Insulin receptor substrate (IRS)-1 is a key protein in insulin signaling. Several studies have shown that the expression of IRS-1 can be modulated by protein degradation via the proteasome and the degradation of IRS-1 can be related to insulin-resistant states. The degradation of IRS-1 has been shown to be induced by SOCS-1 and SOCS-3 via the ubiquitin pathway. The goal of our study was to determine if the induction of SOCS-3 correlated with increased IRS-1 degradation in cultured 3T3-L1 adipocytes. Interestingly, our studies have shown that there is little correlation between the induction in SOCS-3 expression and the degradation of …

Conceptualizing And Measuring The Self In Chronic Illness And Its Relationship To Adjustment, Jessica Marie Morea
Conceptualizing And Measuring The Self In Chronic Illness And Its Relationship To Adjustment, Jessica Marie Morea
Dissertations and Theses
This research sought to clarify the role of the self in chronic illness by developing and evaluating several new constructs, which intend to describe the extent to which illness permeates the self-concept. Following the works of Lewin and Asch, the concepts of central and peripheral regions of the self were elaborated to describe the state of illness within the self, termed "illness self-concept".
Three subsidiary constructs were introduced to further depict illness self-concept: directionality, pervasiveness, and illness self-consciousness. Directionality refers to whether illness drives the self or self drives illness. Pervasiveness refers to whether illness affects many or few regions …

Faculty Publications
Natural organic matter was collected from the upper Yukon River and size fractionated into the (LMW-DOC), colloidal (COC, 1 kDa to 0.45 mu m) and particulate organic carbon (POC, > 0.45 mu m) phases for characterization of elemental (C and N) and isotopic (C-13, C-14 and N-15) composition to examine their sources and transport. Concentrations of total organic carbon (TOC) decreased from 3010 mu M in mid-May to 608 mu M in September, accompanying an increase in river water delta O-18 from the snowmelt to summer and early fall. COC was the predominant OC species, comprising, on average, 63 +/- 8% …

Spatial And Temporal Patterns Of Feeding And Food In Three Species Of Mellitid Sand Dollars, Susan Elizabeth Hilber
Spatial And Temporal Patterns Of Feeding And Food In Three Species Of Mellitid Sand Dollars, Susan Elizabeth Hilber
USF Tampa Graduate Theses and Dissertations
Sand dollars are abundant and conspicuous macroinvertebrates in sandy habitats in the Gulf of Mexico. They ingest sediment and associated organisms. Given their high abundances, size, and deposit feeding, they likely are the most important consumer coupling primary productivity with the rest of the food chain in these habitats. Moreover, sand dollars bioturbate sediments and affect species diversity and community structure.
Three species of sand dollars in the family Mellitidae,
Mellita tenuis, Encope michelini and Encope aberrans, were studied off the West Coast of Florida. Food and feeding of these sand dollar species were studied to understand their relationship …

Performance Evaluation Of The Self-Organizing Map For Feature Extraction, Yonggang Liu, Robert H. Weisberg, Christopher N. K. Mooers
Marine Science Faculty Publications
Despite its wide applications as a tool for feature extraction, the Self‐Organizing Map (SOM) remains a black box to most meteorologists and oceanographers. This paper evaluates the feature extraction performance of the SOM by using artificial data representative of known patterns. The SOM is shown to extract the patterns of a linear progressive sine wave. Sensitivity studies are performed to ascertain the effects of the SOM tunable parameters. By adding random noise to the linear progressive wave data, it is demonstrated that the SOM extracts essential patterns from noisy data. Moreover, the SOM technique successfully chooses among multiple sets of …

Alu elements represent the largest family of human mobile elements in copy number. A controversial issue with implications for both Alu biology and human genome evolution is whether selective pressures are affecting Alu elements on a large scale. To address this issue, we analyzed the genomic distribution of the three youngest known human Alu subfamilies (Ya5a2, Ya8 and Yb9) in conjunction with their insertion polymorphism status in the human population, since selection can only act on polymorphic elements. Our results indicate that: (i) polymorphic and fixed recently integrated Alu elements are found in genomic regions whose GC contents are statistically …
Knowledge Based Structure Modeling Of The Third Hypervariable Region Of Antibodies, Kevin Galens
Knowledge Based Structure Modeling Of The Third Hypervariable Region Of Antibodies, Kevin Galens
Theses
Protein structure prediction has gained increased attention over the past decades in a wide range of biological disciplines. Creating an accurate visual model of a protein can aid in protein engineering; which has implications in the creation of therapeutic molecules as is the case with antibodies. The third complementarity determining region of the heavy chain of antibodies (CDR-H3) is known to show a large degree of variation in sequence and in length, and therefore has provided difficulties for structure prediction. By separating the CDR-H3 into two logical sections, the apex and base, and using a homology modeling techniques for each …

Estimating The Retrotransposition Rate Of Human Alu Elements, Richard Cordaux, Dale J. Hedges, Scott W. Herke, Mark A. Batzer

Mobile elements such as Alu repeats have substantially altered the architecture of the human genome, and de novo mobile element insertions sometimes cause genetic disorders. Previous estimates for the retrotransposition rate (RR) of Alu elements in humans of one new insertion every ∼100-125 births were developed prior to the sequencing of the human and chimpanzee genomes. Here, we used two independent methods (based on the new genomic data and on disease-causing de novo Alu insertions) to generate refined Alu RR estimates in humans. Both methods consistently yielded RR on the order of one new Alu insertion every ∼20 births, despite …

The Impact Of Genetic Variations In Bipolar Disorder, Lee Edsall
The Impact Of Genetic Variations In Bipolar Disorder, Lee Edsall
Theses
Bipolar disorder is a devastating illness that affects the quality of life for millions of Americans. The current diagnostic system depends on an extremely subjective interview and can frequently result in an incorrect diagnosis and ineffective treatment. An improved, biologically based, classification system requires a thorough understanding of the genetic basis of bipolar disorder. This understanding has been hampered by the difficulty in diagnosing patients and by the heterogeneity of the illness. The number of linkage analysis studies and lack of organization have also added to the challenges involved in understanding the biological basis of the disorder. The Bipolar Disorder …
Identifying The Largest Complete Data Set From Alfred, Mohamed Uduman
Identifying The Largest Complete Data Set From Alfred, Mohamed Uduman
Theses
ALFRED is a central and curated repository for allele frequency data for anthropologically defined human populations. To study and estimate the relationships and similarities between populations, researchers require a large and complete data set. However, the data set within ALFRED is not complete. Specifically, not all the populations in the database have been typed for all the polymorphisms. Mining ALFRED for the largest complete data set is equivalent to the 'Maximal Biclique' problem in graph theory. This is proven to be NP-Complete and no single algorithm can find the perfect solution in polynomial time. This project describes a heuristic (Largest …

Bacterial Cell Biology: Managing Magnetosomes, Craig Stephens
Biology
Sensing of magnetic fields by living organisms — magnetosensing — is best understood in magnetotactic bacteria. Recently work has provided new insight into the biogenesis of bacterial magnetosomes, and links these organelles to a newly recognized prokaryotic cytoskeletal filament which organizes magnetosomes into a sensory structure capable of aligning cells with the geomagnetic field.

The emergence of new genes and functions is of central importance to the evolution of species. The contribution of various types of duplications to genetic innovation has been extensively investigated. Less understood is the creation of new genes by recycling of coding material from selfish mobile genetic elements. To investigate this process, we reconstructed the evolutionary history of SETMAR, a new primate chimeric gene resulting from fusion of a SET histone methyltransferase gene to the transposase gene of a mobile element. We show that the transposase gene was recruited as part of SETMAR 40-58 million years ago, after the insertion …

Pluri-differentiated human mesenchymal progenitor cells (MPCs) are isolated. A method isolates and purifies human mesenchymal progenitor cells from Dexter-type cultures for characterization of and uses, particularly therapeutic uses for such cells. Specifically, isolated MPCs can be used for diagnostic purposes, to enhance the engraftment of hematopoietic progenitor cells, enhance bone marrow transplantation, or aid in the treatment or prevention of graft versus host disease.
