Arts & Sciences Faculty Publications

The renin‐angiotensin‐aldosterone system (RAAS) appears to contribute significantly to osmoregulation of fasting northern elephant seal (Mirounga angustirostris) pups; however, RAAS has not been characterized in fasting adult seals. Therefore, this study examined the contribution of RAAS to water turnover rates in fasting adult male northern elephant seals. Blood samples were obtained twice during their breeding fast at an interval of 6.5 wk, and water efflux rate was estimated by isotopic dilution during the same period. Serum electrolytes (Na+, K+, Cl−) and osmolality were unaltered between the two sampling periods, indicating ionic and …

Novel Cancer Therapeutics, The Generation Of Ros, And Cell Survival, Clint Mitchell Jan 2006

Novel Cancer Therapeutics, The Generation Of Ros, And Cell Survival, Clint Mitchell

Theses and Dissertations

The impact of Ad.mda-7 on the survival of renal cell carcinoma lines (RCC), primary renal epithelial cells, glioblastoma multiforme lines (GBM), and primary rodent astrocytes is unknown. The present studies examine whether the GST fusion protein, GST-MDA-7, and the adenovirus, Ad.mda-7, altered the growth and survival of the A498 and UOK121N RCC lines or radiosensitized GBM, respectively. Due to previous findings that the RCC lines, but not primary renal epithelial cells, were resistant to type 5 adenoviral infection, we used purified GST-MDA-7 protein to show that GST-MDA-7, but not GST, caused a dose-dependent reduction in A498 and UOK121N proliferation but …

Degenerate Oligonucleotide Primed - Polymerase Chain Reaction Evaluation And Optimization To Improve Downstream Forensic Str Analysis Of Low Quality/Low Quantity Dna, Lindsay Paige Thompson Jan 2006

Degenerate Oligonucleotide Primed - Polymerase Chain Reaction Evaluation And Optimization To Improve Downstream Forensic Str Analysis Of Low Quality/Low Quantity Dna, Lindsay Paige Thompson

Theses and Dissertations

When forensic biological samples yield low quality/low quantity DNA, thecurrent STR analysis methods do not generate acceptable profiles. Whole genomeamplification can be used to pre-amplify the entire genome for downstream analyses. A commercially available kit for DOP-PCR, a form of WGA, is currently being used in the clinical for downstream single locus targets. Forensic analyses utilize a multiplex amplification. This study determined that the "home brew" created by our lab performs the same as the commercially available kit. Future optimization studies of DOP-PCR can utilize this "home brew". Additionally, this research determined that a 10 second increase in electrokinetic injection …

Designing Computer Agents With Facial Personality To Improve Human-Machine Collaboration, Brian Esley Tidball

Browse all Theses and Dissertations

The development of computer agents to enhance human-computer interfaces is an evolving field of study. This study examined whether people perceive personality in static digital faces that portray expressions of emotion, and if the digital faces would influence human performance on a simple human-machine collaborative task. The first experiment measured user-perception of personality, based on the emotional expression in two sets of five static digital faces. The results from this first phase revealed that participants provided different ratings, of the Big-Five personality model sub-traits, based on the emotional expression of a static digital face. This indicates a perception of personality …

Digital Channelized Wide Band Receiver Implemented With A Systolic Array Of Multi-Rate Fir Filters, David M. Rodney

Browse all Theses and Dissertations

This thesis proposes the use of a Systolic Array of Multi-Rate FIR Filters to improve performance by eliminating the requirement of the FFT and De-Multiplexer associated with the conventional receiver while achieving the same functionality. The FFT is a major bottle neck for improving system performance for the conventional DCWBR because many complex multiplications and additions are required. The proposed new architecture is designed and evaluated in MATLAB to illustrate its viability. Two approaches for improved channel arbitration are accessed in MATLAB, namely, channel bin’s rms comparison and parallelism of the Systolic Array Multi-Rate FIR Filters. The FIR filters (high-pass …

Browse all Theses and Dissertations

The Global Positioning System or GPS is a satellite based technology that has gained widespread use worldwide in civilian and military applications. Direct Sequence Spread spectrum (DSSS) is the method whereby the data transmitted by the satellite and received by user is kept secure, low power and relatively noise-immune. The first step required in the GPS operation is to perform a lock on the incoming signal, both with respect to time synchronization and frequency resolution. Because of the need for reduced time to lock and also reduced hardware, algorithms based in the frequency domain have been developed. These algorithms take …

The Relationship Between Lactic Acid, Reactive Oxygen Species And The Hypoxia-Induced Acidification Seen In Chemosensitive Neurons Of The Nucleus Tractus Solitarius (Nts), Trevor Downing

Browse all Theses and Dissertations

The NTS is one of many sites of chemoreception meaning that it responds to changes in CO2 and pH. NTS neurons also acidify approximately 0.13 pH units in response to hypoxia. Experiments involved brainstem slices exposed to a control hypoxic bout followed by hypoxia in conjunction with different drugs. The drugs included fluorocitrate, 4-hydroxycinnamate, oxygen-glucose deprivation and iodoacetate. Iodoacetate produced the strongest blunting of the hypoxia-induced acidification and decreased the response by ~53%. Results from the studies using the superoxide probe DHE showed an increase in the levels of ROS during the hypoxic exposure prior to reoxygenation. Two ROS scavengers …

Electronic Properties Of Silicon-Based Nanostructures, Gian Giacomo Guzman-Verri

Browse all Theses and Dissertations

We have developed a new unifying tight-binding theory that can account for the electronic properties of recently proposed Si-based nanostructures, namely, Si graphene-like sheets and Si nanotubes. We considered the sp2s* and sp3 models up to first- and second-nearest neighbors, respectively. Our results show that the corresponding Si nanotubes follow the so-called Hamada's rule. Comparison to a recent ab initio calculation is made.

Over the past two decades, much research has been done in the field of human immunodeficiency virus (HIV) and acquired immunodeficiency syndrome (AIDS). However, many of the aspects of pathogenesis of HIV infection and its persistence in the body, despite treatment, remain a mystery. Recent evidence suggests that HIV positive patients develop eosinophilia, especially in the later stages of infection and AIDS. Eosinophils are CD4 positive cells that have the potential to be infected by HIV. Studies have shown that an eosinophilic cell line, AML14.3D10, can be productively infected with a T-cell tropic, CXCR4-using (X4) strain of HIV-1. In this …

This Master thesis considers the use of formal languages representation for image processing, especially texture patterns and determining texture paths in them. The texture paths are detected and extracted by using image processing techniques, such as image segmentation to isolate regions of interest, and then the extraction of repeating textures. The detection of the texture blocks is obtained by recursively using 27 X 27, 9 X 9, and 3 X 3 windows. Predefined repeating texture patterns are also searched for in each of the sets. For each set of texture blocks with similar or same characteristics, curve fitting techniques are …

Escherichia coli (E. coli) is a gram negative bacterium commonly found as a commensal in the intestinal tract of humans and other warm blooded animals. The commensal strains of E. coli are non-pathogenic and do not cause an infection in the host. However, some strains of E. coli are pathogenic and can cause several diseases in humans that include neonatal meningitis, intestinal infections and urinary tract infections. Almost 80-85% of the uncomplicated urinary tract infections are caused by uropathogenic E. coli. Our project involved the characterization of 12 strains of E. coli, isolated from humans in health and disease. These …

Mutant screening and genetic selection in various organisms have shown that residues far from the active site of ribulose-1,5-bisphosphate carboxylase/oxygenase (RubisCO) can influence catalytic efficiency and CO2/O2 specificity. Because RubisCO catalyzes the rate-limiting step of photosynthesis, further study of these sites distant from the primary reaction center may provide the necessary information for engineering an increase in primary productivity of crop plants. In a previously described system of random mutagenesis and bioselection (Smith, 2002), the RubisCO genes from the cyanobacterium Synechococcus PCC6301, were randomly mutated and introduced into the photosynthetic bacterium Rhodobacter capsulatus. An A47T substitution resulted in a very …

Pyrrole Carboxamides As Potential Carbonic Anhydrase Inhibitors, Nishrin Ismailbhai Marketwala Jan 2006

Pyrrole Carboxamides As Potential Carbonic Anhydrase Inhibitors, Nishrin Ismailbhai Marketwala

Browse all Theses and Dissertations

Carbonic anhydrases (CAs) are metalloenzymes that catalyze the reversible interconversion of CO2 and bicarbonate. We recently initiated a screen of a broad range of indoles and pyrroles possessing acyl-, carboxyl-, amido-, sulfonyl-, sulfamyl- and oxime- functionalities as potential inhibitors of á-CA II isozyme. As slight inhibition was observed in the case of 2-carboxamido-1-(phenylsulfonyl)pyrrole, and aware that pyrrole carboxamide derived sulfonamides had been previously demonstrated to possess CA inhibition, we sought to prepare a series of 1-(phenylsulfonyl)pyrrole-2-carboxamides but lacking a sulfonamide moiety to ascertain if the activity in our case might be due to a hitherto unrecognized pyrrole pharmacophore. To this …

The Modeling Of Solubility, Frank Christopher Campanell

Browse all Theses and Dissertations

In this work the solubilities of gases in liquids and liquids in liquids were modeled using both physical properties and topological descriptors of the solutes. Quantitative structure-activity relationship (QSAR) methods were employed to create single-linear regression (SLR) and multiple-linear regression (MLR) models of the solubilities. Factor analysis was employed to determine the number of significant factors present in the solubilities. The solubilities of monoalcohols in water, halogenated alkanes in water, gases in water, gases in alkanes, and gases in alcohols were examined and modeled.

Diatom Paleolimnological Record Of Holocene Climatic And Environmental Change In The Altai Mountains, Siberia, Karlyn S. Westover, Sherilyn C. Fritz, Tatyana A. Blyakharchuk, Herbert E. Wright

Department of Earth and Atmospheric Sciences: Faculty Publications

The sedimentary diatom records of three shallow lakes in the Altai Mountains, southern Siberia, were examined to assess the nature and timing of Holocene environmental changes. Few paleoenvironmental records, especially reconstructions not based on pollen, have been reported from this region. The lakes differ in elevation, annual precipitation, and catchment vegetation. Diatom assemblages in all lakes were dominated for the entire period of record by small benthic species of Pseudostaurosira Williams & Round, Staurosira Ehrenberg, and Staurosirella Williams & Round. Planktonic taxa only occur in very low abundances (<5%). The most diverse diatom flora was found in Dzhangyskol, which is situated at the lowest elevation within a forested catchment. A lack of detailed information on the ecological preferences of the dominant taxa and the complexity of environmental drivers make direct interpretation of the diatom record difficult. However, other proxies suggest that dramatic shifts in dominance between Staurosira elliptica and Staurosirella pinnata in Grusha Ozero reflect millennial-scale variability …

Many of the Indiana Department of Transportation (INDOT) sites are contaminated with aromatic hydrocarbons and chlorinated solvents; these contaminants often occur in the form of light non-aqueous phase liquids (LNAPLs) or dense non-aqueous phase liquids (DNAPLs). Considerable effort has recently been focused on developing in-situ technologies for removing or destroying NAPL source zones, and several potentially viable methods have emerged including cosolvent-enhanced source zone removal. Most recently, site-specific remedial designs involving a combination of in-situ methods (often referred to as a treatment train) have been proposed for which data are still sparse. The primary goal of the laboratory experiments is …

Remediation Of Aromatic Hydrocarbons In Low Permeability Soils: Updating The Remediation Decision Tree (Synthesis Study), Inez Hua, Andy Hopf

JTRP Technical Reports

Because of the large number of technologies for in situ remediation, the very different types of contaminants to which these technologies are applicable, and the wide range of field conditions, it can be difficult to choose an optimal technology for a specific site. Sorting and prioritizing the various factors which contribute to the success of a particular clean-up can be daunting. Furthermore, non-technical factors, such as those in the legal, political, or financial realm, may also influence which technology is ultimately chosen. Most in situ treatment methods are effective in permeable soil. However, much of Indiana soil is low-permeability, so …
