Habitat destruction has driven much of the current biodiversity extinction crisis, and it compromises the essential benefits, or ecosystem services, that humans derive from functioning ecosystems. Securing both species and ecosystem services might be accomplished with common solutions. Yet it is unknown whether these two major conservation objectives coincide broadly enough worldwide to enable global strategies for both goals to gain synergy. In this article, we assess the concordance between these two objectives, explore how the concordance varies across different regions, and examine the global potential for safeguarding biodiversity and ecosystem services simultaneously. Effective resource management requires robust baseline datasets and efficient monitoring programs to identify and quantify temporal change. The Parque Nacional Sistema Arrecifal Veracruzano (Veracruz Coral Reef System National Park) encompasses a total of 52000ha including 23 coral reefs in two island groups separated by the mouth of the Jamapa River; one near the port of Veracruz, Mexico and one approximately 20km south near Punta Antón Lizardo. Both groups receive substantial fisheries pressure and other anthropogenic impacts. Using non-destructive, visual methods we surveyed fish and benthic assemblages at 18 sites, which included 10 individual coral reefs within the Park. For fishes, …

Neisseria meningitidis serogroup B is a major cause of bacterial meningitis in younger populations. The available vaccines are based on outer membrane vesicles obtained from wild-type strains. In children less than 2 years old they confer protection only against strains expressing homologous PorA, a major, variable outer membrane protein (OMP). We genetically modified a strain in order to eliminate PorA and to overproduce one or several minor and conserved OMPs. Using a mouse model mimicking children's PorA-specific bactericidal activity, it was demonstrated that overproduction of more than one minor OMP is required to elicit antibodies able to induce complement-mediated killing …

A series of novel 10-N-substituted acridones, bearing alkyl side chains with tertiary amine groups at the terminal position, were designed, synthesized, and evaluated for the ability to enhance the potency of quinoline drugs against multidrug-resistant (MDR) Plasmodium falciparum malaria parasites. A number of acridone derivatives, with side chains bridged three or more carbon atoms apart between the ring nitrogen and terminal nitrogen, demonstrated chloroquine (CQ)-chemosensitizing activity against the MDR strain of P. falciparum (Dd2). Isobologram analysis revealed that selected candidates demonstrated significant synergy with CQ in the CQ-resistant (CQR) parasite Dd2 but only additive (or indifferent) interaction in the CQ-sensitive …

The phenolic profile and antioxidant activities of oolong tea extract were investigated after tea was steeped in 90 or 100 oC water for 3 or 10 min. The extraction yield increased with increasing temperature and extended steeping time. However, higher temperature and longer time (100 oC water for 10 min) led to loss of phenolics. The aqueous extract of oolong tea (AEOT) at 100 oC for 3 min exhibited the strongest antioxidant activity. The major polyphenolic components of the AEOT were identified as (-)-epigallocatechin (EGC), (-)-epigallocatechin gallate (EGCG) and (-)-epicatechin-3-gallate (ECG). Cerebrospinal Fluid And Serum Biomarkers Of Cerebral Malaria Mortality In Ghanaian Children, Winston Anderson

Plasmodium falciparum can cause a diffuse encephalopathy known as cerebral malaria (CM), a major contributor to malaria associated mortality. Despite treatment, mortality due to CM can be as high as 30% while 10% of survivors of the disease may experience short- and long-term neurological complications. The pathogenesis of CM and other forms of severe malaria is multi-factorial and appear to involve cytokine and chemokine homeostasis, inflammation and vascular injury/repair. Identification of prognostic markers that can predict CM severity will enable development of better intervention.Postmortem serum and cerebrospinal fluid (CSF) samples were obtained within 2-4 hours of death in Ghanaian children …

The impact of extreme prematurity and related hypoxic-ischemic events on brain development recently has begun to be characterized with modern neuroimaging methods, although comparatively less is known about the neuropathology in those born at heavier birth weights. Even subclinical levels of perinatal hypoxia-ischemia, as indexed by perinatal blood pH, are related to intelligence in school-aged children born preterm. Given the impact of hypoxia-ischemia on white matter and the emerging evidence of specific executive and mathematic deficits in children born preterm, the impact of perinatal hypoxia-ischemia on these outcomes was explored in children at relatively low-risk for sequelae. Optimization Of Electrostatic Interactions In Protein-Protein Complexes, Kelly Brock, Kemper Talley, Kacey Coley, Petras Kundrotas, Emil Alexov

In this article, we present a statistical analysis of the electrostatic properties of 298 protein-protein complexes and 356 domain-domain structures extracted from the previously developed database of protein complexes (ProtCom, http://www.ces.clemson.edu/compbio/protcom). For each structure in the dataset we calculated the total electrostatic energy of the binding and its two components, Coulombic and reaction field energy. It was found that in a vast majority of the cases (>90%), the total electrostatic component of the binding energy was unfavorable. At the same time, the Coulombic component of the binding energy was found to favor the complex formation while …

Virtual microscopy has replaced microscopes representing a sea change in how microscopic anatomy is taught at Thomas Jefferson University (TJU).

The class of 2009 inaugurated virtual microscopy made possible by updating a wet laboratory with new computer stations. Virtual microscopy retains the learning features of microscopes while promoting the learning of relationships through small group interactions. Multi-station computers, equipped with new hardware technology that allows up to 5 monitors and keyboards to run from a single CPU, were installed providing a cost and space effective way to bring virtual microscopy to a large number of students in a small group …

Biological systems have long since perfected the development of nanoengineering. Cellular structure and machinery is based, to a significant extent, on the formation of nanometer to micron-sized assemblies of proteins. These architectures start with unique primary, secondary, tertiary, and quaternary structural complexity and can be constructed from a relatively simple set of building blocks. Using this strategy as inspiration, assemblies based on [Ru(phenanthroline)3]2+ building blocks have been synthesized forming polyruthenium species bridged by the tpphz (tetrapyrido[3,2-a:2’,3’-c:3’’,2’’-h:2’’’,3’’’-j]phenazine) and tatpp (9,11,20,22- tetraaza tetrapyrido[3,2-a:2’,3’-c:3’’,2’’-l:2’’’’,3’’’’-n]-pentacene) ligands. With the International Classification of Functioning, Disability and Health (ICF) the World Health Organization (WHO) has prepared the ground for a comprehensive understanding of Human Functioning and Rehabilitation Research, integrating the biomedical perspective on impairment with the social model of disability. This poses a number of old and new challenges regarding the enhancement of adequate research capacity. Here we will summarize approaches to address these challenges with respect to 3 areas: the organization of Human Functioning and Rehabilitation Research into distinct scientific fields, the development of suitable academic training programmes and the building of university centres and collaboration networks.

Amatoxins, the lethal constituents of poisonous mushrooms in the genus Amanita, are bicyclic octapeptides. Two genes in A. bisporigera, AMA1 and PHA1, directly encode α-amanitin, an amatoxin, and the related bicyclic heptapeptide phallacidin, a phallotoxin, indicating that these compounds are synthesized on ribosomes and not by nonribosomal peptide synthetases. α-Amanitin and phallacidin are synthesized as proproteins of 35 and 34 amino acids, respectively, from which they are predicted to be cleaved by a prolyl oligopeptidase. AMA1 and PHA1 are present in other toxic species of Amanita section Phalloidae but are absent from nontoxic species in other sections. …

In mammalian cells, mRNA decay begins with deadenylation, which involves two consecutive phases mediated by the PAN2-PAN3 and the CCR4-CAF1 complexes, respectively. The regulation of the critical deadenylation step and its relationship with RNA-processing bodies (P-bodies), which are thought to be a site where poly(A)-shortened mRNAs get degraded, are poorly understood. Using the Tet-Off transcriptional pulsing approach to investigate mRNA decay in mouse NIH 3T3 fibroblasts, we found that TOB, an antiproliferative transcription factor, enhances mRNA deadenylation in vivo. Results from glutathione S-transferase pull-down and coimmunoprecipitation experiments indicate that TOB can simultaneously interact with the poly(A) nuclease complex CCR4-CAF1 and …
