Prostaglandins are anticancer agents known to inhibit tumor cell proliferation both in vitro and in vivo by affecting the mRNA stability. Here we report that a MAR-binding protein SMAR1 is a target of Prostaglandin A2 (PGA2) induced growth arrest. We identify a regulatory mechanism leading to stabilization of SMAR1 transcript. Our results show that a minor stem and loop structure present in the 5' UTR of SMAR1 (1-UTR) is critical for nucleoprotein complex formation that leads to SMAR1 stabilization in response to PGA2. This results in an increased SMAR1 transcript and altered protein levels, that in turn causes downregulation of …

OxyR is a conserved bacterial transcription factor with a regulatory role in oxidative stress response. From a genetic screen for genes that modulate biofilm formation in the opportunistic pathogen Serratia marcescens, mutations in an oxyR homolog and predicted fimbria structural genes were identified. S. marcescens oxyR mutants were severely impaired in biofilm formation, in contrast to the hyperbiofilm phenotype exhibited by oxyR mutants of Escherichia coli and Burkholderia pseudomallei. Further analysis revealed that OxyR plays a role in the primary attachment of cells to a surface. Similar to what is observed in other bacterial species, S. marcescens OxyR …

This dissertation describes the conceptual and empirical framework that guides the definition of food insecurity and hunger, the present status of federal programmatic responses, and the status of current research on the topic. It also examines relations between hunger and federal food assistance program participation. Logistic regression is utilized to build two predictor models. Model 1 predicts federal food assistance program participation from household structure, income, community characteristics, and demographics. Model 2 predicts food insecurity/hunger from household structure, income, community characteristics, demographics, and federal food assistance program participation. Physiological compliance (PC) refers to the correlation between physiological measures of team members over time. The first goal of the current analyses was to generate several means to measure PC from heart rate variability (HRV) data. A second goal was to examine the relationship between PC and team performance during a building clearing task performed by 4-man teams. Teams were tasked with entering and clearing both real and simulated rooms populated with combatants (individuals with a weapon) and non-combatants (individuals without a weapon). In order for water resource managers to effectively and efficiently react to algal growths that are prohibiting use of a lake, reservoir or stream, information must be obtained on the response of the specific algae in site waters to algaecide exposures. In the absence of this information, ineffective algaecides or excessive treatments may be implemented. Research on development of an efficient algaecide assay using site specific samples could contribute to better decisions regarding algaecide applications and increase margins of safety for non-target species. The use of classical and quantum mechanics has been employed in this dissertation to study temperature control algorithms and a hydrated electron. A comparison of the effects of four commonly used temperature control algorithms on the energetics and dynamics of liquid water has been executed, in efforts to better understand the non-equipartitioning effects caused by some thermostats. The Berendsen, Andersen, Langevin, and velocity rescaling temperature control algorithms were applied to both dimer and bulk water systems, using the TIP4P water model. Lack of complete chloroplast genome sequences is still a limiting factor determining phylogenetic relationships, discerning evolutionary forces, and extending chloroplast genetic engineering to useful crops. Therefore, the chloroplast genomes from six economically important crops were isolated and sequenced. The results will have an impact on chloroplast biology and biotechnology.
The complete soybean chloroplast genome was compared to the other completely sequenced legumes, Lotus and Medicago. The rpl22 gene was found to be missing from all three legumes, a very informative phylogenetic marker. The current trends in molecular phylogenetics are towards assembling large data matrices from many independent loci and employing realistic probabilistic models. Large genome-scale data sets shall reduce the sampling error, whereas complex models accommodating heterogeneity among sites and along the phylogenetic tree can decrease systematic errors. The theme of this dissertation project is using both bioinformatic and experimental approaches to develop genome-scale nuclear gene markers and applying them in studies of phylogeny of ray-finned fish (Actinopterygii) and systematics of clupeiforms. Methods and procedures for modeling university student populations, predicting course enrollment, allocating course seats, and timetabling final examinations are studied and proposed. The university enrollment model presented uses a multi-dimensional state space based on student demographics and the Markov property, rather than longitudinal data to model student movement. The procedure for creating adaptive course prediction models uses student characteristics to identify groups of undergraduates whose specific course enrollment rates are significantly different than the rest of the university population. Historical enrollment rates and current semester information complete the model for predicting enrollment for the coming semester. The course prediction model …

The objectives of this project are 1) the setup and refinement of peptide-SAM surface model systems with explicit water using CHARMM and 2) the development of up-to-date simulation protocols for the accurate calculation of adsorption free energy by incorporating the recent development of non-Boltzmann sampling methods in molecular dynamics and applying it for the calculation of adsorption free energy of short peptides onto well-characterized self-assembled monolayers, which is important for understanding protein/surface interactions.

This dissertation describes the design and implementation of various nonlinear control strategies for robot manipulators whose dynamic or kinematic models are uncertain. Chapter 2 describes the development of an adaptive task-space tracking controller for robot manipulators with uncertainty in the kinematic and dynamic models. The controller is developed based on the unit quaternion representation so that singularities associated with the otherwise commonly used three parameter representations are avoided. Experimental results for a planar application of the Barrett whole arm manipulator (WAM) are provided to illustrate the performance of the developed adaptive controller.

In this work computational approaches to the numerical simulation of steady-state viscoelastic fluid flow are investigated. In particular, two aspects of computing viscoelastic flows are of interest: 1) the stable computation of high Weissenberg number Johnson-Segalman fluids and 2) low-order approaches to simulating the flow of fluids obeying a power law constitutive model.
The numerical simulation of viscoelastic fluid flow becomes more difficult as a physical parameter, the Weissenberg number, increases. Specifically, at a Weissenberg number larger than a critical value, the iterative nonlinear solver fails to converge. For the nonlinear Johnson-Segalman constitutive model, defect-correction and continuation methods are examined …

Biodiesel synthesis from biomass provides a means for utilizing effectively renewable resources, a way to convert waste vegetable oils and animal fats to a useful product, a way to recycle carbon dioxide for a combustion fuel, and production of a fuel that is biodegradable, non-toxic, and has a lower emission profile than petroleum-diesel. Microcantilevers are one the most commonly utilized microelectromechanical systems in a variant of the ultra-precise scanning and characterization application. Of particular interest, the problem of vibrations of microcantilevers has recently received considerable attention due to its application in several nanotechnological instruments such as atomic force microscopy, nanomechanical cantilever sensors and friction force microscopy. For applications such as these, the problem of coupled flexural-torsional nonlinear vibrations of a piezoelectrically-actuated microcantilever beam is considered in this dissertation. In recent years, there has been immense interest in the utilization of photopolymerized hydrogels as carriers for controlled protein delivery and cell scaffolds for tissue engineering applications. Although poly(ethylene glycol) (PEG)-based hydrogels formed from mild photopolymerization methods have been suggested as biocompatible matrices that allow for safely encapsulating biomolecules including proteins, peptides, DNA, and cells, the adverse effects of photopolymerization reactions on the encapsulated proteins have largely been overlooked. In addition, conventional hydrophilic hydrogels fail to effectively control protein delivery rates due to their high permeability. The prenatal environment has been shown to have lasting effects on cardiovascular health. In the present study, pregnant rats were fed a 0.7% NaCl normal salt (NS) diet or an 8% NaCl high salt (HS) diet throughout pregnancy. Adult offspring were fitted with radiotelemetry probes to continuously measure blood pressure and heart rate. Rats were placed in restraining cages to test for a programmed acute stress hyperresponsiveness. Offspring were challenged with HS diet for one week to determine if blood pressure salt sensitivity had been programmed by the prenatal HS diet. Many studies have documented the ways in which shyness can be a barrier to personal well-being and social adjustment throughout childhood and adolescence; however, less is known regarding shyness in emerging adulthood. Shyness as experienced during emerging adulthood may continue to be a risk factor for successful development. The purpose of this study was to compare shy emerging adults with their non-shy peers in (a) internalizing behaviors, (b) externalizing behaviors, and (c) close relationships. Professional and scientific psychology appears to have rediscovered spirituality and religion during recent years, with a large number of conferences, seminars, workshops, books, and special issues in major professional journals on spirituality and psychology integration. The purpose of this commentary is to highlight some of the more compelling ethical principles and issues to consider in spirituality and psychology integration with a focus on psychotherapy. Follicular dendritic cells (FDCs) reside in the lymphoid follicles of the secondary lymphoid tissues (sLTs). Following the infection of an individual with human immunodeficiency virus type 1 (HIV-1), viral particles are trapped in massive quantities on the surfaces of FDCs. HIV-1 viral compartments are cell types or tissues between which there is a restriction of virus flow. Compartmentalization of HIV-1 creates numerous sites within the body in which the virus can undergo independent evolution, giving rise to a more diverse total viral population.
