Selenoproteins In Parasites, Gustavo Salinas, Alexei V. Lobanov, Vadim N. Gladyshev Jun 2006

Selenoproteins In Parasites, Gustavo Salinas, Alexei V. Lobanov, Vadim N. Gladyshev

Vadim Gladyshev Publications

Parasites, which cause an enormous burden in the population of the third world, are a diverse group of organisms, many of which are sensitive to oxidative stress imposed by their hosts. In recent years, several selenoprotein families, some with antioxidant properties, have been described and characterized in metazoan parasites. Glutathione peroxidase and thioredoxin glutathione reductase (TGR) appear to be essential selenoproteins in flatworms (phylum Platyhelminthes). TGR is the single enzyme that provides reducing equivalents to both thioredoxin and glutathione pathways, in contrast to hosts, which evolve parallel pathways. In roundworms (phylum Nematoda), selenoproteins have recently been described, revealing …

The 15-Kda Selenoprotein (Sep15): Functional Analysis And Role In Cancer, Vyacheslav M. Labunskyy, Vadim N. Gladyshev, Dolph L. Hatfield

Vadim Gladyshev Publications

The 15-kDa selenoprotein (Sep15) was identified several years ago as a protein of unknown function. In recent years, several lines of evidence implicated Sepl5 in the effect of dietary selenium in cancer prevention. These lines of evidence include: 1) protein expression patterns in normal and malignant cells; 2) identification of polymorphic sites that regulate Sep15 levels and differentially respond to selenium supplementation; 3) location of the Sep15 gene in the human genome; and 4) correlation between Sep15 haplotype and susceptibility to cancer. Functional analyses revealed a specific interaction between Sep15 and a protein folding sensor in the endoplasmic reticulum of …

Selenocysteine (Sec) tRNA occupies a prominent position in the expression of selenoproteins as it is essential for their synthesis and it provides the means by which selenium is co-translationally inserted into protein as the amino acid, Sec. Thus, Sec tRNA is regarded as the principle constituent in selenoprotein synthesis. Many features unique to this tRNA have been characterized over the years in mammals and other eukaryotes. In the last five years, the major advances have been in an elucidation of the different roles that the two major Sec tRNA isoforms play in selenoprotein biosynthesis and in Sec biosynthesis. One isoform …

Perspective Greenhouse Gas Mitigation By Covers On Livestock Slurry Tanks And Lagoons?, Soren O. Petersen, Daniel N. Miller Jun 2006

Perspective Greenhouse Gas Mitigation By Covers On Livestock Slurry Tanks And Lagoons?, Soren O. Petersen, Daniel N. Miller

Department of Animal Science: Faculty Publications

Liquid manure (slurry) storage facilities are important point sources of atmospheric pollution. Being point sources, containment of gaseous emissions via improved storage conditions may be possible, and permeable surface covers (natural crusts and artificial covers) are increasingly recognized for their capacity to reduce various gaseous emissions. Microbial transformations in permeable surface covers include bacterial-methane oxidation, but this interface between nitrogen-rich slurry and the atmosphere is also an environment with intense nitrogen turnover that can lead to nitrous oxide emissions. Both methane and nitrous oxide are greenhouse gases, and strategies to reduce environmental impact of slurry stores must consider the total …

Simple water-soluble lanthanum and europium complexes are effective at detecting neutral sugars as well as glycolipids and phospholipids. In solutions at physiologically relevant pH the fluorescent lanthanum complex binds neutral sugars with apparent binding constants comparable to those of arylboronic acids. Interference from commonly occurring anions is minimal. The europium complex detects sialic acid-containing gangliosides at pH 7.0 over an asialoganglioside. This selectivity is attributed, in large part, to the cooperative complexation of the oligosaccharide and sialic acid residues to the metal center, based on analogous prior studies. In MeOH, lysophosphatidic acid (LPA), a biomarker for several pathological conditions including …

Is A Shutdown Of The Thermohaline Circulation Irreversible?, Jianjun Yin, Michael E. Schlesinger, Natasha G. Andronova, Sergey Malyshev, Bin Li

Faculty Publications

The Atlantic thermohaline circulation (THC) plays a vital role in explaining past abrupt climate changes and in maintaining the current climate. Its remarkable nonlinear dynamics, first demonstrated by H. M. Stommel, have been supported by various types of climate models. This has led to severe concerns that global warming may shut down the THC irreversibly, with consequent catastrophic climate changes, particularly for Europe. Here we use an uncoupled ocean general circulation model (OGCM) and a coupled atmosphere-ocean general circulation model (AOGCM) to investigate the nonlinear response of the THC to freshwater perturbations in the northern North Atlantic. We find that …

Implications For Cognitive Quantum Computation And Decoherence Limits In The Presence Of Large Extra Dimensions, Jonas R. Mureika Jun 2006

Implications For Cognitive Quantum Computation And Decoherence Limits In The Presence Of Large Extra Dimensions, Jonas R. Mureika

Physics Faculty Works

An interdisciplinary physical theory of emergent consciousness has previously been proposed, stemming from quantum computation-like behavior between 109 or more entangled molecular qubit states (microtubulin). This model relies on the Penrose-Diósi gravity-driven wavefunction collapse framework, and thus is subject to any secondary classical and quantum gravity effects. Specifically, if large extra spatial dimensions exist in the Universe, then the resulting corrections to Newtonian gravity cause this model to suffer serious difficulties. It is shown that if the extra dimensions are larger than 100 fm in size, then this model of consciousness is unphysical. If the dimensions are on the …

Genetic Code Based Coding And Mathematical Formulation For Dna Computation, Mingjun Zhang, Maggie Xiaoyan Cheng, Tzyh-Jong Tarn Jun 2006

Genetic Code Based Coding And Mathematical Formulation For Dna Computation, Mingjun Zhang, Maggie Xiaoyan Cheng, Tzyh-Jong Tarn

Computer Science Faculty Research & Creative Works

DNA computation is to use DNA molecules for information storing and processing. Challenges currently faced by DNA computation are (1) lack of theoretical computational models for applications, and (2) high error rate for implementation. This paper attempts to address these problems from genetic coding and mathematical modeling aspects. The proposed genetic coding approach provides a promising alternative to reduce high error rate. The mathematical formulation lays down groundwork for studying theoretical aspects of DNA computation.

Anadromous Rainbow Smelt And Tomcod In Connecticut: Assessment Of Populations, Conservation Status, And Need For Restoration Plan, Heather A. Fried, Eric T. Schultz

EEB Articles

(beginning of rainbow smelt executive summary)

Evidence indicates that anadromous rainbow smelt (Osmerus mordax) populations in Connecticut and elsewhere in the northeast United States have severely declined. Several sampling programs have documented declines in Connecticut’s smelt populations over the last three decades (Marcy 1976a, Marcy 1976b, Millstone Environmental Laboratory 2005). Similar declines have also been documented in the Hudson River (ASA Analysis & Communication 2005) and in Massachusetts (personal communication, Brad Chase, MA Division of Marine Fisheries 2004). Recreational and commercial fisheries in the region for this species have virtually ceased (Blake and Smith 1984). The Connecticut Fish Advisory Committee …

Patterns Of Habitat Use By Primates In Eastern Ecuador, Seema Nayan Sheth Jun 2006

Patterns Of Habitat Use By Primates In Eastern Ecuador, Seema Nayan Sheth

Theses

Lowland tropical rain forests of western Amazonia are characterized by the most speciose primate communities in the Neotropics, immediately leading to the question of to what extent does niche partitioning by primate species serve as a mechanism to promote species co-existence. Because the primate assemblages that we observe today reflect a combination of ecological and evolutionary processes, this study examines habitat occupancy and its relationship to phylogeny and space in a diverse diurnal primate community in an undisturbed lowland rain forest of Amazonian Ecuador. Specifically, the following null hypotheses are explored as potential factors that shape community structure: (1) mean …

Multivariate Analysis And Visualization Of Splicing Correlations In Single-Gene Transcriptomes, Mark C. Emerick, Giovanni Parmigiani, William S. Agnew Jun 2006

Multivariate Analysis And Visualization Of Splicing Correlations In Single-Gene Transcriptomes, Mark C. Emerick, Giovanni Parmigiani, William S. Agnew

Johns Hopkins University, Dept. of Biostatistics Working Papers

Through ‘combinatorial splicing’, RNA metabolism may create enormous structural diversity in the proteome. Functional interactions among multiple alternative domains can have a disproportionate impact on the phenotype, requiring integrated RNA-level regulation of molecular composition. Splicing correlations within molecules expressed from a single gene, where these effects would be greatest, provide valuable clues to functional relationships and targets for splicing regulation. We present tools to visualize complex splicing patterns in full-length cDNA libraries. Developmental changes in pair-wise correlations are presented vectorially in ‘clock plots’ and linkage grids. Higher-order correlations are assessed via a loglinear model and Monte Carlo analysis with an …

The Environment Transport Of Radium And Plutonium: A Review By Brice Smith And Alexandra Amonette, Institute For Energy And Environmental Research (Ieer), Brice Smith, Alexandra Amonette

Institute for Energy and Environmental Research (IEER)

In this report we will provide a brief review of the environmental transport of two specific radionuclides. In Chapter Two we will consider the mobility of radium. This naturally occurring radionuclide is part of the uranium and thorium decay series, and is thus a potential concern in many areas where these elements have been mined or processed. In addition to the large number of sites with radium bearing waste, we chose to focus on this radionuclide in part due to the high concentrations of radium-226, and its thorium-230 parent, in the raffinate waste from the former Fernald Feed Material Production …

The Importance Of The Strictly Conserved, C-Terminal Glycine Residue In Phosphoenolpyruvate Carboxylase For Overall Catalysis: Mutagenesis And Truncation Of Gly-961 In The Sorghum C4 Leaf Isoform, Wenxin Xu, Shaheen Ahmed, Hideaki Moriyama, Raymond Chollet

Hideaki Moriyama Publications

Phosphoenolpyruvate carboxylase (PEPC) is a “multifaceted,” allosteric enzyme involved in C4 acid metabolism in green plants/microalgae and prokaryotes. Before the elucidation of the three-dimensional structures of maize C4 leaf and Escherichia coli PEPC, our truncation analysis of the sorghum C4 homologue revealed important roles for the enzyme’s C-terminal α-helix and its appended QNTG961 tetrapeptide in polypeptide stability and overall catalysis, respectively. Collectively, these functional and structural observations implicate the importance of the PEPC C-terminal tetrapeptide for both catalysis and negative allosteric regulation. We have now more finely dissected this element of PEPC structure-function by modification of the absolutely conserved …
