Γ-Tocotrienol Inhibits Nuclear Factor-Κb Signaling Pathway Through Inhibition Of Receptor-Interacting Protein And Tak1 Leading To Suppression Of Antiapoptotic Gene Products And Potentiation Of Apoptosis, Seok Ahn Kwang, Gautam Sethi, Koyamangalath Krishnan, Bharat B. Aggarwal

ETSU Faculty Works

Unlike the tocopherols, the tocotrienols, also members of the vitamin E family, have an unsaturated isoprenoid side chain. In contrast to extensive studies on tocopherol, very little is known about tocotrienol. Because the nuclear factor-κB (NF-κB) pathway has a central role in tumorigenesis, we investigated the effect of γ-tocotrienol on the NF-κB pathway. Although γ-tocotrienol completely abolished tumor necrosis factor α (TNF)-induced NF-κB activation, a similar dose of γ-tocopherol had no effect. Besides TNF, γ-tocotrienol also abolished NF-κB activation induced by phorbol myristate acetate, okadaic acid, lipopolysaccharide, cigarette smoke, interleukin-1β, and epidermal growth factor. Constitutive NF-κB activation expressed by certain …

A Bayesian Hierarchical Model For Constrained Distributed Lag Functions: Estimating The Time Course Of Hospitalization Associated With Air Pollution Exposure, Roger Peng, Francesca Dominici, Leah J. Welty Jan 2007

A Bayesian Hierarchical Model For Constrained Distributed Lag Functions: Estimating The Time Course Of Hospitalization Associated With Air Pollution Exposure, Roger Peng, Francesca Dominici, Leah J. Welty

Johns Hopkins University, Dept. of Biostatistics Working Papers

Numerous time series studies have provided strong evidence of an association between increased levels of ambient air pollution and increased levels of hospital admissions, typically at 0, 1, or 2 days after an air pollution episode. An important research aim is to extend existing statistical models so that a more detailed understanding of the time course of hospitalization after exposure to air pollution can be obtained. Information about this time course, combined with prior knowledge about biological mechanisms, could provide the basis for hypotheses concerning the mechanism by which air pollution causes disease. Previous studies have identified two important methodological …

High-Frequency Ultrasound Drug Delivery And Cavitation, Mario Alfonso Diaz Jan 2007

High-Frequency Ultrasound Drug Delivery And Cavitation, Mario Alfonso Diaz

Theses and Dissertations

The viability of a drug delivery system which encapsulates chemotherapeutic drugs (Doxorubicin) in the hydrophobic core of polymeric micelles and triggers release by ultrasound application was investigated at an applied frequency of 500 kHz. The investigation also included elucidating the mechanism of drug release at 70 kHz, a frequency which had previously been shown to induce drug release. A fluorescence detection chamber was used to measure in vitro drug release from both Pluronic and stabilized micelles and a hydrophone was used to monitor bubble activity during the experiments. A threshold for release between 0.35 and 0.40 in mechanical index was …

The Effects Of Novel Serms On Endothelial And Epithelial Tumor Cell Estrogen Receptor Activation, Palvos Zacharias Kaimakliotis Jan 2007

The Effects Of Novel Serms On Endothelial And Epithelial Tumor Cell Estrogen Receptor Activation, Palvos Zacharias Kaimakliotis

Yale Medicine Thesis Digital Library

Premenopausal women are relatively protected against the development of coronary heart disease, when compared with age-matched men. This difference dissolves after menopause and is accepted to be hormonally mediated. Despite recent clinical controversies in the use of hormone replacement therapy in postmenopausal women, there have been numerous in vitro and in vivo studies that demonstrate favorable effects of estrogen on the endothelium. The Bender laboratory has shown that 17β-estradiol (E2) rapidly induces nitric oxide (NO) release from human endothelial cells (EC) in vitro. This occurs through a c-Src/phosphatidylinositol 3-kinase (PI3-kinase)/Akt pathway, in a rapid fashion in the absence of modulated …

Glycaemic Index, Glycaemic Load And Risk Of Endometrial Cancer: A Prospective Cohort Study, Stephanie A. Navarro Silvera, Thomas E. Rohan, Meera Jain, Paul D. Terry, Geoffrey R. Howe, Anthony B. Miller

Department of Public Health Scholarship and Creative Works

Objective

High-glycaemic-load diets may increase endometrial cancer risk by increasing circulating insulin levels and, as a consequence, circulating oestrogen levels. Given the paucity of epidemiological data regarding the relationship between the dietary glycaemic index and glycaemic load and endometrial cancer risk, we sought to examine these associations using data from a prospective cohort study.

Design, setting, and subjects

We examined the association between dietary glycaemic load and endometrial cancer risk in a cohort of 49 613 Canadian women aged between 40 and 59 years at baseline who completed self-administered food-frequency questionnaires between 1982 and 1985. Linkages to national mortality and …

Risk And Protective Factors For Children Of Adolescents: Maternal Depression And Parental Sense Of Competence, Lisa Knoche, Jami E. Givens, Susan M. Sheridan Jan 2007

Risk And Protective Factors For Children Of Adolescents: Maternal Depression And Parental Sense Of Competence, Lisa Knoche, Jami E. Givens, Susan M. Sheridan

Department of Educational Psychology: Faculty Publications

We investigated the relationship between depression and parental sense of competence to child cognitive outcomes for a sample of 49 adolescent mothers and their young children (Mean age = 9 1/2 months) enrolled in a student parenting program. Cognitive development of the infants and toddlers was assessed using the Bayley Scales of Infant Development. Maternal depression was assessed with the CES-D and parental competence measured with the Parental Sense of Competence Scale. Results indicated that maternal depression and parental sense of competence alone did not predict children’s cognitive scores; the interaction of the variables significantly predicted children’s outcomes. Mothers reporting …

Animals were treated with GnRH agonist, GnRH antagonist, or vehicle (Control) for 28 days. Serial blood samples were collected on day 21 of treatment, and at several intervals after treatment. Injections of natural sequence GnRH were used to evaluate the capacity of the pituitary to release gonadotropins during and after treatment. Treatment with GnRH agonist increased basal LH and T concentrations in both rams and bulls, with a greater relative increase …

Identification Of Genes Encoding Acyl-Coa Reductases And Aldehyde Reductases In Mycobacterial Genome By Characterization Of The Reductases Expressed In E. Coli, Harminder Singh Jan 2007

Identification Of Genes Encoding Acyl-Coa Reductases And Aldehyde Reductases In Mycobacterial Genome By Characterization Of The Reductases Expressed In E. Coli, Harminder Singh

Electronic Theses and Dissertations

Mycobacterium tuberculosis has been long known to produce wax esters. However, the enzymes involved in their biosynthesis have not been identified. Here we report the identification of Rv3391 and Rv1543 as genes that encode fatty acyl-CoA reductases and Rv1544 as one that encodes an aldehyde reductase. When expressed in E.coli, the products of Rv3391 and Rv1543 catalyzed reduction of fatty acyl-CoA to fatty alcohol with the corresponding aldehyde as an intermediate with an optimal pH of 7.0. Both enzymes showed a strong preference for NADPH over NADH as a reductant. Apparent Km for NADPH was 38 [micro]M for Rv3391 product …

Social Pathogenic Sources Of Poor Community Health, Hayden Smith Jan 2007

Social Pathogenic Sources Of Poor Community Health, Hayden Smith

Electronic Theses and Dissertations

The United States currently provides a health care system that is neither efficient nor equitable. Despite outspending the world on health care, over three-fourths of developed countries produce better health outcomes (Auerbach et al., 2000). Simultaneously, the "Ecological School of Thought" has documented the large impact that social, economic, and environmental circumstances play in health outcomes. Unfortunately, these 'ecological" studies are frequently conducted without theoretical justification, and rely solely on a cross-sectional research design and a myriad of unrelated variables. This study represents an important step towards the development of a true theory of "ecology". More specifically, we argue that …

Correlation Of Rpob Gene Mutation With Clinical Rifabutin And Rifampicin Resistance For Treatment Of Crohn's Disease, Daniel Beckler Jan 2007

Correlation Of Rpob Gene Mutation With Clinical Rifabutin And Rifampicin Resistance For Treatment Of Crohn's Disease, Daniel Beckler

Electronic Theses and Dissertations

Emerging rise in microbial drug resistance and the slow-growing characteristic of some intracellular pathogens such as MAP (Mycobacterium avium subspecies paratuberculosis) strongly urges the need for an effective approach for unconventional drug susceptibility testing. We designed a molecular-based PCR method for the evaluation of rifabutin (RFB) and rifampicin (RIF) resistance based on probable determinant regions within the rpoB gene of MAP, including the 81 bp variable site located between nucleotides 1363 and 1443. The minimum inhibitory concentration (MIC) for RIF was also determined against 10 MAP isolates in attempt to seek correlation with rpoB sequences. We determined that MAP strain …

Mechanism Of Action And Regulation Of Membrane Serine Protease Prostasin In The Prostate And Prostate Cancer, Mengqian Chen Jan 2007

Mechanism Of Action And Regulation Of Membrane Serine Protease Prostasin In The Prostate And Prostate Cancer, Mengqian Chen

Electronic Theses and Dissertations

The glycosylphosphatidylinositol (GPI)-anchored serine protease prostasin (PRSS8) is expressed at the apical membrane surface of epithelial cells and acts as a suppressor of tumor invasion when re-expressed in highly invasive human prostate and breast cancer cell lines. To better understand the molecular mechanisms underlying the anti-invasion phenotype associated with prostasin re-expression in prostate cancer cells, we expressed wild-type human prostasin or a serine active-site mutant prostasin in the PC-3 human prostate carcinoma cells. Molecular changes were measured at the mRNA and the protein levels. The expression of several invasion-promoting molecules is regulated by prostasin re-expression, mediated by a protein-level down-regulation …

A Member Of The Novel Fikk Family Of Plasmodium Falciparum Putative Protein Kinases Exhibits Diacylglycerol Kinase Activity And Is Exported To The Host Erythrocyte, David Floyd Curtis

Electronic Theses and Dissertations

Plasmodium falciparum is one of four species known to cause malaria in humans and is the species that is associated with the most virulent form of the disease. Malaria causes nearly two million deaths each year, many of these occurring among children in under-developed countries of the world. One reason for this is the prevalence of drug resistant strains of malaria that mitigate the efficacy of existing drugs. Hence, the identification of a new generation of pharmacological agents for malaria is extremely urgent. The recent identification of a group of novel protein kinases within the Plasmodium falciparum genome has provided …
