OBJECTIVE: To assess serum levels of nitric oxide and peroxynitrite in patients presenting with cerebral infarction resulting from middle cerebral artery occlusion, at 48 hours from stroke onset. METHODS: We conducted the study in the Department of Pharmacology and in cooperation with Al-Yarmouk Teaching Hospital and the Department of Medicine, College of Medicine, Al-Mustansiriya University, Baghdad, Iraq from October 2003 to May 2004. OBJECTIVE: To rapidly detect rifampin resistance in Mycobacterium tuberculosis isolates causing meningitis in northeast Iran. METHODS: This study presents the results of a polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) analysis for the evaluation of rifampin resistance directly from the CSF of 47 patients strongly suspicious to have tuberculosis meningitis in Emam Reza University Hospital, Mashhad, Iran over 3 years (2002 to 2005). Each CSF sample underwent microscopic examination, culture and DNA amplification by 2 PCR protocols and subsequent detection of mutations by SSCP analysis. OBJECTIVE: To comparatively analyze the ultrastructural changes in the submandibular and parotid glands and in the exocrine pancreas following diabetes induced by Streptozotocin exposure and the effects of fasting and insulin treatment on these alterations. METHODS: For experimental procedure, we included 48 Sprague-Dawley type rats in July 2001-March 2002 at Gazi University, Turkey. We divided the rats into 8 groups following the infusion of Streptozotocin. High Frequency Measurements Of Soil Carbon Dioxide Flux At Harvard Forest, Stephen C. Phillips

Soil carbon dioxide flux was measured by automatic chambers at Harvard Forest over a four-year period, 2003-2006. The autochambers were installed along a moisture gradient from upland to wetland soils. In 2003, fluxes from the upland and mid-slope chambers exceeded the fluxes from the wetland margin. In 2004-2006, the mid-slope fluxes were significantly larger than both the upland and wetland margin chambers. The differences in flux between chamber location were most pronounced in the late summer and early fall. Residuals from a non-linear temperature regression exhibit a distinct seasonal pattern in 2003, 2004, and 2006, but not in 2005. On …

The state is coordinating an Instream flow study for the Lamprey River in the Seacoast of NH and this research adds a key component to the existing study. This thesis explains how the values and objectives of stakeholders and the general public were elicited. Fourteen stakeholders participated in an interview and survey process and 229 randomly selected residents of the Lamprey River Watershed completed surveys about they how use and value the river. …

We describe three software tools related to research in comparative genomics, a growing research area that explores the variation within and between organisms. We developed a set of tools that explore sequence similarity and differences in genomes. Two of these tools are specifically aimed at examining DNA sequence data from two or more genomes: (1) The Magenta's OPUS tool compares genomic sequences to identify shared or unique segments between closely related species. This tool looks for functional similarities and differences in genomic data by classifying sequences into groups based on genomic categories: Orthologs, Paralogs, and Unique Sequence. (2) The DSNP …

Invasion Of Transition Hardwood Forests By Exotic Rhamnus Frangula: Chronology And Site Requirements, Hanna S. Wingard

The invasion chronology and site requirements of the exotic, invasive shrub, glossy buckthorn (Rhamnus frangula), were studied within intact transition hardwood---hemlock---white pine forests in Durham, NH. Data were collected from 63 plots (7 sites with 9 plots each). Within sites, plots were arranged over a topographic gradient with 3 plots in each of 3 topographic positions (upper, middle, and lower slope). From each plot, information on soil nutrients and texture, soil moisture, overstory community composition, and canopy openness was collected. Densities of R. frangula in two size classes (≥1 m in height, <1 m in height) within each plot were calculated. Morphological data (height, diameter at stem base, number of live and dead stems, and age) were collected from each individual ≥1 m tall. R. frangula invaded these intact, closed-canopy forests over 30 years ago and the invasion has continued, with population density increasing over time. Nominal logistic regression suggested that site, topographic position, and overstory community type played a role in whether or not R. frangula was present in a plot. Plots on upper slopes dominated by Tsuga canadensis were less likely to become invaded than other plots. Multiple linear regression showed that density of R. frangula (≥1 m) increased with time since invasion and increasing soil quality. Density of R. frangula (<1 m) was positively influenced by time since invasion, canopy openness, and density of R. frangula (≥1 m). Multiple regressions also showed that mean age and age of oldest R. frangula per plot were higher in plots with higher soil moisture content. Size of R. frangula individuals (i.e. height, diameter) generally increased with mean age of stems. The results show that successional, transition hardwood forests can be invaded by R. frangula and should not be overlooked when managing against this species. Special attention should be given to low topographic positions and areas with rich soils, while upland areas with dense hemlock canopy are much less likely to experience heavy invasion by R. frangula..

The rapid proliferation of invasive Phragmites australis in New England has challenged resource managers to curb its expansion and reduce the loss of functional salt marsh. The interactive effects of sediment elevation, nutrient additions and community structure (plant competition and diversity) on the success and health of Phragmites were examined within a salt marsh in Hampton, NH. Along with four native halophytes, Phragmites was transplanted to vegetation plots (0.09 m2) comprising three community structure types (Phragmites alone, PA+1sp., PA+4spp.). Plots were arranged across elevation and nutrient enrichment gradients. Elevation and fertilization had no effect on Phragmites success, likely due to …

Surface plasmon coupled emission (SPCE) is a process by which isotropic fluorescent emission, from a fluorescent dye or biological agent labeled with a fluorescent dye, is channeled into highly directional emission. This process relies on the interaction between fluorescent dye molecules and thin silver films in close proximity to each other. Recent studies have shown that SPCE greatly increases the sensitivity of spectroscopic methods by increasing the amount of light that can be collected from a fluorescent molecule. The Notch signaling pathway regulates adipogenesis. To determine specific effects the Notch ligand Jagged-1 has on the regulation of adipogenesis, we transfected 3T3-L1 preadipocytes with either a constitutively expressed full-length human Jagged-1 construct or an empty vector control. These cells were grown in conditions promoting differentiation and then evaluated for lipid accumulation and gene expression. To test the role of Jagged-1 in adipose tissue development in vivo, transfected cells were examined for their ability to form fat pads after injection into nude mice. In vitro, cells constitutively expressing Jagged-1 accumulated less lipid and exhibited a delay in adipocyte protein expression. …

Humans experience climate variability and climate change primarily through changes in weather at local and regional scales. In this work, changes in northeastern United States winter climate are documented using meteorological observations from 1965-2005. Spatial coherence analysis is utilized to remove stations with non-climatic influences from the analysis. Trends over the past four decades in snowfall, snow-covered days, mean, minimum, and maximum temperature are estimated using linear regression.

Northeastern United States regional winter maximum temperatures (+0.43 +/-.08°C/decade) are warming greater rate than minimum (+0.37 +/-0.10°C/decade) and mean (+0.39 +/-0.10°C/decade). Regional winter snowfall decreased by -2.5 +/-0.8 inches/decade. Overall snowfall decreases …

Eicosanoids are potent mediators of inflammatory response whose role has been well established in inflammatory disorders. Release of arachidonic acid by group IVA cytosolic phospholipase A2 α (cPLA2α) is the initial rate limiting step for the production of eicosonoids in response to inflammatory mediators. Previous findings from our laboratory have demonstrated that cPLA2α is directly activated by the emerging bioactive sphingolipid, ceramide-1-phosphate (C1P). In this study, we have developed a modified Triton X-100/phosphatidylcholine (PC) mixed micelle assay which was utilized to determine the kinetics and specificity of this lipid-enzyme interaction. Pre-treatment, post-overbite correction and post-treatment records were gathered from 32 patients who presented with deep overbite malocclusions to the Virginia Commonwealth University orthodontic clinic. The occurrence of thrombosis in several pathophysiological conditions creates a huge need for anticoagulation therapy. Thrombin and factor Xa have been prime targets for regulation of clotting through the direct and indirect mechanism of inhibition. This work investigates chemo-enzymatically prepared oligomers of 4-hydroxycinnamic acids (DHPs) as potential anticoagulants. Oligomers were prepared through peroxidase-catalyzed oxidative coupling of 4-hydroxycinnamic acids. The products resulting from this reaction are called CDs, FDs and SDs. Structurally, these sulfated DHPs are unique and do not resemble any of the anticoagulants known in the literature.DHP oligomers were found to increase clotting times at concentrations comparable to heparin. …

Copper-organonitrogen complexes are studied by threshold photoionization and zero electron kinetic energy photoelectron spectroscopy. These complexes are prepared in pulsed laser vaporization supersonic molecular beams. Adiabatic ionization energies of the neutral species and vibrational frequencies of the neutral and ionic complexes were measured. Metal-ligand bond dissociation energies were obtained from the theoretical calculations or the experiments. More importantly, by combining the spectroscopic measurements, quantum chemical calculations, and spectral simulations, metal-ligand bonding structures are determined for copper complexes of diamines, pyridine, diazines, aminopyridines, polypyridines, and imidazole. Understanding the causes of individual differences in brain structure may give clues about the etiology of cognition, personality, and psychopathology, and also may identify endophenotypes for molecular genetic studies on brain development. We performed a comprehensive statistical genetic study of anatomic neuroimaging data from a large pediatric sample (N=600+) of twins and family members from the Child Psychiatry Branch at the NIMH. These analyses included variance decomposition of structural volumetric endophenotypes at several levels of resolution, voxel-level analysis of cortical thickness, assessment of gene by age interaction, several multivariate genetic analyses, and a search for genetically-mediated brain-behavioral relationships. I quantified seasonal variations in corticular photosynthesis in 1st through 5th order branches of Myrica cerifera L. (Myricaceae) in order to determine whether corticular photosynthesis contributes to whole plant carbon gain by reducing respirational CO2 loss. Maximum % refixation was 110 ± 39 % of CO2 efflux in the dark (Rd) in 1st order branches during winter, minimum was 18 ± 3 % in 5th order branches during summer. Variations in % refixation paralleled changes in photosynthetically active radiation (PAR). As light attenuated with increasing branch order % refixation decreased.
