Design And Fabrication Of A Re-Configurable Micromirror Array For An Optical Microspectrometer, Vandana Upadhyay

This thesis presents the design and fabrication of a re-configurable micromirror array which can be used as a component of an optical microspectrometer. In an optical microspectrometer, an array of mechanically positionable micromirrors can be implemented as a reconfigurable exit slit to selectively focus particular wavelengths of a diffracted spectrum onto the detector stage. The signal to noise ratio and response time of an optical microspectrometer can be vastly improved by this technique.

In the approach presented here, a hybrid bulk- and surface- micromachining process is demonstrated for fabrication of a 1XN array of micromirrors. The reconfigurable micromirrors presented here …

Method Of Producing A Branched Carbon Nanotube For Use With An Atomic Force Microscope, Rudiger Schlaf, Zhifeng F. Ren, Jianguo Wen, David L. Carnahan

A method of producing a branched carbon nanotube (CNT) is disclosed. The branched CNT is used with an atomic force microscope having a cantilever and a tip and that is able to measure a surface of a substrate as well as an undercut feature of the substrate that protrudes from the surface. A catalytic material is deposited onto the tip of the microscope, and the catalytic material is subjected to chemical vapor deposition. This initiates growth of a primary branch of the branched carbon nanotube such that the primary branch extends from the tip. A secondary branch is then introduced …

Alterations In Uterine And Placental Sodium Pump Abundance May Contribute To The Onset Of Mouse Labor, Carlos Jacob Vance

Objective: Reductions in sodium pump (SP) abundance can give rise to increases in contractile force in uterine and vascular smooth muscle as well as an increased secretion in secretory cells, including potentially those of the placenta. To determine whether the mouse might serve as a model for human pregnancy in terms of the SP and to determine whether changes in SP abundance anticipate or follow labor, we studied pregnant mice over the final trimester of their pregnancy.

Study Design: C57Bl6 dams (n=46) were bred and studied during their pregnancy. Animals (n=4) were sacrificed at specific gestational time points. Other mice …

The Microbial Selenoproteome Of The Sargasso Sea, Yan Zhang, Dmitri E. Fomenko, Vadim Gladyshev

Background: Selenocysteine (Sec) is a rare amino acid which occurs in proteins in major domains of life. It is encoded by TGA, which also serves as the signal for termination of translation, precluding identification of selenoprotein genes by available annotation tools. Information on full sets of selenoproteins (selenoproteomes) is essential for understanding the biology of selenium. Herein, we characterized the selenoproteome of the largest microbial sequence dataset, the Sargasso Sea environmental genome project.
Results: We identified 310 selenoprotein genes that clustered into 25 families, including 101 new selenoprotein genes that belonged to 15 families. Most of these proteins were predicted …

The existing scientific literature on offshore benthic assemblages (OBA) residing along the US East and Gulf of Mexico continental shelf was reviewed. Identification was made of any associations between the dominant OBA and particular sediment types and/or bathymetry. Of special interest was the evaluation of reported effects of sand dredge/mining activities on the dominant OBA and recognition of data deficiencies. One hundred and twenty-two references were selected and classified as to type of study with pertinent results extracted. Polychaetes were predominantly cited as the principal infaunal taxa present in studies from both the Gulf of Mexico and US Atlantic coast. …

Part 1:

Prophage induction has been demonstrated to be a sensitive indicator for a wide variety of toxic and mutagenic compounds and, as a consequence, has been utilized for biologically based carcinogen screenings. Fourteen marine bacterial isolates were screened for development into the Marine Prophage Induction Assay (MPIA), for marine samples. The selected isolate (P99-4S3) was identified by 16S rDNA sequencing as Pseudomonas aeruginosa. This isolate demonstrated a log-linear response to increasing dose of mutagens, and sensitivity to known environmental contaminants. Field-testing of the assay over two years demonstrated the MPIA would be a useful screening tool for environmental …

Arf and Rab family GTPases regulate membrane traffic in cells, yet little is known about how they are targeted to distinct organelles. To identify sequences in Arf-1 necessary for Golgi targeting, we examined the localization of chimeras between Arf-1 and Arf-6. Here, we identify a 16-amino acid sequence in Arf-1 that specifies Golgi targeting and contains a motif (MXXE) that is important for Arf-1 binding to membrin, an ER-Golgi SNARE protein. The MXXE motif is conserved in all Arfs known to localize to the Golgi and enables Arf-1 to localize to the early Golgi. Arf-1 lacking these 16 aa can …

Ion Transmission In The First Vacuum Stage Of An Inductively Coupled Plasma Mass Spectrometer, Jeffrey Harris Macedone

The inductively coupled plasma mass spectrometer (ICP-MS) is the instrument of choice for trace and ultra-trace elemental analysis. However, the ICP-MS suffers from matrix effects. Matrix effects occur when instrument response varies as the composition of the sample matrix is changed. Matrix effects, or non-spectroscopic interferences, limit the accuracy of routine analysis. Identification of the sources of matrix effects provide a basis for reducing or eliminating them. As inaccuracies in the ICP-MS are more severe than those in the ICP atomic emission spectrometer, the problem may be due, at least in part, to the vacuum interface used to couple the …

Forensic anthropology is the identification of skeletal remams in medicolegal death investigations. The foundation of forensic anthropology rests on "documented" skeletal collections, which provide a crucial repository of information from which experts can rely upon to make determinations about human skeletal remains. A "documented" skeletal collection is one in which the demographics (e.g., ancestry, sex, age, height) of each individual are supported by known facts about the decedent at the time of his/her death. The Effects Of Interracial Interaction On Behavior As A Function Of Prejudice And Race, Jason R. Read

In a series of two experiments, the first involving 121 participants and the second 114, I investigated whether level of racial prejudice is related to performance on a cognitive task and helping behavior in participants who had just interacted with the target of their prejudice. The moderating effect of control was tested and, unlike previous research, the responses of African-American participants were studied too. It was proposed that when people interact with the target of their prejudice, they will experience stress and the aftereffects of stress will lead to a decrement in Stroop task performance and a lower likelihood of …

A Phylogenetic Approach For The Study Of Variation And Determination Of Population Affiliation Of Indigent Human Skeletal Remains, Hattie Bea Wetherington

Mitochondrial DNA (mtDNA) has played a major role in human population studies over the past decade due to its maternal inheritance and negligible recombination (Macaulay, 1999). The mtDNA control region has been the focus of these studies due to the highly polymorphic nature of this non-coding region. Forensic scientists also use mtDNA to help determine the identity of missing individuals when nuclear DNA is not present. However, when skeletal remains are unclaimed, identification becomes near impossible. Therefore, mtDNA can play a valuable role in identification in terms of population affiliation, especially in conjunction with morphological analysis.
