The characterization of perphenazine and scopolamine aerosols generated using the capillary aerosol generator (CAG) was reported. Variables including steady state power, the formulation vehicle, the drug concentration and the formulation flow rate were studied for their effects on the chemical stability and particle size of these drug aerosols.Stability-indicating HPLC and LC-MS assays were developed and validated for perphenazine and scopolamine, respectively. The chemical stability of each compound was investigated under a variety of stress conditions and the structure of degradation products was proposed.Perphenazine aerosols were generated from propylene glycol (PG) formulations with concentrations of 9, 48 and 90mM at formulation …

Diffuse Brain Injury Triggers Ultra-Rapid Perisomatic Traumatic Axonal Injury, Wallerian Change, And Non-Specific Inflammatory Responses, Brian Joseph Kelley

A significant component of diffuse brain injury (DBI) is diffuse axonal injury (DAI) which is responsible for the morbidity and mortality associated with this condition. DAI and its experimental counterpart traumatic axonal injury (TAI) result in scattered microscopic pathology characterized by focal impairment of axonal transport leading to progressive swelling and delayed axotomy. DBI-mediated perisomatic axotomy does not result in acute neuronal death suggesting that delayed axotomy was responsible for this unanticipated response. To evaluate this hypothesis, we examined the spatiotemporal progression of DBI-mediated perisomatic TAI. LM / TEM identified impaired axonal transport within 15 - 30 min post-injury. Perisomatic …

CD23, the low affinity receptor for IgE, is expressed mainly on B cells and has been shown to regulate IgE production. Previously, recombinant mouse and human CD23 were constructed with a trimerizing isoleucine zipper motif attached in frame to the N-terminus of the entire extracellular CD23 (lz-ECCD23). The goal was to examine the role of the necessity of the CD23 stalk for binding IgE. Using PCR-based mutagenesis to delete the majority of the stalk, binding to IgE was lost. Further studies examined the effect of lz-ECCD23 in preventing IgE from binding FcεRI and therefore acting as a therapeutic agent. It …

The Wilms' tumor 1 gene is overexpressed in many types of cancer and is associated with poor prognosis and resistance to anti-cancer therapies. In vitro studies in non-glioma cells types have demonstrated that WTl plays a role in increased proliferation, resistance to apoptosis, and increased cellular invasion. We aimed to thoroughly characterize the expression pattern of Wilms' tumor 1 in human malignant glioma and discern its function in this complex disease process. Leukocytes are activated in women with preeclampsia, but the class of leukocyte most likely to cause vascular dysfunction is not known. We hypothesized that neutrophils may be the class of leukocyte most involved in causing this dysfunction because neutrophils are the most abundant of the leukocytes and their numbers increase during pregnancy. In this study we compared vascular infiltration of neutrophils (CD66b) with monocytes/macrophages (CD14) and lymphocytes (CD99) in normal non-pregnant women (NNP), normal pregnant women (NP), and preeclamptic women (PE). Convection-enhanced delivery (CED) is a proven method for targeted drug delivery to the brain that circumvents the blood-brain barrier (BBB). Little study has been conducted in understanding CED in pathological brain states. This is of importance when dealing with chemotherapeutic agent delivery to brain tumors, where vasogenic edema (VE) exists. The current study aims to characterize a model of VE suitable for studying CED.VE was produced in the right hemisphere of the rat brain using multiple infusions of hyperosmotic mannitol (0.25mL/kg/s over 30 seconds) delivered through the right internal carotid artery. Objective: Numerous studies trying to find the causes and implications of binge drinking on college campuses have focused their attention on the heavy drinkers. The purpose of this study was to understand why and how college students choose to abstain from drinking. The study also examined the experiences of the abstaining college students on a campus where 83% of the student body drinks. Methods: Twelve undergraduate students from the University of Richmond participated in this qualitative study. Effects of norepinephrine (NE) on macrophage cytokine release are complex because the cells have both α2 and β2 adrenergic receptors, which mediate opposing actions. Furthermore, β2-adrenergic agonists are reported to have both stimulatory and inhibitory effects on interleukin-6 (IL-6). This study was designed to clarify the autocrine role of macrophage-derived NE on IL-6 production in activated peritoneal macrophages. Effects of NE on IL-6 production in the RAW264.7 macrophage cell-line also were investigated. Treatment of activated peritoneal macrophages with endotoxin, the α2-adrenergic antagonists yohimbine or RS79948 revealed that the α2-adrenergic receptor mediates a stimulatory autocrine action of catecholamines on IL-6 production. …

The distribution of obligate and facultative phreatophytic vegetation reflects the gradient of ecohydrological habitats in a landscape. Preliminary investigations of Banksia vulnerability to xylem embolism have reported that obligate phreatophytes are more susceptible to water stress than facultative phreatophytes (Froend & Drake 2006). A quantitative measure of plant susceptibility to water stress is vital when establishing environmental water requirements. The New York City Housing Court faces significant challenges in addressing housing disputes, particularly due to the high number of pro se litigants and the intersection of legal and social issues that contribute to homelessness. A 2004 conference brought together diverse stakeholders to examine these challenges and propose reforms, focusing on six key areas: pre-adjudication steps, the adjudicative process, the right to counsel, litigants of diminished capacity, preserving housing stock, and social services. The conference emphasized the need for systemic changes to improve access to justice and efficiency in the court.

For most older people in the United States, Social Security is the major source of income: nine out of ten people age 65 or older receive benefits, which represent an average of 41 percent of their income. Largely as a result of Social Security, poverty rates for the elderly are at an all-time low, just 10 percent. But pockets of poverty persist: older unmarried persons, blacks, and Hispanics experience poverty rates in excess of 20 percent, and over 40 percent of all older single black women live in poverty. Our research addresses fundamental long-standing concerns in the compensating wage differentials literature and its public policy implications: the econometric properties of estimates of the value of statistical life (VSL) and the wide range of such estimates from about $0.5 million to about $21 million. We address most of the prominent econometric issues by applying panel data, a new and more accurate fatality risk measure, and systematic selection of panel estimator in our research. Controlling for measurement error, endogeneity, individual heterogeneity, and state dependence yields both a reasonable average level and narrow range for the estimated value of a statistical life …

Research has shown that determinants of PDV victimization are different for male and female adolescents. However, inconsistent findings entail that further research needs to be done using a representative sample of male and female adolescents.Objective: To identify gender-specific determinants of PDV victimization utilizing a nationally representative sample of high school adolescents.Methods: Data from the 2005 National Youth Risk Behavior Survey was used for this analysis. Pharmacological Studies Of Nicotine And Galantamine Effects On Age Associated Memory Impairment And The Neurotrophic System In Aged Female Rats, Kristen L. French

Nicotine and Galantamine were studied for their pharmacological effects on age associated memory impairment (AAMI) and the neurotrophin system in the aged female F344 rat. Nicotine was found to have dose dependent effects on cognition in the 24 month old rat. A low dose of nicotine improved both working and reference memory (WM, RM) performance on the water radial arm maze and also improved the ability of aged rats to handle an increasing WM load. A high dose did not improve WM and caused deficits in RM, and high cotinine levels in these subjects correlated with worse performance. Nicotine treatment …
