Cisplatin-Based Chemotherapy For Treatment Of Head And Neck Cancers, Osteosarcomas Of The Jaw Bone, And Ovarian Cancers, Tara Kersten

In this project, I have done extensive research on two chemotherapy regimens, cisplatin/5-fluorouracil and cisplatin/Adriamycin. My hope is to compare and contrast the effectiveness of cisplatin-based regimens in the treatment of head and neck squamous cell carcinomas, osteosarcomas of the jaw, and ovarian cancers. In addition to literature searches, I have studied the medical files of two patients who were treated with cisplatin. One patient had recurrent, unresectable head and neck cancer and one patient has a resected, metastatic osteosarcoma of the mandible. After studying the case of the patient with head and neck cancer, I found exceptional results. After …

Nodule formation is one of the main cellular mechanisms insects utilize in response to infection. The nodulation reaction accounts for clearing nearly 90% of the bacterial cells from circulation. Furthermore, the nodulation assay is a useful tool for developing an understanding of how insects immune response to challenge. Little is known of environmental effects such as heavy metal toxicity on insect immunity. This study was designed to investigate a variety of heavy metals influence on the cellular immune response of the tobacco homworm, Manduca sexta, to bacterial challenge. What kind of a system or process would portray a twelve-year-old girl from the Yellow Quill First Nation as a sexual threat to three twenty-something white men, who promising her a ride home, picked her up and got her drunk? The answer: The Canadian legal system. In R v Edmondson a twelve-year old Aboriginal girl, who was sexually assaulted by Dean Edmondson, Jeffrey Brown, and Jeffrey Kindrat from Tisdale Saskatchewan, was portrayed by the judge as a "sexual aggressor". The formation of viable genetic chimeras in mammals through the transfer of cells between siblings in utero is rare. Using microsatellite DNA markers, we show here that chimerism in marmoset (Callithrix kuhlii) twins is not limited to blood-derived hematopoietic tissues as was previously described. All somatic tissue types sampled were found to be chimeric. Notably, chimerism was demonstrated to be present in germ-line tissues, an event never before documented as naturally occurring in a primate. In fact, we found that chimeric marmosets often transmit sibling alleles acquired in utero to their own offspring. Thus, an individual that contributes gametes to …

Exploring The Mechanism Of Flexible Biomolecular Recognition With Single Molecule Dynamics, H. Peter Lu, Qiang Lu, Jin Wang

Combining a single-molecule study of protein binding with a coarse grained molecular dynamics model including solvent (water molecules) effects, we find that biomolecular recognition is determined by flexibilities in addition to structures. Our single-molecule study shows that binding of CBD (a fragment of Wiskott-Aldrich syndrome protein) to Cdc42 involves bound and loosely bound states, which can be quantitatively explained in our model as a result of binding with large conformational changes. Our model identified certain key residues for binding consistent with mutational experiments. Our study reveals the role of flexibility and a new scenario of dimeric binding between the monomers: …

The Mentors in Violence Prevention (MVP) Program is the subject of this research study. MVP is a high school leadership and mentoring program that focuses on promoting gender respect and preventing harassment, sexual harassment and teen dating abuse in middle schools and high schools. This program evaluation study of the MVP program utilizes both quantitative and qualitative data. The findings suggest the MVP training supports the mentors in their ability to become active bystanders and leaders in this work, not only in the short term and while they are in the program, but over the long term, when they are …

The emergence of cell phones with embedded Global Positioning System (GPS) chips provides opportunities to push personalized real-time travel information subject to an individual's current location. One such application, a travel assistant device, notifies cognitively disabled public transportation users when it is time to request a stop and exit the vehicle. GPS-enabled cell phones must provide highly accurate real-time location data for this type of service. The components used in the data-collection process are identified, and a quantitative analysis of real-time GPS data obtained with a cell phone while walking, driving a vehicle, and riding public transportation is provided. A great deal of research on multiculturalism looks at different approaches to multicultural education and visions of multicultural teaching and learning. Though some research theorizes about how preservice teachers might learn about race or gender, there is very little work that helps teacher educators understand what learning about diversity more broadly, might look like. This study uses the conceptual framework developed by Paine to raise questions about and illuminate differences in the learning outcomes of preservice teachers who participated in two similar yet notably different service-learning experiences. Nitric oxide (NO) plays an important role in physiological functions like vasodilation, neurotransmission, and inhibition of platelet aggregation. The endothelium-derived NO diffuses into the vascular lumen where it interacts with flowing blood as well as the smooth muscles where it modulates vascular tone. However, uncertainty exists on how NO escapes the rapid scavenging by hemoglobin (Hb) and reaches smooth muscles. Several proposed hypotheses include 1) a reduced reaction rate of NO with Hb contained inside red blood cells (RBCs) and 2) NO preservation in the bound form of s-nitrosohemoglobin or nitrite. Nitric oxide (NO) produced by endothelial cells is a key component for blood-vessel dilation. Dilation is achieved through smooth muscle relaxation as a response to NO transport. Inhibition of this process occurs through the inactivation of NO by reactive oxygen species, especially superoxide (O2 -). NO and superoxide react quickly, forming peroxynitrite (ONOO-). Both superoxide and peroxynitrite apply oxidative stress on vascular tissue. Experimental studies investigating NO interactions are difficult since these reactions occur rapidly and over small distances. This study presents a computational model to describe the interactions of NO, superoxide, and peroxynitrite across an arteriole/venule pair. Based on …

Mitochondria are essential for promoting cell survival and growth through aerobic metabolism and energy production. Mitochondrial function is typically analyzed using mitochondria freshly isolated from tissues and cells because they yield tightly coupled mitochondria, whereas those from frozen tissue can consist of broken mitochondria and membrane fragments. A method, utilizing a well-characterized cryoprotectant such as dimethyl sulfoxide (DMSO), is described. Such mitochondria show preserved structure and function that presents us with a possible strategy to considerably expand the time-frame and the range of biochemical, molecular and metabolic studies that can be performed without the constraints of mitochondrial longevity ex vivo …
