A pivotal mediator of actin dynamics is the protein cofilin, which promotes filament severing and depolymerization, facilitating the breakdown of existing filaments, and the enhancement of filament growth from newly created barbed ends. It does so in concert with actin interacting protein 1 (Aip1), which serves to accelerate cofilin's activity. While progress has been made in understanding its biochemical functions, the physiologic processes the cofilin/Aip1 complex regulates, particularly in higher organisms, are yet to be determined. We have generated an allelic series for WD40 repeat protein 1 (Wdr1), the mammalian homolog of Aip1, and report that reductions in Wdr1 function …

Although abundant in well-differentiated rat thyroid cells, Rap1GAP expression was extinguished in a subset of human thyroid tumor-derived cell lines. Intriguingly, Rap1GAP was downregulated selectively in tumor cell lines that had acquired a mesenchymal morphology. Restoring Rap1GAP expression to these cells inhibited cell migration and invasion, effects that were correlated with the inhibition of Rap1 and Rac1 activity. The reexpression of Rap1GAP also inhibited DNA synthesis and anchorage-independent proliferation. Conversely, eliminating Rap1GAP expression in rat thyroid cells induced a transient increase in cell number. Strikingly, Rap1GAP expression was abolished by Ras transformation. "Children love to explore the world around them. In doing so they are not aware of disciplinary boundaries or even of disciplines. They move freely from watching ants (biology) to building structures (engineering) to counting rocks (mathematics and geology) to seeing what things dissolve in water (chemistry). Only as they go to school do they learn that humans divide up the way we learn about the universe and start to think within disciplinary boundaries. Uganda’s “National Policy for the Conservation and Management of Wetland Resources” was launched in 1995 to promote the protection of Uganda’s wetlands in order to sustain their ecological and socioeconomic functions. Despite the formal policy, wetlands continue to be drained and converted. This report examines factors that account for the divide between the National Wetlands Policy and reality. Six weeks were spent researching the challenges associated with implementing the National Wetlands Policy. In South Africa, many children find themselves in at risk situations arising from issues like HIV, abuse, and poverty, which highly impact their mental well being by forcing them into unlivable situations. The Durban Children’s Home, located in Glenwood, Durban, works to tackle this problem, providing facilities, meals, and care for 75 homeless, abused, terminally ill, and/or poverty stricken children. Yet in coming from situations where children no longer have support of their primary caregiver or environment, these children require quality psychosocial attention in the Home. I worked for four weeks at the University of Antananarivo (Ankatso), spending two weeks each in the chemistry and pharmacology laboratories. There I ran a total of eight experiments or “screens” with an alcoholic extract of the medicinal plant Mandravasarotra. This plant is found mainly in northwestern Madagascar, and is often used to stimulate contractions for women having difficulty giving birth. The plant tested positive for three secondary metabolites: tannins, steroids and flavanoids. In the pharmacological tests, the plant exhibited anti-inflammatory properties, and high toxicity in doses exceeding 10ml/kg. Together, conjectures may be made between what is known of the …

Ultraviolet radiation has far-reaching effects in marine ecosystems, but many marine organisms have UV-absorbing compounds that protect them from sun-induced damage. The mucus of coral reef fish has been found to contain mycosporine-like amino acids that absorb UV light from 309-360 nm. Using UV spectrophotometry, we examined whether fish are able to allocate these MAA sunscreen compounds to areas of the body that receive the most UV radiation. We compared absorbance spectra of mucus from the body surface of dorsal, ventral, caudal and head areas in two species of Scaridae (Scarus schlegeli and Chlorurus sordidus) from Coral Bay, Western Australia. …

Development of transcriptional pulsing approaches using the c-fos and Tet-off promoter systems greatly facilitated studies of mRNA turnover in mammalian cells. However, optimal protocols for these approaches vary for different cell types and/or physiological conditions, limiting their widespread application. In this study, we have further optimized transcriptional pulsing systems for different cell lines and developed new protocols to facilitate investigation of various aspects of mRNA turnover. We apply the Tet-off transcriptional pulsing strategy to investigate ARE-mediated mRNA decay in human erythroleukemic K562 cells arrested at various phases of the cell cycle by pharmacological inhibitors. Chondrocyte gene regulation is important for the generation and maintenance of cartilage tissues. Several regulatory factors have been identified that play a role in chondrogenesis, including the positive transacting factors of the SOX family such as SOX9, SOX5, and SOX6, as well as negative transacting factors such as C/EBP and delta EF1. However, a complete understanding of the intricate regulatory network that governs the tissue-specific expression of cartilage genes is not yet available. Quercetin, one of the major flavonoids in some fruits and vegetables, has much stronger antioxidative and anticarcinogenic activities. The aim of this study was to evaluate the protective role of quercetin in attenuating the histological changes of spleen and chromosome aberrations in rat bone marrow after NiCl2 acute poisoning. Male Wistar rats received either NiCl2 alone in the dose of 50, 150, and 300 mg/kg and pretreatment with quercetin in the dose of 50, 100, and 200 mg/kg for one hour before administration of NiCl2 300 mg/kg by single oral gavage. Chlamydomonas reinhardtii is a unicellular green alga whose lineage diverged from land plants over 1 billion years ago. It is a model system for studying chloroplast-based photosynthesis, as well as the structure, assembly, and function of eukaryotic flagella (cilia), which were inherited from the common ancestor of plants and animals, but lost in land plants. We sequenced the ∼120-megabase nuclear genome of Chlamydomonas and performed comparative phylogenomic analyses, identifying genes encoding uncharacterized proteins that are likely associated with the function and biogenesis of chloroplasts or eukaryotic flagella. The changes in diet and food consumption practices experienced after displacement can have serious implications for the physiological health of forced migrants and refugees, as noted in Part One.1 These changes can also express cultural loss and social dislocation; the exchange, sharing, preparation and consumption of foods carry important social and cultural values and significances that often reflect and reinforce the very modes of social organization and codes of conduct which become undermined and fractured during displacement (Counihan et al. 1997:3, see also Levi-Strauss 1978, Mead 1970, Appadurai 1981). Mutations in DNA mismatch repair (MMR) lead to increased mutation rates and higher recombination between similar, but not identical sequences, as well as resistance to certain DNA methylating agents. Recently, a component of human MMR machinery, MutLα, has been shown to display a latent endonuclease activity. The endonuclease active site appears to include a conserved motif, DQHA(X)2E(X)4E, within the COOH-terminus of human PMS2. Mosquito control programs at seven urban sites in Kenya, Egypt, Israel, Costa Rica, and Trinidad are described and compared. Site-specific urban and disease characteristics, organizational diagrams, and strengths, weaknesses, obstacles and threats (SWOT) analysis tools are used to provide a descriptive assessment of each mosquito control program, and provide a comparison of the factors affecting mosquito abatement. The information for SWOT analysis is collected from surveys, focus-group discussions, and personal communication. SWOT analysis identified various issues affecting the efficiency and sustainability of mosquito control operations. Vitamin A and its metabolite retinoic acid (RA) are essential elements for normal lung development and the differentiation of lung epithelial cells. We previously showed that RA rapidly activated cyclic AMP response element-binding protein (CREB) in a nonclassical manner in normal human tracheobronchial epithelial (NHTBE) cells. In the present study, we further demonstrated that this nonclassical signaling of RA on the activation of CREB plays a critical role in regulating the expression of airway epithelial cell differentiation markers, the MUC2, MUC5AC, and MUC5B genes. The graduation requirements of the Illinois Mathematics and Science Academy are in concert with those maintained by the State of Illinois with additional requirements as established by the IMSA Board of Trustees. Each semester students must take a minimum of 2.5 credits and a maximum of 3.5 credits. One-semester classes generally receive .5 credits and two semester classes (e.g., World Languages) generally receive 1.0 credit. Most students will take between 5 and 7 academic classes per semester. Field trials were conducted in Kilifi District, Kenya on the activity of Piper guineense and Spilanthes mauritiana powders against field populations of Anopheles gambiae s.l. and Culex quinquefasciatus larvae. Pools containing mosquito larvae were sampled and larval populations determined before and after application of plant powders. Four doses, 0.5, 1.0, 1.5 and 2.0 g L-1, were used in the trials and larval mortality monitored after 24, 48 and 72 h. After 24 h, P. guineense powder at 0.5 g L-1 gave larval mortalities of 18.8 and 23.6% for An. gambiae s.l. and Cx. quinquefasciatus, respectively. At 2.0 g L-1, mortalities …

The mite Tetranychus evansi Baker & Pritchard (Tetranychidae) probably originated in South America. Because of its importance as a tomato pest in Africa, an extensive project has been conducted to detect potentially effective natural enemies in South America for the classical biological control of the pest in Africa. A search for the natural enemies of T. evansi was conducted in the Province of Tucumán, northwestern Argentina, in December 2004, and this report describes the results. One hundred predatory mites of the family Phytoseiidae referring to 11 species were collected on 11 examined species of solanaceous plants. The most abundant phytoseiid …

This study investigated district location and the beliefs of school board members regarding Young Earth Creationism (YEC) and Old Earth Creationism (OEC) associated with the inclusion or exclusion of creationism in the district science curriculum of Georgia public schools. A random sampling of 144 mailed of a total population of 1,034 local school board members was selected as subjects. Data analysis indicated that beliefs of school board members in Young Earth Creationism had a weak, positive association with the permitted inclusion of creationism in the district science curriculum while board member beliefs in OEC had a moderate, negative relationship.
