Birds serve as an excellent group of organisms from which to introduce the study of ecology, being of inherent aesthetic interest to many otherwise uninterested in science, and are also ubiquitous in the immediate environment of many students. By extension, images of birds might serve as a valuable resource for the ecology educator, and bird artists – as a subset of ecologists - might provide useful models for expertise in ecology. Cryopreservation And Intracytoplasmic Sperm Injection With Bovine Epididymal Spermatozoa, Carlos Andres Guerrero

Recently, interest in the preservation of epididymal sperm as a potential source of valuable genes for genome resource banks has escalated. The development of a successful protocol to recover and cryopreserve sperm harvested from the epididymides would salvage germplasm from genetically valuable males that are injured and can no longer mate or have unexpectedly died and can be used as a model for the preservation of male gametes from endangered species. In a series of experiments, epididymal sperm was successfully harvested, cryopreserved and used for intracytoplasmic sperm injection. In Experiment I, ethylene glycol was found to cause significantly (P<0.05) less osmotic damage to bovine sperm during a one step addition and/or removal at 4°C as compared with glycerol in all concentrations evaluated. Furthermore, prolonged exposure (5 days at 4°C) of ethylene glycol was found to be less toxic than glycerol to sperm. In Experiment II, it was demonstrated that glycerol was more effective than ethylene glycol in providing protection against freezing injury during the cryopreservation process in the concentrations evaluated. In Experiment III, it was demonstrated that epididymal sperm retrieval using seminal plasma is beneficial to enhance sperm overall and progressive motility characteristics and to protect it from morphological abnormalities derived from the freezing process. In Experiment IV, a one step dilution process for removal of glycerol from cryopreserved epididymal sperm was found to significantly affect plasma membrane integrity and mitochondrial function of sperm previously exposed to seminal plasma. However, seminal plasma exposure did not have any significant detrimental effect on acrosome integrity. Furthermore, it was demonstrated that the longevity and survivability in vitro during a 4-hour incubation period at 37°C of post-thaw epididymal sperm exposed to seminal plasma prior to cryopreservation was not compromised when compared with the control extended sperm. In Experiment V, we have demonstrated that fertilization, blastocyst and fetal development could be achieved with cryopreserved bovine epididymal sperm by intracytoplasmic sperm injection (ICSI). To our knowledge, this is the first report in the United States and second in the world to use bovine epididymal sperm for ICSI. We achieved far markedly improved blastocyst rates over those results recently reported in the first study originating in Japan.

Salt marshes are dynamic, highly productive habitats and serve as nursery grounds for many commercially and economically important species. Benthic microalgae (BMA) are considered an important food source for benthos and provide the principal source of nutrition that fuels secondary production. Estuarine sediments around the world are a repository for many contaminants from anthropogenic sources. In particular, hydrocarbons and metals are ubiquitous contaminants in coastal systems. West Nile virus (WNV), a member of the genus Flavivirus transmitted by mosquitoes, first appeared in the New York in 1999. Within five years WNV was detected throughout the contiguous 48 states causing disease in reservoirs and accidental hosts alike. In Louisiana, WNV was first detected in 2001 with one human case, ten equine cases, and six dead birds reported. The introduction of WNV into Louisiana presented an unique opportunity to observe an emerging disease unfold, so a study was launched to gain insight into the epidemiology of WNV in Louisiana. The purpose of this study was to examine the effect of tempo on movement responses of children ages one to three. For two weeks, 17 children between the ages of 22 and 36 months were videotaped twice per week to observe and measure their movement responses to fast and slow musical stimuli. During these sessions, the children were videotaped in their classrooms, engaged in either free play or in a quiet group activity. This dissertation explores the spatial ecology and potential pathways of infection of anthrax, Bacillus anthracis, in North America. A multi-scale approach was used to evaluate the components required for disease agent survival in the environment, interactions with wildlife, and the potential role that vectors play in anthrax transmission. First, ecological niche modeling with the Genetic Algorithm for Rule-set Production (GARP) was used to predict the geographic distribution of anthrax in the continental U.S. using case data from outbreaks between 1957 and 2005. Pregnant women ingest contaminated fish, which transfers high mercury concentrations to the unborn fetus. This exposure may result in methyl mercury buildup in the brain of the unborn fetus. Symptoms ranging from minor to severe may be observed as special educational needs in school systems. These include learning disorders, developmental delays, and autism. Terracing is a novel technique used to combat coastal marsh loss in Louisiana and Texas. Terraces are assumed to slow marsh erosion, decrease pond depth, and encourage vegetation expansion. Terraced ponds have never been evaluated as habitat for waterbirds, which heavily depend on Louisiana�s coastal marshes. From April 2005 to April 2006, I monitored waterbird species richness and density through time to estimate effects that terracing has on habitat quality. Water quality (turbidity, salinity, conductivity, water temperature, and water depth) also was measured. The objective of this research was to evaluate color and tenderness in chilled or frozen pork loin chops after antioxidant dipping and modified atmosphere packaging. Loin chops were dipped in 0.3 M calcium chloride, 2.0 % sodium ascorbate, 0.2 M calcium ascorbate, or 0.3 M calcium ascorbate. Non-dipped chops served as controls. Chops were packaged in high oxygen (80% O2 / 20% CO2) or no oxygen (80% N2 / 20% CO2) and stored chilled (4° C) for 7 days or frozen (-18° C) for 21 days. After storage, chops were displayed under continuous fluorescent lighting for 3 or 6 days. …

This project examines themes of race, gender, and nation in a series of four novels by nineteenth-century Louisiana author Sidonie de la Houssaye. The series, called Les Quarteronnes de la Nouvelle-Orléans (The Quadroons of New Orleans), is based on the system of plaçage. Plaçage, a system of concubinage in which white men took women of mixed racial heritage (such as “quadroons”) as mistresses, becomes a source of conflict and contradiction in the series. The author sees plaçage as a tragic necessity for some educated and morally “upright” quarteronnes. For others, those quarteronnes depicted as libidinous and avaricious, it is a …

In this dissertation I describe a number of patterns and interesting aspects associated with the evolution of snake mitochondrial genomes (mtDNA). I also attempt to resolve the phylogeny of squamates, focusing on the relationship between the snakes and lizards. The results of this study indicate that snakes and worm lizards (amphisbaenians) appear to share an exclusive common ancestor, and snakes appear to have undergone strong selective pressure that shaped snake mtDNAs. Snake mtDNAs have several unique features, including a compact size, duplicated control regions, and an elevated evolutionary rate. Amacrine cells mediate complex lateral signaling in the inner plexiform layer of the vertebrate retina. Glutamate is the primary excitatory neurotransmitter that shapes the retinal circuitry by activating ionotropic glutamate receptors (iGluRs)and metabotropic glutamate receptors (mGluRs). One step in understanding the signaling in the inner retina is to understand the role and expression patterns of these glutamate receptors. Self-assembly is scientifically interesting, technologically important, and sometimes life-threatening. A striking example: Alzheimer’s disease is caused by the self-assembly of the â-amyloid fragment of amyloid precursor protein. Such natural amphiphiles normally are very expensive and complicated to isolate. Synthetic mimics that are much cheaper and easier to prepare can facilitate the development of efficient characterization methods, even as they suggest new materials science applications. Two-directional arborols are synthetic bolaform amphiphilic dendrimers. They dissolve in hot water and form reversible gels at room temperature. Louisiana is within one of the major cow-calf production areas in the US (the Southeast). Louisiana accounts for about 1.72% of the total US cattle operations. This research explored whether there was a difference between two methods of phonemic awareness instruction. Two groups of kindergarten students were selected. One group received an implicit method of phonemic awareness instruction. A second group received an explicit model of phonemic awareness instruction. Of particular concern were these questions: What is the impact of phonemic awareness instruction on reading achievement? How does phonemic awareness instruction delivered via an implicit instructional method differ from an explicit, scripted method of instruction in their respective impact on reading achievement? The Southern region of the United States historically has a high rate of violent crime, especially homicide. This has led to a number of studies tackling the issue by relying on subcultural theory or by using structural correlates of crime to account for the South versus non-South difference in homicide. Macro level research has focused on pitting culture (usually measured by a dummy variable for South) against structural characteristics such as poverty and measures of income inequality, but suffers from a lack of direct cultural measures needed to successfully evaluate the subcultural thesis. Nineteenth-century Americans took great pride in the completion of the first transcontinental railroad in 1869. This pride was not solely grounded in the knowledge that a grand, technological feat had been accomplished. When placed in its historical context, the celebration surrounding the completion of the railroad suggests a clear and visible statement of unity following a bitter and divisive civil war. The transcontinental railroad of 1869 undeniably unified the States. Elizabeth I of England was one of the most celebrated monarchs in history. Authors, playwrights, and artists venerated her in their art. At her accession, however, her subjects were unsure about their new queen. She was an illegitimate female ruling a religiously divided kingdom. In response to this, Elizabeth and her council initiated a propaganda campaign that created an image of Elizabeth as a wise, just, and well-beloved ruler. Stilbene concentration was analyzed in juice and tissue of muscadine grape (Vitis rotundifolia Michx.) and bunch grape (Vitis labrusca L.) from fresh fruit, after processing and after postharvest treatments. Five filter types were evaluated for percent resveratrol recovery when filtering standard and spiked juice samples in preparation for HPLC analysis. Only two (polycarbonate and Anopore) of the five filter types had more than 90 percent recovery. Polycarbonate was chosen for sample preparation since it was more durable during handling. I tested the scope of the Spring Condition Hypothesis in explaining the continental population decline of lesser scaup (Aythya affinis; hereafter scaup) and explored potential mechanisms affecting female body condition to assist conservation efforts for population recovery. Epoxy fatty acids (EXA) are valuable to industry as they are used in synthesizing plasticizers such as of poly vinyl chloride, resins, adhesives, coating materials such as paint, lubricant, lubricant additives, insecticides, insect repellants, crop oil concentrates and formulations of carriers for slow release pesticides and herbicides. There is interest in developing commercial oilseeds accumulating epoxy fatty acids to at least 50% of the seed oil. Previous studies on market power hypothesis strictly assumed that the data used in the analysis were a stationary process. This assumption has been argued that not all time series exhibit a stationary process such that conventional asymptotic theory cannot be applied. This study adopts the “New Empirical Industrial Organization” (NEIO) approach developed by Bresnahan (1982) and Lau (1982) to measure the degree of market power in the export market for soybean complex. In the last twenty years, interferon-α (IFN-α) has gained success as an immunotherapy for treating such cancers as hairy cell leukemia, malignant melanoma, and renal cell cancer. Our goal was to improve the effectiveness of IFN-α therapy by genetically modified the IFN-α gene to encode a tumor-targeting peptide fused to a functional IFN-α protein. To ensure the targeting peptide worked, a genetically modified reporter gene encoding a secreted alkaline phosphatase (SEAP) gene and different mini-peptides were used to determine distribution and targeting ability. A major task in proteomics is to identify proteins from a biological sample using two-dimensional (2-D) separation prior to mass spectrometry of peptides generated via proteolytic digestion of the proteins. For 2-D separations, microfluidic devices are superior to bench top and capillary-based systems since they potentially provide higher separation efficiencies due to the minimal dead volumes produced during peak transfer between the two separation dimensions. In addition, fast separations can be envisioned because the column lengths are typically shorter in microfluidic platforms without scarifying peak capacity. Diets are typically poorer and risk of chronic disease is greatest in low-income populations. A relationship has been established in the literature between food costs and diet quality, where lower cost diets are generally those of the poorest quality. Food group intake, energy/nutrient intake, and diet cost were assessed in 64 female food stamp recipients in Southeast Louisiana. From one 24-hour dietary recall collected at the beginning of the monthly resource cycle (Day 1) and one at the end (Day 2), nutrient intakes and diet costs were able to be analyzed between different time frames.
