrid striped bass (Morone saxatilis x M. chrysops) to capture unexposed prey, fathead minnows (Pimephales promelas). Chemically targeted brain …

Activation Of Degp Chaperone-Protease Via Formation Of Large Cage-Like Oligomers Upon Binding To Substrate Proteins, Jiansen Jiang, Xuefeng Zhang, Yong Chen, Yi Wu, Z Hong Zhou, Zengyi Chang, Sen-Fang Sui Aug 2008

Activation Of Degp Chaperone-Protease Via Formation Of Large Cage-Like Oligomers Upon Binding To Substrate Proteins, Jiansen Jiang, Xuefeng Zhang, Yong Chen, Yi Wu, Z Hong Zhou, Zengyi Chang, Sen-Fang Sui

Faculty, Staff and Student Publications

Cells use molecular chaperones and proteases to implement the essential quality control mechanism of proteins. The DegP (HtrA) protein, essential for the survival of Escherichia coli cells at elevated temperatures with homologues found in almost all organisms uniquely has both functions. Here we report a mechanism for DegP to activate both functions via formation of large cage-like 12- and 24-mers after binding to substrate proteins. Cryo-electron microscopic and biochemical studies revealed that both oligomers are consistently assembled by blocks of DegP trimers, via pairwise PDZ1-PDZ2 interactions between neighboring trimers. Such interactions simultaneously eliminate the inhibitory effects of the PDZ2 domain. …

Drift By Drift: Effective Population Size Is Limited By Advection, John P. Wares, James M. Pringle Aug 2008

Drift By Drift: Effective Population Size Is Limited By Advection, John P. Wares, James M. Pringle

Earth Sciences

Background: Genetic estimates of effective population size often generate surprising results, including dramatically low ratios of effective population size to census size. This is particularly true for many marine species, and this effect has been associated with hypotheses of "sweepstakes" reproduction and selective hitchhiking. Results: Here we show that in advective environments such as oceans and rivers, the mean asymmetric transport of passively dispersed reproductive propagules will act to limit the effective population size in species with a drifting developmental stage. As advection increases, effective population size becomes decoupled from census size as the persistence of novel genetic lineages is …
