Dartmouth Scholarship

Understanding how nutrients affect gene expression will help us to understand the mechanisms controlling plant growth and development as a function of nutrient availability. Nitrate has been shown to serve as a signal for the control of gene expression in Arabidopsis. There is also evidence, on a gene-by-gene basis, that downstream products of nitrogen (N) assimilation such as glutamate (Glu) or glutamine (Gln) might serve as signals of organic N status that in turn regulate gene expression. To identify genome-wide responses to such organic N signals, Arabidopsis seedlings were transiently treated with ammonium nitrate in the presence or absence of …

Amphiod Densities And Indices Of Wetland Quality Across The Upper-Midwest, Usa, Michael J. Anteau, Alan D. Afton

United States Geological Survey, Northern Prairie Wildlife Research Center: Publications

Nutritional, behavioral, and diet data for lesser scaup (Aythya affinis [Eyton, 1838]) indicates that there has been a decrease in amphipod (Gammarus lacustris [G. O. Sars, 1863] and Hyalella azteca [Saussure, 1858]) density and wetland quality throughout the upper-Midwest, USA. Accordingly, we estimated densities of Gammarus and Hyalella in six eco-physiographic regions of Iowa, Minnesota, and North Dakota; 356 randomly selected semi-permanent and permanent wetlands were sampled during springs 2004 and 2005. We also examined indices of wetland quality (e.g., turbidity, fish communities, aquatic vegetation) among regions in a random subset of these wetlands (n = 267). Gammarus …

Species of Perkinsus are responsible for high mortalities of bivalve molluscs world-wide. Techniques to accurately estimate parasites in tissues are required to improve understanding of perkinsosis. This study quantifies the number and tissue distribution of Perkinsus marinus in Crassostrea virginica by modern stereology and immunohistochemistry. Mean total number of trophozoites were (mean ± SE) 11.80 ± 3.91 million and 11.55 ± 3.88 million for the optical disector and optical fractionator methods, respectively. The mean empirical error between both stereological approaches was 3.8 ± 1.0%. Trophozoites were detected intracellularly in the following tissues: intestine (30.1%), Leydig tissue (21.3%), hemocytes (14.9%), digestive …

Application Of A Geomorphic And Temporal Perspective To Wetland Management In North America, Loren M. Smith, Ned H. Euliss Jr., Douglas A. Wilcox, Mark M. Brinson

United States Geological Survey, Northern Prairie Wildlife Research Center: Publications

The failure of managed wetlands to provide a broad suite of ecosystem services (e.g., carbon storage, wildlife habitat, ground-water recharge, storm-water retention) valuable to society is primarily the result of a lack of consideration of ecosystem processes that maintain productive wetland ecosystems or physical and social forces that restrict a manager’s ability to apply actions that allow those processes to occur. Therefore, we outline a course of action that considers restoration of ecosystem processes in those systems where off-site land use or physical alterations restrict local management. Upon considering a wetland system, or examining a particular management regime, there are …

Genome-Wide Analysis Of Aux/Iaa And Arf Gene Families In Populus Trichocarpa, Udaya C. Kalluri, Stephen P. Difazio, Amy M. Brunner, Gerald A. Tuskan

Faculty & Staff Scholarship

Background

Auxin/Indole-3-Acetic Acid (Aux/IAA) and Auxin Response Factor (ARF) transcription factors are key regulators of auxin responses in plants. We identified the suites of genes in the two gene families in Populus and performed comparative genomic analysis with Arabidopsis and rice.

Results

A total of 35 Aux/IAA and 39 ARF genes were identified in the Populus genome. Comparative phylogenetic analysis revealed that several Aux/IAA and ARF subgroups have differentially expanded or contracted between the two dicotyledonous plants. Activator ARF genes were found to be two fold-overrepresented in the Populusgenome. PoptrIAA and PoptrARF gene families appear to have expanded due …

United States Public Health Resources

BACKGROUND. Gallbladder cancer (GBC) is rare; however, it disproportionately affects the American Indian and Alaska Natives (AI/AN) population. The purpose of the study was to characterize GBC among AI/AN in the US population.

METHODS. Cases of GBC diagnosed between 1999 and 2004 and collected by state-based cancer registries were included. Registry records were linked with Indian Health Service (IHS) administration records to decrease race misclassification of AI/AN. GBC rates and/or percent distributions for AI/AN and non-Hispanic whites (NHW) were calculated by sex, IHS region, age, and stage for all US counties and IHS Contract Health Service Delivery Area …

Conditional Protein Alleles And Regulation Of Bacterial Transcription: A Thesis In Two Acts, Edmund Ching Schwartz Jan 2008

Conditional Protein Alleles And Regulation Of Bacterial Transcription: A Thesis In Two Acts, Edmund Ching Schwartz

Student Theses and Dissertations

The use of small molecule modulators of protein function (drugs) has gained in popularity due to the speed and precision with which they can work. Unfortunately, a small molecule with the required specificity and potency is not always available. We have developed two methods to generate small molecule sensitive protein alleles without the need to screen for a new drug. In the first, which we term conditional protein splicing (CPS), an intein is split into two inactive fragments which are activated by heterodimerization. By fragmenting the target protein and fusing the pieces to this split intein, we were able to …

Biotinyl-Methyl 4-(Amidomethyl)Benzoate Is A Competitive Inhibitor Of Human Biotinidase, Keyna Kobza, Kittichai Chaiseeda, Gautam Sarath, James M. Takacs, Janos Zempleni

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Posttranslational modification of histones by biotinylation can be catalyzed by both biotinidase (BTD) and holocarboxylase synthetase. Biotinylation of histones is an important epigenetic mechanism to regulate gene expression, DNA repair, and chromatin remodeling. The role of BTD in histone biotinylation is somewhat ambiguous, given that BTD also catalyzes removal of the biotin tag from histones. Here, we sought to develop BTD inhibitors for future studies of the role of BTD in altering chromatin structure. We adopted an existing colorimetric BTD assay for use in a novel 96-well plate format to permit high-throughput screening of potential inhibitors. Biotin analogs were chemically …

Nazariyah Al-Ahliyah Dirasat Tahliliyah Muqaranah Bayna Al-Fiqh Wa-'Ilm Al-Nafs = Qualification Theory Analytical Comparative Study Between Islamic Jurisprudence And Developmental Psychology / Huda Mohammad Hassan Hilal, Huda Mohammad Hassan Hilal

Student Works (2000-2009)

This study attempts to formalize a theory for Qualification based on modern psychology science. Such theory resolves many of juristic disagreements and modern issues related to Qualification. This requires researching the gap between old transcripts on one hand and modern studies in Psychology and Islamic jurisprudence on the other hand. The study adopts an analytical inductive methodology through the analysis of juristic books, psychology literature, and uses deductive reasoning in stating logical findings. The study yields a number of results. Firstly, the gap in juristic literacy in Qualification studies appears in three aspects. The lack of reference to psychology which …

Negotiating For Nature: Conservation Diplomacy And The Convention On Nature Protection And Wildlife Preservation In The Western Hemisphere, 1929--1976, Keri Lewis

Doctoral Dissertations

In 1941, as the United States entered the Second World War, leaders from twenty American nations signed into effect a broad-based treaty for the protection of migratory wildlife at the Convention on Nature Protection and Wild Life Preservation in the Western Hemisphere. This dissertation examines the unique set of questions, problems, and concerns framers of the Convention dealt with in the development of a conservation program to ensure the protection of migratory wildlife as it crossed political borders. Although it provided no solid system of enforcement, the provisions of the Convention opened the door for new, more specific conservation treaties …
