Escherichia coli O157:H7 is an emerging food pathogen that was first identified as a cause of illness in 1982. According to CDC (The Center for Disease Control and Prevention) estimates, E. coli O157:H7 is responsible for about 73,000 illnesses, 2,000 hospitalizations and 60 deaths in the United States each year. Cattle are the principal reservoir of E. coli O157:H7. Contamination of feed and drinking water with cattle feces is an importance factor in the prevalence of infection in cattle which, in turn, results in contamination of food and the environment. At elevated levels, homocysteine (Hcy) is a risk factor for cardiovascular diseases and Alzheimer’s disease. It’s also implicated in a number of other diseases such as neural tube defects, pregnancy complications, and renal failures. The monitoring of Hcy levels in plasma is of current concern. To date, the biochemical mechanism by which Hcy is involved in disease states is unclear. Herein, we describe highly selective colorimetric methods for the direct determination of Hcy. Inexpensive, commercially available materials are reported. The results show potential application for the detection of Hcy in human blood plasma. Measuring Player Perceptions Of Advertising In Online Games, Ben Lewis

An experiment with 100 participants aged 18-24 was conducted to measure the effects of advertising in an online role-playing computer game on perceived interactivity and other aspects of gameplay experience. Results from a post-test questionnaire revealed insight into players' attitudes toward advertising in video game environments, and reflected varying levels of advertising awareness and recall, message recognition, and factors in purchasing habits. Results suggested that while advertising in online games can sometimes trigger high advertising awareness rates, it can also reduce a game's perceived sense of realism and genuinely annoy players if not appropriately coordinated with the game environment. Survey data collected from randomly selected participants within the four geographical regions of the U.S. were used to evaluate consumer attitudes towards functional foods and determine their willingness to pay for these foods. Contingent valuation using the payment card method was used to elicit premiums that consumers are willing to pay for a spread that maintains a healthy heart (spread A), a spread that is proven to significantly reduce cholesterol (spread B) and a loaf of bread that may reduce the risk of heart disease and certain cancers (bread A). Domangue, Mathews, Sun, Roussel, and Guidry (2004) trained participants to generate valid exemplars from an artificial grammar using either memory-based or model-based processing. Their results showed that learning by memory-based processing resulted in fast but inaccurate performance, while model-based learning resulted in slow but accurate performance. Attempts to integrate both types of training did not result in fast and accurate string generation. Fast and accurate performance was achieved by Sun and Mathews (2004) using a computer animated display to train participants. My study focuses on the literature of democratic morality, with specific reference to the question of "heroic individualism." I attempt to elucidate the notion of heroic individualism by examining three modern democratic moralists whose work occupies the space between politics and literature: Jean Jacques Rousseau, Thomas Carlyle and Friedrich Nietzsche. In brief, I conclude that the central aspiration of heroic individualism is to bridge the gap between writing and action, the Text and the Voice. The dialogue among Rousseau, Carlyle, and Nietzsche reveals that the problem of writing as action is central to heroic-individualist morality. Each of these authors demonstrates …

Vibrio vulnificus is a Gram-negative organism that is found in estuarine environments. The bacterium is capable of causing gastroenteritis, wound infections, and septicemia, often resulting in death for susceptible individuals. Monoclonal antibodies (MAbs) to five lipopolysaccharide (LPS) O-serovars have previously been used as a serotyping system in epidemiological studies. Here is the first report of two additional MAbs capable of recognizing two new LPS serovars (O6 and O7), further increasing the number of V. vulnificus that can be typed. Effects of hydrocarbons, salinity, tidal height, bioenergetics and competition on the oyster reef fauna were studied. Dried oyster shell was exposed to crude oil in the laboratory and plastic trays filled with control and oil-exposed shell were then deployed at two locations, in two seasons and at two tidal levels, with immersion periods of two and five weeks. Salinity exhibited a significant effect with sub tidal site having higher diversity. Hydrocarbon effects were less prominent. To determine effects of cleaners, Corexit 9580 was applied both alone and in combination with oil on cultch. At high concentrations, the cleaner ameliorated hydrocarbon …

The first purpose of the research project was to examine the relationship between child weight status and adult quality of life. The second purpose of the research project was to test psychosocial variables as mediators of the relationship between child weight status and adult quality of life. A total of 164 undergraduate and graduate students from Louisiana State University participated in the study. The students completed five online questionnaires that were used to assess variables such as child weight status, adult weight status, history of childhood teasing experiences, child self-concept, and adult quality of life. Analysis of the mechanical and neural regulatory mechanisms of the flexion-relaxation phenomenon (FRP), observed in deep trunk flexion, was performed since it is believed these mechanisms provide insight into the causes of low back injury and pain. Three methods were used to analyze the behavior of the lumbar tissues during trunk flexion-extension exercises: 1) active continuous cyclic movement, 2) acute cyclic movement at different orientations, and 3) passive continuous cyclic movement. All activities were performed at a rate of 0.1 Hz (6 cycles•min-1) while monitoring the surface electromyography (EMG) of the lumbar paraspinal muscles. Brine incorporation into meat products has become a well established practice in the United States. Brines are used to enhance the eating quality of pork by improving its tenderness and juiciness. Carbon dioxide and carbon monoxide have been used in modified atmosphere packaging as antimicrobial agents and color stabilizers, respectively, to increase the shelf life of retail meats. Southeastern coastal plain pine savannas lack direct evidence of past fire regimes. As a result, uncertainty exists regarding the range of variation in frequency and seasonal timing of past fire regimes and the relative importance of anthropogenic and lightning-ignited fires. Characterization of past fire regimes is needed for effective restoration and management of these high-biodiversity ecosystems. I used dendrochronologically dated fire scars from stumps of old growth longleaf pines in a large coastal, mainland pine savanna and from dead slash pines on a small, coastal barrier island in north Florida to explore past fire regimes. Evaluation Of A Plasmid Delivery System For Production Of Gnrh And Ghrh In The Horse And Goat, William Andrew Storer

The efficacy of a novel plasmid delivery system was assessed for long-term expression of gonadotropin releasing hormone (GnRH) and growth hormone releasing hormone (GHRH) in horses and goats. The efficacy of the technology was demonstrated using 3 novel plasmids: pSEAP [expressing secreted embryonic alkaline phosphatase (SEAP)], pGHRH (expressing GHRH), and pGnRH (expressing GnRH). Geldings were electroporated with a reporter plasmid expressing SEAP in 3 muscle sites. Expression of SEAP, measured from jugular plasma samples, indicated muscle specificity for uptake and expression of the plasmid. Concentrations of SEAP were greatest (P < 0.05) after pectoralis injection, which was chosen as the site for electroporation in subsequent studies. In a second experiment, stallions were electroporated with pGHRH or pSEAP to evaluate the effect of long-term GHRH treatment on the growth hormone (GH) axis and testicular function. Stallions treated with pGHRH had increased (P < 0.05) plasma concentrations of IGF-I, increased (P < 0.05) volume of accessory sex gland fluid, and increased (P < 0.05) number of normal spermatozoa in the ejaculate relative to controls. In the third experiment, stallions were electroporated with pGnRH or pSEAP to test the effects of GnRH on the reproductive axis. Treatment with pGnRH increased (P < 0.05) plasma testosterone concentrations to d 56 and increased (P < 0.01) the LH response to GnRH on d 21, but did not alter (P > 0.1) seminal characteristics evaluated after 36 d of treatment. Access to a running wheel causes a feeding suppression in adult rats (Afonso & Eikelboom, 2003) but it appears this wheel induced feeding suppression is not evident in weanlings (Cortright, Chandler, Lemon, and DiCarlo, 1997). This ad lib wheel and food model (ALWF) has been suggested as an animal model of anorexia nervosa (AN). Two characteristic features of AN are the disorder's very specific age of onset and the marked sex difference in incidence, with the majority of cases first becoming evident in females during puberty. The contextual interference hypothesis holds that simple motor skill tasks are best learned when practiced under blocked, or repetitive conditions, but that retention and transfer are best accomplished when the skill has been practiced in varied conditions. The purpose of this study was to measure the effects of contextual interference practice conditions on the acquisition, retention, and transfer of a complex task—right hand lead percussion sticking technique among university musicians. All participants (N = 120) demonstrated rhythmic competency for the task, and were necessarily unable to perform the sticking technique with accuracy at the start of treatment. The purpose of this study was to examine the usefulness of the MacArthur- Bates Communicative Development Inventory (CDI) as an assessment tool for low-income, African American (AA) children. The data were from eighty-seven typically developing AA children, aged 8 to 30 months; these children were recruited from childcare centers that served low-income populations in Baton Rouge, Louisiana. Each participant’s primary caregiver completed a biographical sketch and a CDI inventory. Two analyses were completed. Of the 36 species of felines in the world, all except the domestic cat are listed as endangered or threatened. To preserve the genetic diversity of felines and other species, genome resource banks have been established. Due to limited availability of germ cells for research, studies must use models to optimize the techniques before they are applied to endangered species. In this study, preservation of oocytes and spermatozoa was examined using the bovine as a model for felines. This dissertation explores effects of windstorm and fire disturbances on the clonal and reproductive biology of cane (Arundinaria gigantea Muhl., Poaceae). In this collection of studies, multiple disturbances interact in complex ways, and some interactions appear only after a substantial lag. One implication of this research is that multiple, interacting disturbances might strongly influence the boundary between monodominant and species-rich plant communities. Cane is the only bamboo native to the United States, and once covered vast areas of bottomlands in the southeastern U.S. in monodominant stands called “canebrakes.” The study took place on the Buckhorn Wildlife Management Area in Tensas …

Some personal or collective histories can never be completely integrated into the continuum of one's emotional life. Such stories produced in traumatic times or in disastrous events are likely to remain only partially understood or accepted. Examining the human consequence of traumatic events such as the enslavement of Africans in the United States or the attempted extermination of the Jewish people in Europe is one challenging focus of this work. It is comparatively productive, however, if these events are approached from the perspective of the trauma they have produced-an approach that suspends chronological and geographical barriers of time and space. …
