Probing The Reaction Mechanism Of The D-Ala-D-Ala Dipeptidase, Vanx, By Using Stopped-Flow Kinetic And Rapid-Freeze Quench Epr Studies On The Co(Ii)-Substituted Enzyme, Megan L. Matthews, Gopalraj Periyannan, Christine E. Hajdin, Tara K. Sidgel, Brian Bennett, Michael W. Crowder

Physics Faculty Research and Publications

In an effort to probe the reaction mechanism of VanX, the D-ala-D-ala dipeptidase required for high-level vancomycin resistance in bacteria, stopped-flow kinetic and rapid-freeze quench EPR studies were conducted on the Co(II)-substituted enzyme when reacted with d-ala-d-ala. The intensity of the Co(II) ligand field band at 550 nm decreased (ε550 = 140 to 18 M-1 cm-1) when VanX was reacted with substrate, suggesting that the coordination number of the metal increases from 5 to 6 upon substrate binding. The stopped-flow trace was fitted to a kinetic mechanism that suggests the presence of an intermediate whose breakdown …

Fire Making Fuel: How A Surface Fire In A Primary Forest Affected The Availability Of Potential Fuel One Year Later, Alex Leckie Oct 2006

Fire Making Fuel: How A Surface Fire In A Primary Forest Affected The Availability Of Potential Fuel One Year Later, Alex Leckie

Independent Study Project (ISP) Collection

Fires affect millions of hectares of tropical forests around the world. These fires result in great environmental damage and economical losses. Many farmers are dependent on fire for managing their lands and many times their fires accidentally spread into forests via fuels on the forest floor. This study attempted to analyze and quantify the difference of potential fuel in a primary forest which burned one year before and an unburned part of the same forest at a primary forest fragment surrounded by farms and with a history of anthropogenic accidental fires burning it. This was done by making three sample …

Liberty Of Ecological Conscience, Aaron Lercher Oct 2006

Liberty Of Ecological Conscience, Aaron Lercher

Faculty Publications

Our concern for nonhuman nature can be justified in terms of a human right to liberty of ecological conscience. This right is analogous to the right to religious liberty, and is equally worthy of recognition as that fundamental liberty. The liberty of ecological conscience, like religious liberty, is a negative right against interference. Each ecological conscience supports a claim to protection of the parts of nonhuman nature that are current or potential sites of its active pursuit of natural value. If we acknowledge the fallibility of each conscience in its pursuit of genuine natural value, a policy of indefinitely extensive …

It addresses concurrency and active perception by modeling the world as concurrent schemas represented in Robot Schemas and implementing RS within Soar. The paper discusses the design issues involved in integrating cognition, real-time sensorimotor behavior, learning, and natural language.

Adaptive Estimation Of Autoregressive Models With Time-Varying Variances, Ke-Li Xu, Peter C.B. Phillips Oct 2006

Adaptive Estimation Of Autoregressive Models With Time-Varying Variances, Ke-Li Xu, Peter C.B. Phillips

Cowles Foundation Discussion Papers

Stable autoregressive models of known finite order are considered with martingale differences errors scaled by an unknown nonparametric time-varying function generating heterogeneity. An important special case involves structural change in the error variance, but in most practical cases the pattern of variance change over time is unknown and may involve shifts at unknown discrete points in time, continuous evolution or combinations of the two. This paper develops kernel-based estimators of the residual variances and associated adaptive least squares (ALS) estimators of the autoregressive coefficients. These are shown to be asymptotically efficient, having the same limit distribution as the infeasible generalized …

Simulating The Maximum Domain Wall Speed In A Magnetic Nanowire, Andrew Kunz Oct 2006

Simulating The Maximum Domain Wall Speed In A Magnetic Nanowire, Andrew Kunz

Physics Faculty Research and Publications

The dynamics of domain wall motion in permalloy nanowires have been simulated utilizing the Landau-Lifshitz-Gilbert (LLG) equation of motion. The simulation results are presented in terms of the domain wall speed for ranges of the Gilbert damping parameter alpha and nanowire width. The maximum domain wall speed is independent of alpha. The speed of the domain wall can be increased by increasing the nanowire width, but this lowers the critical field. For applied fields below the critical field, the wall moves uniformly along the wire and the speed of the wall increases with increases in the driving field. This behavior …

The Effects Of Tgf-Beta1 And Igf-I On The Biomechanics And Cytoskeleton Of Single Chondrocytes, Nic Leipzig Oct 2006

The Effects Of Tgf-Beta1 And Igf-I On The Biomechanics And Cytoskeleton Of Single Chondrocytes, Nic Leipzig

Chemical, Biomolecular, and Corrosion Engineering Faculty Research

Objective Ascertaining how mechanical forces and growth factors mediate normal and pathologic processes in single chondrocytes can aid in developing strategies for the repair and replacement of articular cartilage destroyed by injury or disease. This study examined effects of transforming growth factor-β1 (TGF-β1) and insulin-like growth factor-I (IGF-I) on the biomechanics and cytoskeleton of single zonal chondrocytes. Method Superficial and middle/deep bovine articular chondrocytes were seeded on tissue culture treated plastic for 3 and 18 h and treated with TGF-β1 (5 ng/mL), IGF-I (100 ng/mL), or a combination of TGF-β1 (5 ng/mL) + IGF-I (100 ng/mL). Single chondrocytes from all …

Monoreflections Of Archimedean ℓ-Groups, Regular Σ-Frames And Regular Lindelöf Frames, A. W. Hager, J. J. Madden Oct 2006

Monoreflections Of Archimedean ℓ-Groups, Regular Σ-Frames And Regular Lindelöf Frames, A. W. Hager, J. J. Madden

Faculty Publications

We prove that in the category of Archimedean lattice-ordered groups with weak unit there is no homomorphism-closed monoreflection strictly between the strongest essential monoreflection (the so-called "closure under countable composition") and the strongest monoreflection (the epicompletion). It follows that in the category of regular σ-frames, the only non-trivial monoreflective subcategory that is hereditary with respect to closed quotients consists of the boolean σ-algebras. Also, in the category of regular Lindelöf locales, there is only one non-trivial closed-hereditary epi-coreflection. The proof hinges on an elementary lemma about the kinds of discontinuities that are exhibited by the elements of a composition-closed l-group …

Support Properties And Holmgren's Uniqueness Theorem For Differential Operators With Hyperplane Singularities, Gestur Ólafsson, Angela Pasquale Oct 2006

Support Properties And Holmgren's Uniqueness Theorem For Differential Operators With Hyperplane Singularities, Gestur Ólafsson, Angela Pasquale

Faculty Publications

Let W be a finite Coxeter group acting linearly on Rn. In this article we study the support properties of a W-invariant partial differential operator D on Rn with real analytic coefficients. Our assumption is that the principal symbol of D has a special form, related to the root system corresponding to W. In particular the zeros of the principal symbol are supposed to be located on hyperplanes fixed by reflections in W. We show that conv (supp D f) = conv (supp f) holds for all compactly supported smooth functions f so that conv (supp f) is W-invariant. The …

Estimation Of The Rigid-Body Motion From Three-Dimensional Images Using A Generalized Center-Of-Mass Points Approach, B. Feng, P. P. Bruyant, P. H. Pretorius, R. D. Beach, H. C. Gifford, J. Dey, M. A. Gennert, M. A. King

Faculty Publications

We present an analytical method for the estimation of rigid-body motion in sets of three-dimensional (3-D) SPECT and PET slices. This method utilizes mathematically defined generalized center-of-mass points in images, requiring no segmentation. It can be applied to compensation of the rigid-body motion in both SPECT and PET, once a series of 3-D tomographic images are available. We generalized the formula for the center-of-mass to obtain a family of points comoving with the object's rigid-body motion. From the family of possible points we chose the best three points which resulted in the minimum root-mean-square difference between images as the generalized …
