Novel Cyanine-Amp Conjugates For Efficient 5' Rna Fluorescent Labeling By One-Step Transcription And Replacement Of [γ-32P] Atp In Rna Structural Investigation, Na Li, Changjun Yu, Faqing Huang

Two novel fluorescent cyanine-AMP conjugates, F550/570 and F650/670, have been synthesized to serve as transcription initiators under the T7 φ2.5 promoter. Efficient fluorophore labeling of 5′ RNA is achieved in a single transcription step by including F550/570 and F650/670 in the transcription solution. The current work makes fluorescently labeled RNA readily available for broad applications in biochemistry, molecular biology, structural biology and biomedicine. In particular, site-specifically fluorophore-labeled large RNAs prepared by the current method may be used to investigate RNA structure, folding and mechanism by various fluorescence techniques. Are Leptodactylus Didymus And L. Mystaceus Phylogenetically Sibling Species (Amphibia, Anura, Leptodactylidae)?, Rafael O. De Sá, W. R. Heyer, Arley Camargo

The Leptodactylus fuscus species group consists of 25 currently recognized species; within this species group and distributed throughout the Amazon Basin, Atlantic Forests, Gran Chaco, and cerrados is the L. mystaceus species complex. This species complex consists of L. didymus, L. elenae, L. mystaceus, L. notoaktites, and L. spixi. Adult morphologies have been used to distinguish these species from each other except for L. didymus and L. mystaceus (Heyer, 1978; Heyer et al., 1996). Leptodactylus didymus and L. mystaceus are morphologically indistinguishable; the species are recognizable only by the characteristics of their advertisement calls: non-pulsed in L. didymus and pulsed …

Biodegradable polymer materials, specifically poly(lactic-co-glycolide) (PLGA) can be used as bone replacements for bone regeneration. Scaffolds can be prepared to be porous to induce bone growth into a scaffold so that it is replaced with natural tissue as the polymer degrades. However, simply using PLGA will result in formation of scar tissue rather than regeneration of natural bone. Therefore focus has turned to attaching growth factors to the PLGA molecules to elicit a specific cellular response when the implant is placed in the body. A restored elk (Cervus elaphus) population in eastern Kentucky may be affecting ecosystem processes in a landscape composed of reclaimed grassland expanses and isolated forest remnants. Elk routinely select forested ridge-tops as resting and ruminating sites. These locations are characterized by sparse or absent leaf litter, trampled and diminished vegetation, large deposits of dung, and urine-saturated soils. In fall 2003, a series of fenced ungulate exclosures were constructed; 8 on highly disturbed forested ridge-tops and 8 on reclaimed grasslands. Interleukin-10 (IL-10) is a pleiotropic cytokine which limits inflammatory responses by balancing the hosts immune response against infection. Mammalian Toll-like receptors (TLRs) are pattern recognition receptors that recognize specific molecular pattens on microbial pathogens and activate intracellular signaling via the transcription factors NF-B and IRF-3. In this study we evaluate the contribution of the TLR ligands Poly I:C, Pam3CSK4, LPS and LTA to IL-10 receptor dysfunction in murine peritoneal macrophages (PM). Degenerative suspensory ligament desmitis (DSLD) is a debilitating disease of connective tissues seen in many breeds but has become prevalent in the Peruvian Pasohorse. DSLD is believed to be a genetic disorder caused by one primary founder and most likely has a recessive mode of inheritance although a dominant or co-dominant mode of inheritance has not been ruled out.

Population Dynamics Of Oryzomys Palustris And Microtus Pennsylvanicus In Virginia Tidal Marshes, Christopher P. Bloch, Robert K. Rose

Oryzomys palustris (marsh rice rat) and Microtus pennsylvanicus (meadow vole) cohabit coastal marshes in the mid-Atlantic US. Both were live-trapped for 23 months at two tidal marsh sites in Virginia to assess their demography near the margins of their distributions. In the presence of dense vegetation, population dynamics of the two species were seasonal and positively correlated, with densities declining through the winter. At the more sparsely vegetated site, densities of both species were lower, and densities of M. pennsylvanicus were negatively correlated with those of O. palustris. Patterns of reproduction differed between the species. O. palustris was reproductively …

We are living in a globalizing world, characterized by constant and rapid innovation. As we are inclined to go with this flow and its accompanying discourse of mobility, there is a danger that we overlook the persistence of cultural traditions. However, this paper argues that important differences exist between pre-modern, modern and post-modern traditions with regard to the form mobility takes. After a short discussion of the role information and communication technologies play in post-modern traditions, it is argued that these technologies transform our world, and not only our electronic files, into a global database and, as a consequence, generate …

The Non-Modularity Of Moral Knowledge: Implications For The Universality Of Human Rights, Theresa W. Tobin

Many contemporary human rights theorists argue that we can establish the normative universality of human rights despite extensive cultural and moral diversity by appealing to the notion of overlapping consensus. In this paper I argue that proposals to ground the universality of human rights in overlapping consensus on the list of rights are unsuccessful. I consider an example from Islamic comprehensive doctrine in order to demonstrate that apparent consensus on the list of rights may not in fact constitute meaningful agreement and may not be sufficient to ground the universality of human rights. The structural and electronic properties of Gd₂(Ti_1−yZr_y)₂O₇ (y=0–1) pyrochlores following a 2.0-MeV Au²⁺ ion-beam irradiation (~5.0 X 10¹⁴ Au²⁺/cm²) have been investigated by Ti 2p and O 1s near-edge x-ray absorption fine structure (NEXAFS). The irradiation of Gd₂(Ti_1−yZr_y)₂O₇ leads to the phase transformation from the ordered pyrochlore structure (Fd3m) to the defect fluorite structure (Fm3m) regardless of Zr concentration. The fragmentation dynamics of core-excited H₂S has been studied by means of partial anion and cation yield measurements around the S L_2,3-subshell ionization thresholds. All detectable ionic fragments are reported, and significant differences between partial ion yields are observed. Possible dissociation pathways are discussed by comparison to previous studies of electron spectra.

In this article Professor Guruswamy advances an argument for new energy agreements that address the immense global environmental challenge presented by the increasing global energy demands of both the developed and developing world. Arguing that new energy accords are needed to meet this challenge, he identifies and describes the decidedly interdisciplinary knowledge base and analytics required to negotiate such international instruments. The construction of these knowledge bases call for scientific, engineering, technological, legal, social, economic and behavioral expertise.
