Innate Antiviral Response Targets Hiv-1 Release By The Induction Of Ubiquitin-Like Protein Isg15, Atsushi Okumura, Gengshi Lu, Ian Pitha-Rowe, Paula M. Pitha

The goal of this study was to elucidate the molecular mechanism by which type I IFN inhibits assembly and release of HIV-1 virions. Our study revealed that the IFN-induced ubiquitin-like protein ISG15 mimics the IFN effect and inhibits release of HIV-1 virions without having any effect on the synthesis of HIV-1 proteins in the cells. ISG15 expression specifically inhibited ubiquitination of Gag and Tsg101 and disrupted the interaction of the Gag L domain with Tsg101, but conjugation of ISG15 to Gag or Tsg101 was not detected. The inhibition of Gag-Tsg101 interaction was also detected in HIV-1 infected, IFN-treated cells. Elimination …

With this dissertation I developed a new theory and measure of ethical work climate (EWC). Currently, there exists one dominant theory and measure of EWC developed by Victor and Cullen (1988, 1987). Even though researchers have identified problems with this theory, such as inconsistencies with regard to its limited theoretical scope and troubling psychometric properties, it is the most widely utilized framework for conceptualizing and testing EWC. Therefore, I propose to develop an improved theory and measure of EWC, one capable of addressing some of the principle shortcomings of earlier efforts. Atmospheric mercury deposition, known to be a major source of mercury to aquatic and terrestrial environments, was studied at an urban site in Orlando, FL. Precipitation sampling was conducted from September 2003 to May 2006 at a Mercury Deposition Network site located on the University of Central Florida campus. Weekly rainfall and mercury wet deposition data were gathered from this site, which provided the framework of data for this study. Historical mercury wet deposition data from several sites in Florida were used to develop a regression model to predict mercury deposition at any location in Florida. Stormwater runoff from a …

Ubvi Ccd Photometry Of The Old Open Cluster Berkeley 17, Zosia A. C. Krusberg, Brian Chaboyer

Photometric UBVI CCD photometry is presented for NGC 188 and Berkeley 17. Color-magnitude diagrams (CMDs) are constructed and reach well past the main-sequence turnoff for both clusters. Cluster ages are determined by means of isochrone fitting to the cluster CMDs. These fits are constrained to agree with spectroscopic metallicity and reddening estimates. Cluster ages are determined to be 7.0 ± 0.5 Gyr for NGC 188 and 10.0 ± 1.0 Gyr for Berkeley 17, where the errors refer to uncertainties in the relative age determinations. These ages are compared to the ages of relatively metal-rich inner halo/thick-disk globular clusters and other …

The goal of the current study was to generate a comprehensive, multi-tissue perspective of the effects of chronic hypoxic exposure on carbohydrate metabolism in the Gulf killifish Fundulus grandis. Fish were held at approximately 1.3·mg·l–1 dissolved oxygen (~3.6·kPa) for 4·weeks, after which maximal activities were measured for all glycolytic enzymes in four tissues (white skeletal muscle, liver, heart and brain), as well as for enzymes of glycogen metabolism (in muscle and liver) and gluconeogenesis (in liver). The specific activities of enzymes of glycolysis and glycogen metabolism were strongly suppressed by hypoxia in white skeletal muscle, which may reflect decreased energy …

High-performance computing increasingly occurs on “computational grids” composed of heterogeneous and geographically distributed systems of computers, networks, and storage devices that collectively act as a single “virtual” computer. A key challenge in this environment is to provide efficient access to data distributed across remote data servers. Our parallel I/O framework, called Armada, allows application and data-set providers to flexibly compose graphs of processing modules that describe the distribution, application interfaces, and processing required of the dataset before computation. Tree squirrels (Sciurus spp.) have exhibited high seroprevalence rates, suggesting that they are commonly exposed to West Nile virus (WNV). Many characteristics of WNV infections in tree squirrels, such as the durations and levels of viremia, remain unknown. To better understand WNV infections in fox squirrels (S. niger), we subcutaneously inoculated fourteen fox squirrels with WNV. Peak viremias ranged from 104.00 plaque-forming units (PFU)/mL of serum on day 2 post-infection (DPI) to 104.98 PFU/mL on 3 DPI, although viremias varied between individuals. A reversed-phase ion-pair liquid chromatogaphic analysis combined with a solid-phase extraction clean-up method is used to assess the quantity of diphacinone residue found in invertebrates. Three invertebrate species are exposed to commercially available diphacinone-fortified bait used for rat control. The invertebrate samples are collected, frozen, and shipped to the laboratory. The samples are homogenized after cryogenic freezing. A portion of the homogenized samples are extracted with acidified chloroform-acetone, followed by cleanup with a silica solid-phase extraction column. Diphacinone is detected by UV absorption at 325 nm after separation by the chromatographic system. The method limit of detection (MLOD) for snail …

Blackbirds are ubiquitous members of the avian fauna in the Prairie Pothole Region. However, their abundance combined with their food habits make blackbirds significant agricultural pests on sunflower. Cost estimates for blackbird damage to sunflower in the northern Great Plains range from 4-11 million U.S. dollars per year. Because of their economic impact on agriculture, it is imperative to understand the environmental factors that influence blackbird abundance patterns. This study attempts to quantify the effects of landscape-level land use and climate patterns on blackbird abundance in the Prairie Pothole Region of the United States.

Biofilms are structured communities of bacterial cells enclosed in a self-produced polymeric matrix and adherent to an inert or living surface; they have clinical, industrial and environmental impacts. Biofilms that are established by bacteria on plants are found on the surfaces of roots, leaves, seeds and internal vascular tissues where the microbes live in commensal, mutualistic or parasitic/pathogenic associations with their host. The study of the structure of plant-associated biofilms has been considerably helped by the development of techniques using fluorescent markers coupled with confocal scanning laser microscopy as well as scanning electron microscopy.
