Panulirus argus Virus 1 (PaV1) is a pathogenic virus that infects Caribbean spiny lobsters P. argus in the Florida Keys. We have developed a PCR detection assay for PaV1 for the purpose of studying the natural history of the virus and for monitoring the prevalence of infection. The detection of the virus in hemolymph and other tissues is based on the PCR amplification of a 499 bp product using specific primers designed from a cloned fragment of the PaV1 genome. The sensitivity limit for the assay was 1.2 fg of purified viral DNA. The PaV1 primers did not react with …

An Avidin-Based Assay For Histone Debiotinylase Activity In Human Cell Nuclei, Yap Ching Chew, Gautam Sarath, Janos Zempleni

The covalent binding of biotin to histones participates in heterochromatin formation, cell cycle progression and cellular response to DNA breaks. Biotinylation of histones appears to be a reversible process, but the identity of enzymes that remove biotin marks is largely unknown. Our long-term goal is to identify histone debiotinylases in human cells. Here we developed an avidin-based plate assay to quantify histone debiotinylase activities in nuclear extracts. This assay is an essential first step in purifying and identifying histone debiotinylases from human cells. Description Of Harmonic Generation In Terms Of The Complex Quasienergy. I. General Formulation, M.V. Frolov, A.V. Flegel, N. L. Manakov, Anthony F. Starace

Using the Hellmann-Feynman theorem for quasistationary quasienergy states of a quantum system in an intense laser field, we present the high-order harmonic generation (HHG) amplitude in terms of the complex quasienergy of a bound electron, thereby avoiding the necessity for an explicit form for the electron wave function in HHG calculations. This formulation for the HHG amplitude confirms use of the dual dipole moment (instead of the dipole moment expectation value) in wave-function-based HHG calculations in order to properly account for both the ionization and the Stark shift of the initial bound state in a strong laser field.

Nitric oxide is emerging as an important modulator of many physiological processes including olfaction, yet the function of this gas in the processing of olfactory information remains poorly understood. In the antennal lobe of the moth, Manduca sexta, nitric oxide is produced in response to odor stimulation, and many interneurons express soluble guanylyl cyclase, a well-characterized nitric oxide target. We used intracellular recording and staining coupled with pharmacological manipulation of nitric oxide and soluble guanylyl cyclase to test the hypothesis that nitric oxide modulates odor responsiveness in olfactory interneurons through soluble guanylyl cyclase-dependent pathways. Acceptance and commitment therapy (ACT) has previously been shown to alter stigmatizing attitudes and to be relatively useful for psychologically inflexible participants. The present study is the first to bring those two findings together by comparing ACT to an education intervention for reducing stigma toward people with psychological disorders, and examining whether results differ for psychologically inflexible versus flexible individuals. A sample of college students (N=95) was randomly assigned to a 21/2 h ACT or educational workshop. Measures were taken before and after the workshop and at a 1-month follow-up. The authors report experiments with a diffraction-based biosensor based on self-assembly of target-containing nanobeads that form optical diffraction gratings. They demonstrate that the diffraction signal is a function of the bead size, and that noise is minimized by normalizing the intensities of the diffraction modes. They characterize the dependence of the diffraction signal on equivalent bead size and demonstrate the potential of the scheme in detecting biologically significant molecules.

This study examined competition effects on ADG in the feedlot of 1,882 Hereford bulls representing 8 birth years from a selection experiment. Each year, 8 feedlot pens were used to feed bulls in groups, with 2 pens nested within each of the 4 selection lines. Gains were recorded for up to 8 periods of 28 d. Models for analyses included pen effects (fixed or random), fixed effects such as year and line, and random direct genetic, competition genetic (and in some analyses competition environmental), and environmental effects. The conformational change of a host protein, PrPC, into a disease-associated isoform, PrPSc, appears to play a critical role in the pathogenesis of prion diseases such as Creutzfeldt–Jakob disease and scrapie. However, the fundamental mechanism by which infectious prions are produced in neurons remains unknown. To investigate the mechanism of prion formation biochemically, we conducted a series of experiments using the protein misfolding cyclic amplification (PMCA) technique with a preparation containing only native PrPC and copurified lipid molecules. These experiments showed that successful PMCA propagation of PrPScmolecules in a purified system requires accessory polyanion molecules. In addition, we found that …
