Microvascular Endothelial Cells Express A Phosphatidylserine Receptor: A Functionally Active Receptor For Phosphatidylserine-Positive Erythrocytes, Yamaja Setty, Suhita Gayen Betal

Phosphatidylserine (PS)-positive erythrocytes adhere to endothelium and subendothelial matrix components. While thrombospondin mediates these inter-actions, it is unknown whether PS-associated erythrocyte-endothelial adhesion occurs in the absence of plasma ligands. Using ionophore-treated PS-expressing control HbAA erythrocytes, we demonstrate that PS-positive erythrocytes adhered to human lung microendothelial cells in the absence of plasma ligands, that this adhesion was enhanced following endothelial activation with IL-1alpha, TNF-alpha, LPS, hypoxia, and heme, and that this adhesive interaction was selective to erythrocyte PS. We next explored whether microendothelial cells express an adhesion receptor that recognizes cell surface-expressed PS (PSR) similar to that expressed on activated macrophages. …

This paper describes a new national learning center designed to improve the delivery of science-based information to non-research customers. The vision of the Livestock and Poultry Environmental Learning Center is to provide individuals involved in public policy issues, animal production, and delivery of technical services for confined animal systems with on-demand access to the nation’s best science-based resources that is responsive to priority and emerging environmental issues associated with animal agriculture. Multiple assessments of ambient odor were made by trained individuals in the vicinity of a swine finishing operation in eastern Nebraska during the summers of 2005 and 2006. This paper addresses an analysis of assessor responses in Year 1 of this field study to determine what relationships existed between field odor measurements/ratings and ratings of annoyance potential, and to identify candidate measurement threshold values for odors that are likely to cause an annoyance. The first-year results showed that the likelihood of odor causing annoyance increased as ambient odors became more offensive, more intense, and more concentrated, with r2 values …

The introduction of ASABE Standard D384.2, Manure Production and Characteristics, has created the opportunity to integrate feed management decisions and animal performance measures into nutrient planning processes. This paper introduces a software tool that integrates estimates of nutrient excretion based on the new standard with estimates of land need, labor and equipment time allocation, and economic cost and benefit for manure application. This tool will be used to evaluate the economic implications for two scenarios using beef cattle examples as a means of illustrating tool application. Obesity and diabetes are associated with increased fatty acid availability in excess of muscle fatty acid oxidation capacity. This mismatch is implicated in the pathogenesis of cardiac contractile dysfunction and also in the development of skeletal-muscle insulin resistance. We tested the hypothesis that 'Western' and high fat diets differentially cause maladaptation of cardiac- and skeletal-muscle fatty acid oxidation, resulting in cardiac contractile dysfunction. Wistar rats were fed on low fat, 'Western' or high fat (10, 45 or 60% calories from fat respectively) diet for acute (1 day to 1 week), short (4-8 weeks), intermediate (16-24 weeks) or long (32-48 weeks) …

Environmental health professionals are concerned about bites. Animal bites, mosquito bites, and tick bites to name a few. But what about bytes of data? Environmental health information systems (EHIS) are often the "silent partners" in an environmental health (EH) department's daily protection of public health. By Webster's dictionary definition, a silent partner is a cohort that does not have the right to participate in an organization's management process. The Rhinopomatidae, traditionally considered to be one of the most ancient chiropteran clades, remains one of the least known groups of Rhinolophoidea. No relevant fossil record is available for this family. Whereas there have been extensive radiations in related families Rhinolophidae and Hipposideridae, there are only a few species in the Rhinopomatidae and their phylogenetic relationship and status are not fully understood. Background and Purpose: Databases have led to considerable advancements in the fields of biology and astrophysics. Functional Magnetic Resonance Imaging (fMRI) studies are expensive and time consuming to complete. An fMRI database would maximize knowledge gained from any one individual study. Methods: I searched PubMED and the fmridc.org website. I then searched PubMED using single citation manager to find an example of a study that used this database to create a new scientific investigation. Results: The fMRI Data Center was established in the autumn of 1999 to provide a mechanism for sharing fMRI data. Several investigators have used this database …

Elevated systemic levels of the acute phase C-reactive protein (CRP) are predictors of future cardiovascular events. There is evidence that CRP may also play a direct role in atherogenesis. Here we determined whether the proinflammatory interleukin (IL)-17 stimulates CRP expression in hepatocytes (Hep3B cell line and primary hepatocytes) and coronary artery smooth muscle cells (CASMC). Our results demonstrate that IL-17 potently induces CRP expression in Hep3B cells independent of IL-1β and IL-6. IL-17 induced CRP promoter-driven reporter gene activity that could be attenuated by dominant negative IκBα or C/EBPβ knockdown and stimulated both NF-κB and C/EBP DNA binding and reporter …

The Ctr1 family of integral membrane proteins is necessary for high affinity copper uptake in eukaryotes. Ctr1 is also involved in cellular accumulation of cisplatin, a platinum-based anticancer drug. Although the physiological role of Ctr1 has been revealed, the mechanism of action of Ctr1 remains to be elucidated. To gain a better understanding of Ctr1-mediated copper and cisplatin transport, we have monitored molecular dynamics and transport activities of yeast Saccharomyces cerevisiae Ctr1 and its mutant alleles. Co-expression of functional Ctr1 monomers fused with either cyan or yellow fluorescent protein resulted in fluorescence resonance energy transfer (FRET), which is consistent with …

Search Techniques For Multi-Objective Optimization Of Mixed Variable Systems Having Stochastic Responses, Jennifer G. Walston

A research approach is presented for solving stochastic, multi-objective optimization problems. First, the class of mesh adaptive direct search (MADS) algorithms for nonlinearly constrained optimization is extended to mixed variable problems. The resulting algorithm, MV-MADS, is then extended to stochastic problems (MVMADS-RS), via a ranking and selection procedure. Finally, a two-stage method is developed that combines the generalized pattern search/ranking and selection (MGPS-RS) algorithms for single-objective, mixed variable, stochastic problems with a multi-objective approach that makes use of interactive techniques for the specification of aspiration and reservation levels, scalarization functions, and multi-objective ranking and selection. Cellular expression of Mcl-1, an anti-apoptotic Bcl-2 family member, is tightly regulated. Recently, Bcl-2 expression was shown to be regulated by microRNAs, small endogenous RNA molecules that regulate protein expression through sequence-specific interaction with messenger RNA. By analogy, we reasoned that Mcl-1 expression may also be regulated by microRNAs. We chose human immortalized, but non-malignant, H69 cholangiocyte and malignant KMCH cholangiocarcinoma cell lines for these studies, because Mcl-1 is dysregulated in cells with the malignant phenotype. By in silico analysis, we identified a putative target site in the Mcl-1 mRNA for the mir-29 family, and found that mir-29b was highly …
