Dna-Templated Surface Alignment And Characterization Of Carbon Nanotubes., Huijun Xin

Carbon nanotubes are appealing materials for nanofabrication due to their unique properties and structures. However, for carbon nanotubes to be used in mass-fabricated devices, precise control of nanotube orientation and location on surfaces is critical. I have developed a technique to align single-walled carbon nanotubes (SWNTs) on surfaces from a droplet of nanotube suspension under gas flow. Fluid motion studies indicate that alignment is likely due to circulation of SWNTs in the droplet. My work provides a facile method for generating oriented nanotubes for nanodevice applications. I have also devised an approach for localizing SWNTs onto 1-pyrenemethylamine-decorated DNA on surfaces. …

Alpha-thrombin has two separate electropositive binding exosites (anion binding exosite I, ABE-I and anion binding exosite II, ABE-II) that are involved in substrate tethering necessary for efficient catalysis. Alpha-thrombin catalyzes the activation of factor V and factor VIII following discrete proteolytic cleavages. Requirement for both anion binding exosites of the enzyme has been suggested for the activation of both procofactors by alpha-thrombin. We have used plasma-derived alpha-thrombin, beta-thrombin (a thrombin molecule that has only ABE-II available), and a recombinant prothrombin molecule rMZ-II (R155A/R284A/R271A) that can only be cleaved at Arg(320) (resulting in an enzymatically active molecule that has only ABE-I …

Most ecological hypotheses about species coexistence hinge on species differences, but quantifying trait differences across species in diverse communities is often unfeasible. We examined the variation of demographic traits using a global tropical forest data set covering 4500 species in 10 large-scale tree inventories. With a hierarchical Bayesian approach, we quantified the distribution Of mortality and growth rates of all tree species at each site. This allowed us to test the prediction that demographic differences facilitate species richness, as suggested by the theory that a tradeoff between high growth and high survival allows species to coexist. Evolutionary Genetics Of Juvenile Hormone And Ecdysteroid Regulation In Gryllus: A Case Study In The Microevolution Of Endocrine Regulation, Anthony J. Zera

During the past 15 years the first detailed synthesis of endocrinology and population genetics has begun, in which natural genetic variations for endocrine regulators have been characterized, almost exclusively in species of the cricket genus Gryllus. Artificial selection studies have documented that regulators of the juvenile hormone titer can rapidly evolve and exhibit levels of genetic variability similar to other physiological traits. Strong genetic correlations exist between some but not all regulators of the JH titer during the juvenile stage. No genetic correlation exists between regulators functioning in juvenile and adult stages, and thus, endocrine regulation can evolve independently in …

Vengeance is generally accompanied by the moral emotion of resentment and indignation, which are also natural psychological reactions. We can and do give these emotions cognitive content, inasmuch as they have developed and matured over time with culture, but they are primitive. They arise when an individual suffers a non-trivial injury that was inflicted without excuse or justification. Among other injuries suffered, the harm done discounts the value we hold of ourselves as human beings, so that when this discounting (the crime or a substantial tort) occurs and we react defensively; our worth as an individual feels threatened. Evaluating Causal Effect Predictiveness Of Candidate Surrogate Endpoints, Peter B. Gilbert, Michael Hudgens

UW Biostatistics Working Paper Series

Most methods for evaluating surrogate endpoints measure validity in terms of net effects (i.e., treatment effects adjusted for the biomarker measured after randomization). Frangakis and Rubin (2002, Biometrics) criticized these approaches because net effects may reflect selection bias, and suggested an alternative definition of a surrogate endpoint (a "principal" surrogate) based on causal effects. For evaluating principal surrogates we introduce a causal effect predictiveness (CEP) surface, which quantifies how well causal treatment effects on the biomarker predict causal treatment effects on the clinical endpoint. The CEP surface is not identifiable in general due to missing potential outcomes. However, by incorporating …

Electrospray ionization (ESI) has become one of the most commonly used ionization techniques for mass spectrometry (MS) and ion mobility spectrometry (IMS), and efforts continue to improve its performance. ESI-MS is most recognized for its wide application to biomacromolecules where high sensitivity is of paramount importance. However, the major limitation in sensitivity with ESI-MS is due to its low ion transmission efficiency from the ESI source into the sampling orifice and through any stages utilized for transfer of ions from atmosphere to vacuum in the MS. Electric field gradient focusing (EFGF) utilizes a hydrodynamic flow and an electric field gradient to focus and concentrate charged analytes and order them in a separation channel according to electrophoretic mobility. Elution can be achieved by decreasing the applied voltage or increasing the hydrodynamic flow. EFGF has the advantages of concentrating a large volume (100 micro-L) of target proteins without significant band broadening and simultaneously removing unwanted components from the sample. Two types of EFGF devices have been investigated to concentrate and separate proteins: a fiber-based EFGF device and a hydrogel-based EFGF device. Using fiber-based EFGF with UV detection, a …

Understanding the dynamics of coastal marine communities represents a substantial challenge, and one that is actively pursued globally. Within the United States, several sites have been designated as National Estuarine Research Reserves (NERR) with the idea that concentrated research at these sites will lead to greater understanding of the ecosystem. The Great Bay Estuary of New Hampshire is one of these sites. A wide spectrum of research is conducted within the Great Bay, and substantial financial support is committed to that research on an annual basis. Markov Approximations: The Characterization Of Undermodeling Errors, Lei Lei

This thesis is concerned with characterizing the quality of Hidden Markov modeling when learning from limited data. It introduces a new perspective on different sources of errors to describe the impact of undermodeling. Our view is that modeling errors can be decomposed into two primary sources of errors: the approximation error and the estimation error. This thesis takes a first step towards exploring the approximation error of low order HMMs that best approximate the true system of a HMM. We introduce the notion minimality and show that best approximations of the true system with complexity greater or equal to the …
