This study examines the role of artistic education in shaping artistic beliefs and personal philosophy. Its central focus is the origins of a common belief among artists that artmaking or aesthetic response to art can be a form of spiritual activity or experience leading to spiritual insight. The primary data for this study is a series of in-depth oral history interviews with seven painters who studied with Henry Hensche at The Cape School of Art in Provincetown, MA, a school of American plein air painting that is linked with Impressionism. The ABC transporter P-glycoprotein (Pgp) and membrane permeability are determinant factors for absorption and disposition of xenobiotics. These studies investigated the effects of potential modulators of Pgp and/or membrane function on the disposition, bioavailability and bioaccumulation of environmentally relevant pharmaceuticals, tetracycline and ivermectin, and the carcinogen benzo[a]pyrene in catfish. The pesticide dieldrin and surfactant linear alkylbenzene sulfonate (LAS) were selected as mixture components thought to have potential interactions with these determinants of disposition. Initial in situ experiments demonstrated dieldrin and tetracycline both significantly inhibited biliary excretion of Pgp prototypic substrate Rhodamine-123 in isolated perfused livers. Extinction is a very important component of functional communication training (FCT). Thus, the potential undesirable effects of extinction must be considered before this type of treatment is implemented. Resurgence, the recurrence of previously reinforced behavior when another behavior is placed on extinction, is a possible undesirable effect of extinction. Resurgence may account for some instances of treatment relapse in situations where problem behavior recovers following implementation of extinction-based treatments such as FCT. Despite the potential relevance of resurgence to understanding why problem behavior may re-emerge, few applied studies have examined resurgence effects. The purpose of this study was the development and testing of a novel method for assessment of white blood cell (WBC) identification skills used in the field of Clinical Laboratory Sciences (CLS). A dual format exam was administered to both novices (students) and experts (laboratory professionals). Format 1 was similar to current assessment formats, simply presenting a series of single WBC images for identification. Format 2 applied principles of visual cognition, grouping WBCs for identification by patient and presenting multiple example images from the patient before requesting identification of individual cells. Red imported fire ants (RIFA), Solenopsis invicta Buren, are medical, urban, and agricultural pests from South America. Fourth-instar larvae are used by the colony to digest solid food and then regurgitate it for consumption by workers and queens. Larvae are an ideal source of investigations of obligate endosymbiotic bacteria possibly involved in nutrient distributions. I attempted to first identify what bacteria species are in the guts of larvae followed by antibiotic and microscopy work to further study their roles in the ants. In the vertebrate retina, multiple cell types express g protein-coupled receptors linked to phospholipase C. The signaling pathway engendered by activation of this enzyme can involve Ca2+-permeable transient receptor potential (TRP) channels. To begin to understand the role of these channels in the retina, we undertake an immunocytochemical localization of two TRPC channel subunits, TRPC1 and TRPC4. TRPC1 expression was observed in amacrine cells and their process in the chicken retina. Aspects of the population ecology of a parasitoid (Pseudacteon tricuspis) of the red imported fire ant (Solenopsis invicta) in Louisiana were studied. The spatio-temporal abundance patterns, dispersal, population spread, aggregation, direct mutual interference and functional response characteristics of this parasitoid were studied to address deficiencies in our knowledge about phorid flies, particularly Pseudacteon parasitoids. This endoparasitoid was discovered to manipulate host ant behavior in ways that benefit its own survival. Laboratory experiments to gain insights into behavioral and functional responses revealed that fly aggregations were density-dependent and interference was not significant when 1-3 females were simultaneously confined with hosts, although …

In 2003, there were reports of child maltreatment affecting over 5.5 million children in the United States. As a result of this epidemic, over 500,000 children are in foster care with an estimated additional 300,000 in voluntary kin placements. Because of a shortage of foster families and resources, Child Protective Services (CPS) agencies routinely seek and give priority to family members to serve as foster families to maltreated children. Simple water soluble lanthanum and europium complexes are effective at detecting neutral sugars as well as glyco- and phospholipids. In solutions at physiologically relevant pH the fluorescent lanthanum complex binds neutral sugars with apparent binding constants comparable to those of arylboronic acids. Interference from commonly occurring anions is minimal. The europium complex detects sialic acid-containing gangliosides at pH 7.0 over an asialoganglioside. This selectivity is attributed, in large part, to the cooperative complexation of the oligosaccharide and sialic acid residues to the metal center, based on analogous prior studies. My graduate work has focused on the development of new fluorophore materials towards addressing several interdisciplinary research problems. Selective detection of sialic acid via a boron-amide interaction. Concentration changes of sialic acid can be monitored ratiometrically in the visible region using a boronic acid based chemosensor. The technique shows great promise for the detection of sialic acid, a critical cell surface residue involved in cancer, graft rejection and virulence in certain bacteria. The discharge of hydrophobic organic chemicals (HOCs) and other hazardous substances from anthropogenic sources to surface waters and the atmosphere has led to the widespread contamination of bottom sediments throughout the world. HOCs collect in sediments due to their affinity for organic matter and other sorbents present on and within their solid matrices. The conservation equation and the equations of motion are solved for a case where a thin liquid film moves out of a slot onto a horizontal surface. The liquid is allowed to evaporate into air. The evaporation process is taken to be isothermal. Lubrication theory approximation is used where only the tangential velocity and its dependence only in the normal direction are considered. The dynamics of thin films includes the use of disjoining pressure for a pure liquid and where there is a dissolved polymer. This book chapter compares the depictions of Cuban women in Louisa May Alcott's first adult novel Moods (1864) and Elizabeth Stoddard's short story "Eros and Anteros" (1862). Both writers configure a love triangle between an Anglo man and two women, one Anglo and one Cuban. In both texts, the Cuban woman is rejected as an unsuitable choice for an Anglo man. Alcott’s and Stoddard’s decision to re-value the Anglo woman as the more appropriate choice can be read as a rejection of the popular nineteenth-century political doctrine of manifest destiny and, at least with Alcott, of the United States’s dependence …

This thesis assessed the physical characteristics and stability of selected western Minnesota streams that cross the eastern beach ridges of glacial Lake Agassiz. Stream and river channels in this area host unusually diverse aquatic habitat and biodiversity, but incompatible land use along channels and in watersheds impair their quality and function. Previous rapid assessment protocols have been developed for stream attributes and stability, but these methods have been developed elsewhere in the United States and are not necessarily appropriate for western Minnesota’s unique geology and landscape.

Electrical models for biological cells predict that reducing the duration of applied electrical pulses to values below the charging time of the outer cell membrane (which is on the order of 100 ns for mammalian cells) causes a strong increase in the probability of electric field interactions with intracellular structures due to displacement currents. For electric field amplitudes exceeding MV/m, such pulses are also expected to allow access to the cell interior through conduction currents flowing through the permeabilized plasma membrane. Mutations in the Plasmodium falciparum chloroquine (CQ) resistance transporter (PfCRT) can result in verapamil-reversible CQ resistance and altered susceptibility to other antimalarials. PfCRT contains 10 membrane-spanning domains and is found in the digestive vacuole (DV) membrane of intraerythrocytic parasites. The mechanism by which PfCRT mediates CQ resistance is unclear although it is associated with decreased accumulation of drug within the DV. The first full-length mRNA for vitellogenin (Vg) from ticks was sequenced. This also represents the first complete sequence of Vg from the Chelicerata and of a heme binding Vg. The Vg cDNA from the American dog tick, Dermacentor variabilis was 5744nt in length (GenBank Accession number AY885250), which coded for a protein of 1843aa with a calculated molecular weight of 208kD. This protein had an 18 aa signal sequence, a single RXXR cleavage signal that would generate two subunits (49.5 and 157K in molecular weight) and lipoprotein N-terminal and carboxy von Willebrand factor type D domains. The Caranx hippos species complex comprises three extant species: crevalle jack (Caranx hippos) (Linnaeus, 1766) from both the western and eastern Atlantic oceans; Pacific crevalle jack (Caranx caninus) Günther,1868 from the eastern Pacific Ocean; and longfin crevalle jack (Caranx fischeri) new species, from the eastern Atlantic, including the Mediterranean Sea and Ascension Island. Monthly and annual phytoplankton productivity rates of four Virginia tidal rivers were determined based on a 12-year monitoring study that included sampling stations from tidal freshwater, oligohaline, and mesohaline regions in these rivers. The mean monthly rates and range at these locations were 5.52 (Dec.) to 175.12 (Aug.) mg C m-3 h-1 for tidal freshwater, 12.21 (Jan.) to 149.90 (May) mg C m-3 h-1 in oligohaline regions, and 16.20 (Jan.) to 151.33 (May) mg C m-3 h-1 for the mesohaline. The transcript sequence of the Amblyomma americanum Linnaeus (Acari: Ixodidae) defensin, termed amercin (amn), was ascertained and a 219-bp amn coding region identified. The gene encodes a 72-amino acid prepropeptide with a putative 37-amino acid mature peptide. This gene shows little similarity to either of the defensins from Amblyomma hebraeum Koch, the only other Amblyomma species for which a defensin has been described. Fascinating new data, revealed through gene sequencing, comparative genomics, and genetic engineering, precisely establish which genes are involved in mate choice and mating activity—behaviors that are surprisingly understudied from a genetic perspective. Discussed here are some of the recently identified visual and chemosensory genes that are involved in mate choice and mating behavior. These genes’ products are involved in the production, transmission, and receipt of crucial sensory mate-choice cues that affect fitness. A series of novel 10-N-substituted acridones, bearing alkyl side chains with tertiary amine groups at the terminal position, were designed, synthesized, and evaluated for the ability to enhance the potency of quinoline drugs against multidrug-resistant (MDR) Plasmodium falciparum malaria parasites. A number of acridone derivatives, with side chains bridged three or more carbon atoms apart between the ring nitrogen and terminal nitrogen, demonstrated chloroquine (CQ)-chemosensitizing activity against the MDR strain of P. falciparum (Dd2). Isobolograrn analysis revealed that selected candidates demonstrated significant synergy with CQ in the CQ-resistant (CQR) parasite Dd2 but only additive (or indifferent) interaction in the CQ-sensitive …

The Asian snake Rhabdophis tigrinus possesses specialized defensive glands on its neck that contain steroidal toxins known as bufadienolides. We hypothesized that R. tigrinus does not synthesize these defensive steroids but instead sequesters the toxins from toads it consumes as prey. To test this hypothesis, we conducted chemical analyses on the glandular fluid from snakes collected in toad-free and toad-present localities. We also performed feeding experiments in which hatchling R. tigrinus were reared on controlled diets that either included or lacked toads. We demonstrate that the cardiotonic steroids in the nuchal glands of R. tigrinus are obtained from dietary toads. …

Real-time study of the transport and biocompatibility of nanomaterials in early embryonic development at single-nanoparticle resolution can offer new knowledge about the delivery and effects of nanomaterials in vivo, and provide new insights into molecular transport mechanisms in developing embryos. In this study, we directly characterized the transport of single silver nanoparticles into an in vivo model system (zebrafish embryos) and investigated their effects on early embryonic development at single-nanoparticle resolution in real time.
