Selenocysteine (Sec) is co-translationally inserted into selenoproteins in response to codon UGA with the help of the selenocysteine insertion sequence (SECIS) element.The number of selenoproteinsinanimals varies, with humans having 25 and mice having 24 selenoproteins. To date, however, only one selenoprotein, thioredoxin reductase, has been detected in Caenorhabditis elegans, and this enzyme contains only one Sec. Here, we characterize the selenoproteomes of C.elegans and Caenorhabditis briggsae with three independent algorithms,one searching for pairs of homologous nematode SECIS elements, another searching for Cys- or Sec-containing homologs of potential nematode selenoprotein genes and the third identifying Sec-containing homologs of annotated nematode …

A number of studies have demonstrated an increased rate of histories of childhood maltreatment among adults with serious mental illness. The present investigation documented the presence of childhood maltreatment in a sample of 40 psychiatric inpatients with schizophrenia spectrum disorders. The type (neglect, physical abuse, sexual abuse), duration, and severity of childhood maltreatment was examined along with measures of premorbid functioning, current symptomatology, and cognitive functioning. Participants with histories of maltreatment were significantly more likely to have poorer peer relationships in childhood, more diffi culty in school, an earlier age at first hospitalization, more previous hospitalizations, elevated symptoms of anxiety, …

Olfactory communication and associated scent-marking activities play a major role in the behavioral ecology of many mammals. During the mating season (rut), scent marking associated with urine of male cervids is an important chemical cue to relay information to conspecifics. Specifically, adult male moose (Alces alces) dig rutting pits in which they urinate, and females respond strongly to urine deposited in pits. A behavioral bioassay was developed to aid in the identification and function of adult male moose urinary pheromones, which elicited the behaviors observed in females during rut. The serotonin type-3 receptor (5-HT3R) is widely distributed in peripheral and central nervous systems. This pentameric protein is a member of the Cys-loop superfamily of ligand gated ion channels and plays a role in mediating physiological processes in nervous, cardiovascular, and digestive systems. The ligand-binding domain of this receptor is extracellularly located and is composed of multiple putative loop structures. Based on structural and sequence homology with other members of the superfamily, it has been proposed that at least six such loops (loops A to F) contribute to the ligand-binding domain. Hibernation is a unique physiological state characterized by suppressed metabolism and body temperature that is interrupted by multiple, brief periods of arousal throughout the hibernation season. Blood flow fluctuates during hibernation and arousal in a reperfusion-like manner without causing neurological damage. Previous studies show that hippocampal slices from hibernating animals tolerate experimental oxygen nutrient deprivation and N-methyl-D-aspartate (NMDA) toxicity better than slices from euthermic animals. However, the cellular mechanisms underlying these examples of tolerance remain unclear. Tolerance to NMDA toxicity suggests that modulation of NMDA receptors (NMDAR) contributes to intrinsic tissue tolerance in slices from hibernating Arctic ground squirrels (hAGS, …

Heterotrophic bacteria, adapted to small concentrations of substrate, are a main component of the microbial flywheel. However, understandings of their activity, isolation, genetics, and nutrition are restricted to the large, easily isolated and culturable bacteria. By using the dilution culture method, apparent culturabilities could approach 10% in unamended lake water and were inversely proportional to the number of cells inoculated from mixed species in a natural environment from Harding Lake. Substrate additions could not improve bacterial culturability in the dilution cultures. Comparative sequence analyses of 16S rDNA genes showed that all bacterial species have similar lengths in the phylogenetic tree, …

Periglacial landforms, called patterned ground, change the vegetation, microtopography and organic content of the surface soil horizons. Because they are uniquely products of the periglacial environment, changes in that environment affect their distribution and activity. As surface features, they mitigate heat and mass transfer processes between the land and atmosphere. For environmental change detection, the state of the soil and active layer must be monitored across temporal and spatial scales that include these features. The goal of this research was to investigate methods and techniques that enhance mass transfer through the membranes. Two general types of fluids were investigated: synthetic wastewater treated in a membrane bioreactor (MBR) and natural and simulated river water. For both fluids, a wide range of solid concentrations (up to 18 g/L) were tested. The membranes investigated were all tubular modules at pilot scale between 0.75 and 1.20 m length, with tubular diameters of 5.5--6.3 mm, 0.2 mum pore size, and membrane surface areas of 0.036--0.1 m2. In 1960, after nearly a decade of controversy and failed legislative attempts, the Arctic National Wildlife Range was established by an executive order "for the purpose of preserving unique wildlife, wilderness, and recreational values." This is the story of the transformation of this little-known expanse of mountains, forest, and tundra into a place internationally recognized as one of the finest examples of wilderness. Pristane (2,6,10,14-tetramethylpentadecane) concentrations in mussels (Mytilus trossulus) increase abruptly during spring in Prince William Sound (PWS), Alaska. This increase is mainly due to ingestion by mussels of pristane-laden feces produced by nearshore zooplanktivores, especially juvenile pink salmon (Oncorhynchus gorbuscha), and I evaluate whether the increase may be used as an index of foraging success, and hence early marine survival, of pink salmon. Pristane is biosynthesized by Neocalanus copepods. Examination of the trophic and temporal distribution of pristane found in 3,007 samples implicates Neocalanus copepods as the source of pristane in PWS. Neocalanus copepods often dominate the zooplankton biomass during spring …

The Influence Of Water Velocity And Depth On Prey Detection And Capture By Juvenile Coho Salmon And Steelhead: Implications For Habitat Selection And Segregation, John J. Piccolo

I studied the effects of water velocity and depth on drift-foraging by juvenile coho salmon and steelhead to assess how these influence their reported habitat segregation into pools and riffles, respectively. I used three-dimensional video analysis of stream-tank foraging experiments to test how velocity and depth influence prey capture probabilities, and the geometry and dynamics of prey detection and capture. I used the experimental results to develop net energy intake models to predict optimal foraging velocities for coho and steelhead. Prey capture probabilities for both coho and steelhead declined from $65% to 10% with an increase in velocity from 0.29 …

The decline of the Steller Sea lion in the eastern Bering Sea over the last 25 years has resulted in increased management of the pollock fishery due to requirements of the Endangered Species Act, as food competition was hypothesized to contribute to the decline. Our research focused on determining if the pollock fishery was causing significant depletion in the eastern Bering Sea, particularly in Steller sea lion critical habitat. The northern Gulf of Alaska (GOA) shelf is a productive coastal region that supports several commercially important fisheries. The mechanisms supporting such high levels of productivity over this shelf are not understood, however, since it is a downwelling-dominated shelf. In an effort to understand the mechanisms underlying such high biological productivity, nutrient distributions were determined 25 times throughout 1998, 1999, 2000, and 2001 from over the northern GOA shelf and in Prince William Sound (PWS). Deep water (>75 m) nitrate, silicate and phosphate concentrations were positively correlated with salinity indicating an offshore nutrient source. The average annual cycle was …

Currently, the environments in the Arctic are rapidly changing. These changes of climate and ice conditions may alter the quantity, quality, and timing of production of ice algae and phytoplankton in the Arctic Ocean.
