The Drosophila melanogaster lymph gland is a haematopoietic organ1–3 in which pluripotent blood cell progenitors proliferate and mature into differentiated haemocytes. Previous work4 has defined three domains, the medullary zone, the cortical zone and the posterior signalling centre (PSC), within the developing third-instar lymph gland. The medullary zone is populated by a core of undifferentiated, slowly cycling progenitor cells, whereas mature haemocytes comprising plasmatocytes, crystal cells and lamellocytes are peripherally located in the cortical zone. The PSC comprises a third region that was first defined as a small group of cells expressing the Notch ligand Serrate5. Here we show that …

The Puerto Rican tree frog, Eleutherodactylus coqui, is a striking example of a deviation from the indirect mode of development among amphibians. E. coqui does not have an intermediate tadpole stage. It hatches out directly as a free-living froglet, deriving nutrition from the yolky material attached to its intestine. The innermost of the three germ layers, the endoderm, forms the lining of the gut. In the amphibian model Xenopus laevis, the endoderm originates from the vegetal hemisphere. The cells, which are originally present on the vegetal surface of the embryo, end up forming the definitive gut. They, thus, provide nutrition …

Strong evidence indicates that western juniper has significantly expanded its range since the late 1800s by encroaching into landscapes once dominated by shrubs and herbaceous vegetation (fig. 1). Woodland expansion affects soil resources, plant community structure and composition, water, nutrient and fire cycles, forage production, wildlife habitat, and biodiversity. Goals of juniper management include an attempt to restore ecosystem function and a more balanced plant community that includes shrubs, grasses, and forbs, and to increase ecosystem resilience to disturbances. Developing a management strategy can be a difficult task due to uncertainty about how vegetation, soils, hydrologic function, and wildlife will …

Glass fibers are among the most versatile industrial materials known today, where 4 billion pounds of glass are used every year. They are readily produced from raw materials, which are available in virtually unlimited supply. To compensate for its low tensile strength, glass is usually mixed with other materials such as nanofibers and nanospheres, to form what is called "glass nanocomposites". The mainstay of the blood coagulation cascade is the formation of the fibrin clot, catalyzed by the serine protease, thrombin. The prothrombinase complex catalyzes the activation of prothrombin to thrombin and, is composed of the enzyme, factor Xa, and the protein cofactor, factor Va, in the presence of divalent metal ions associated on a membrane surface. The enzyme, factor Xa, alone can activate prothrombin by two sequential proteolytic cleavages at Arg271 and Arg320 resulting in the intermediates, Fragment 1.2 and Prethrombin 2. The environmental movement relies on scientific claims to justify its calls for protectionist policies. These claims can be followed in the scientific literature using bibliometric methods such as citation analysis. Citation analysis was used to deconstruct the literature of endocrine disrupting chemical (EDC) sciences as it emerged and developed time from 1980 through 2004. This study explored how the attributes of scientific papers such as topic, journal, experimental model, document type, and support or negation of hypotheses impacted their influence (quantified as times cited) within the field over time. Temporal Activation Of The Jak-Stat Pathway In Relation To Cardiac Gene Expression In A Mouse Model Of Cardiac Dysfunction, Cassandra Talerico

Background: This project examined JAK-STAT pathway activation in two mouse models of cardiac hypertrophy: autoimmune myocarditis and pressure overload (PO). Methods: Myocarditis was induced with cardiac myosin PO was induced by transverse aortic constriction. STAT1, 3, and 5 binding was assessed by gel shift. STATs, JAKs, SERCA2A, and calsequestrin (CSQ) were quantified. In myocarditis, P-STAT3 localization to cardiac myocyte nuclei was ascertained, and plasma IL-6 and ventricular ANF mRNA were analyzed. Results: In myocarditis, STAT3/3 and STAT1/1 activation, inflammation, increased ventricle weight (P < 0.0001), and ANF mRNA (P = 0.005) occurred on days 14, 21, and 28. In PO, activation appeared on day 7 and persisted to failure. P-STAT3 increased (myocarditis, P < 0.0001 pressure overload, P < 0.05). P-JAK1 increased in myocarditis on days 21 and 28 (P < 0.007). In PO, CSQ and SERCA2A levels did not differ, but in myocarditis CSQ decreased (P = 0.02). In myocarditis, a biphasic elevation in plasma IL-6 occurred (P = 0.003). Conclusions: Although JAK-STAT signaling is activated in both models, it occurs earlier in PO and persists to heart failure, whereas in myocarditis it declines to basal levels as inflammation and plasma IL-6 return to baseline

Metallothioneins (MTs) are ubiquitous low molecular weight, cysteine rich proteins with a pronounced affinity for metal ions with d10 configuration such as Cu, Cd and Zn. These heavy metals form metal-thiolate clusters with cysteine side chains. In contrast to the vertebrate forms, knowledge about the properties of members of the plant metallothionein family is still scarce. We describe here a method of isolation and purification of metallothionein from the plant Helianthus annuus, performed by affinity chromatography using glutathione-agarose column. In this study, alignment of gene sequence of the isolated H. annuus metallothionein cDNA with known MTs showed that it belongs …

A lumped mathematical model of liver metabolism is presented to analyze the effect of ethanol on metabolic processes of 24 hr fasted rats. The model is developed in two parts. In the first part individual kinetic models for important regulatory steps in the liver metabolic pathways are developed and in second part transport and mass balance equations in the two well mixed domains: tissue and blood, are developed to calculate intermediate metabolite concentrations and fluxes in response to the changes in ethanol and lactate concentrations in the perfusion medium. Heart transplantation is a successful therapeutic procedure for the patients with end stage heart disease. Various preservative techniques have been developed to increase the shelf life of organs, but still there can be tissue damage during cold storage. One mechanism leading to damage is disruption of cell plasma membranes as they pass through temperature induced phase transitions. The addition of antifreeze protein (AFP) or glycoprotein (AFGP) to storage solutions has been hypothesized to reduce the leakiness of lipid membranes. External counterpulsation (ECP) has previously been used in treating cardiac patients. Compression of the lower extremities during diastole increases venous return and coronary perfusion. Purpose: To determine if ECP affects delayed onset muscle soreness (DOMS) and markers of muscle inflammation and skeletal muscle damage. Methods: Ten trained runners, 5 males and 5 females, aged 30.5 ł 12.8 years ran 20 miles at 70 of their VO2 max on a pre-determined course on two different occasions, once under control conditions and once under ECP treatment conditions. This report summarizes the activities of the NH Department of Environmental Services (NHDES) Shellfish Program for the period of January 2006 to December 2006, emphasizing those tasks for which NHDES received direct funding from the NH Estuaries Project. The NHDES Shellfish Program conducts a number of activities to minimize the health risks associated with consuming shellfish, and to continue to comply with National Shellfish Sanitation Program guidelines. These include water sampling on a prescheduled/randomized basis, as well as a pollution source identification and evaluation program.
