New Spiked-In Probe Sets For The Affymetrix Hgu-133a Latin Square Experiment, Monnie Mcgee, Zhongxue Chen

The Affymetrix HGU-133A spike in data set has been used for determining the sensitivity and specificity of various methods for the analysis of microarray data. We show that there are 22 additional probe sets that detect spike in RNAs that should be considered as spike in probe sets. We assign each proposed spiked-in probe set to a concentration group within the Latin Square design, and examine the effects of the additional spiked-in probe sets on assessing the accuracy of analysis methods currently in use. Implicit family process rules refer to unspoken rules about family beliefs and expectations about communication, closeness, and organization and are an important dimension of family functioning that may have a powerful influence over adolescent psychological well being. This study focused on the relationship between implicit family process rules and adolescent psychological symptoms such as: Hostility, interpersonal sensitivity, depression, anxiety, and somatization. It was hypothesized that more facilitative implicit family process rules would be related to fewer adolescent psychological symptoms. Network structures based upon metal-organic backbones represent a new class of functional materials that can be rationally constructed by employing the concepts of supramolecular chemistry and crystal engineering. The modularity of design strategies, the diversity of prototypal structures, and the dynamic features of networks have afforded great advantages over traditional materials syntheses. The research presented in this thesis is primarily concerned with developing an in-depth understanding of the basic principles that govern the supramolecular behaviors of metal-organic networks and gaining an experimental control over the structure and function of these new classes of hybrid materials.

BACKGROUND: Clostridium perfringens type A food poisoning is caused by enterotoxigenic C. perfringens type A isolates that typically possess high spore heat-resistance. The molecular basis for C. perfringens spore heat-resistance remains unknown. In the current study, we investigated the role of small, acid-soluble spore proteins (SASPs) in heat-resistance of spores produced by C. perfringens food poisoning isolates.

RESULTS: Our current study demonstrated the presence of all three SASP-encoding genes (ssp1, 2 and 3) in five surveyed C. perfringens clinical food poisoning isolates. beta-Glucuronidase assay showed that these ssp genes are expressed specifically during sporulation. This study utilized paired tones at 1000 Hz with a 500 ms interpair interval and a 10 s interstimulus interval to assess sensory gating. Forty-two participants, ranging from 3-72 years of age were used to observe maturational changes in amplitude, latency, and suppression ratios of the P50 waveform. Previous research has shown that in normal adults the amplitude in response to the second of the paired tones is significantly suppressed compared to the amplitude in response to the first tone. The current study showed amplitude decreased with age to middle adulthood, where it increased slightly to later adulthood. Latencies decreased …

Although the differential flow of metabolites through alternate pathways of intermediary metabolism is thought to be an important functional cause of life-history trade-offs, this phenomenon remains understudied. Using a radiolabeled amino acid, we quantified genetic differences in in vivo amino acid metabolism between morphs of the wing-polymorphic cricket Gryllus firmus that trade off early-age reproduction and dispersal capability. Lines selected for the flight-capable morph, which delays reproduction, oxidized a greater proportion of radiolabeled glycine and converted a greater amount into somatic lipid, mainly triglyceride (flight fuel). By contrast, lines selected for the flightless, reproductive morph converted a substantially greater proportion …

Rabies is a fatal zoonotic disease of mammals; it exacerbates the uncertainty of conserving populations of some threatened mammals (TM). Modeling affords an inexpensive, a priori way of studying key parameters of wildlife rabies transmission, rabies management economics, and TM conservation. Numerous models of rabies transmission have been published. Linear density dependent models predicted that a threshold density (KT 6 1.0), possibly attained by culling or contraception, would eliminate an epizootic through reduced contacts among host animals. Density independent models predicted less advantage of culling and contraception in rabies control due to limited contacts among territorial host animals. Recent stochastic, …

This note/essay examines the evidence on the effect of stronger IP laws introduced during the process of international IP law harmonization initiated by the TRIPS agreement, on the economic development of developing countries. It has been argued by proponents of harmonization that stronger IP laws will provide a needed boost to the economic development of developing (and even least-developed) countries. Critics of harmonization have argued that stronger IP laws will have the opposite effect. What has been largely overlooked in this debate is the strength of the evidentiary foundation upon which the arguments of both sides depend. Many of the …

The Demanding Community: Politicization Of The Individual After Dewey, Matthew C. Flamm

Education and Culture

This article argues that conceptions of community after Dewey despair of an institutional means of recovering individuality, which is the central problem of democracy. They so despair, I contend, because of their politicized view of the individual. I first briefly consider the contrast between Dewey and contemporary proceduralists and civic republicans, before turning to my central discussion: C. Wright Mills, whose critique indicates a historical watershed for Dewey’s view of community. Ultimately, despair of a Deweyan sense of community issues in a contemporary stalemate between what I identify as the political “activist” and “apathist.”

Motivation: Array CGH technologies enable the simultaneous measurement of DNA copy number for thousands of sites on a genome. We developed the circular binary segmentation (CBS) algorithm to divide the genome into regions of equal copy number (Olshen {\it et~al}, 2004). The algorithm tests for change-points using a maximal $t$-statistic with a permutation reference distribution to obtain the corresponding $p$-value. The number of computations required for the maximal test statistic is $O(N^2),$ where $N$ is the number of markers. This makes the full permutation approach computationally prohibitive for the newer arrays that contain tens of thousands markers and highlights the …

Unusually cold seawater temperatures were observed along much of the U.S. eastern seaboard during the summer of 2003. In this study, hydrographic and atmospheric observations from spring through summer were analyzed to track the evolution of the cold water event in the South Atlantic Bight (SAB) and investigate links to various forcing mechanisms. The hydrographic observations included 13 cross‐shelf transects over the central region of the SAB, surface temperature time series from several NDBC stations, and bottom temperatures from a mid shelf mooring. In vivo, endothelial cells are constantly exposed to pulsatile shear and tensile stresses. The main aim of this study was to design and build a physiological simulator, which reproduced homogenous strain profiles of the tensile strain experienced in vivo, and to investigate the effect of this cyclic tensile strain on the cell morphology, cell orientation and protein expression of endothelial cells. The biological response of human umbilical vein endothelial cells to a uniaxial cyclic stretch, in this newly developed simulator, was examined experimentally using immunohistostaining and confocal imaging and it was

2-Bromoisobutyryl bromide was immobilized on poly(methyl methacrylate) (PMMA) substrates activated using an oxygen plasma. Atom-transfer radical polymerization was then performed to graft poly(ethylene glycol) (PEG) on the PMMA surface. PMMA micro capillary electrophoresis (µCE) devices made with the covalently modified surfaces exhibited substantially reduced electroosmotic flow and nonspecific adsorption of proteins. Both column efficiency and migration time reproducibility were one order of magnitude better with derivatized PMMA µCE devices compared to untreated versions. Fast, reproducible, and efficient separations of proteins and peptides were demonstrated using the PEG-grafted PMMA µCE chips. All analyses were completed in less than 60 seconds, and …
